    Abstract It should be evident from this brief account that electron microscopy of thin sections is an invaluable asset in the study of fractions isolated by differential centrifugation. I have tried to indicate how the integrity of particles, purity of fractions, and the existence of new particles can be established through its use. I have also suggested the desirability of a common terminology for all cytology -classic, electron-microscopic, and biochemical. Some have expressed the opinion that neutral terms such as alpha, beta, and gamma membranes (59) are more useful than ergastoplasm, Golgi apparatus, and so forth. As helpful as such neutral terms may be in describing intracellular structures, they do not appear to me to substitute for historically rooted cytological names. Note added in proof: Since this article went to press there has appeared an important article by G. E. Palade and P. Siekevitz (60). These authors consider that the vesicles without granules found in the microsome fraction were "probably derived from the smooth surfaced parts of the endoplasmic reticulum." The latter were found to be continuous with the granule-studded membranes; "the two varieties of profiles represent local differentiation within a common system." The authors confirm the finding of Rouiller (18) and Novikoff et al. (7) of the dense bodies adjacent to the bile canaliculi and describe their presence in the microsome fraction as a "minor component."

    Abstract Most overweight individuals do not develop diabetes due to compensatory islet responses to restore glucose homeostasis. Therefore, regulatory pathways that promote β cell compensation are potential targets for treatment of diabetes. The transient receptor potential cation channel subfamily M member 7 protein (TRPM7), harboring a cation channel and a serine/threonine kinase, has been implicated in controlling cell growth and proliferation. Here, we report that selective deletion of Trpm7 in β cells disrupted insulin secretion and led to progressive glucose intolerance. We indicate that the diminished insulinotropic response in β cell-specific Trpm7-knockout mice was caused by decreased insulin production because of impaired enzymatic activity of this protein. Accordingly, high-fat-fed mice with a genetic loss of TRPM7 kinase activity displayed a marked glucose intolerance accompanied by hyperglycemia. These detrimental glucoregulatory effects were engendered by reduced compensatory β cell responses because of mitigated protein kinase B (AKT)/ERK signaling. Collectively, our data identify TRPM7 kinase as a potentially novel regulator of insulin synthesis, β cell dynamics, and glucose homeostasis under obesogenic diet.

    Abstract The Reuber hepatoma H-35 and Morris hepatoma 5123 have been studied by electron microscopy and by cytochemical staining methods for a number of phosphatases. These studies emphasize the resemblances of the two tumors to rat liver, but they also indicate distinctive features in each of the three tissues. Secretory product accumulates within the cisternae of the Golgi apparatus that dilate to form the Golgi vacuoles. The vacuoles apparently separate, and secretory material undergoes further condensation within them. These "secretory vacuoles" possess acid phosphatase activity and may thus be considered lysosomes. The membranes of the Golgi apparatus are without acid phosphatase activity but show high levels of thiaminepyrophosphatase activity. The endoplasmic reticulum also hydrolyzes thiaminepyrophosphate but at a lower rate; it hydrolyzes the diphosphates of uridine, guanosine, and inosine rapidly. These observations and the electron microscopic images are consistent with the view that the cytomembranes are in a dynamic state of flux, movement, and transformation in the living cell, and that smooth surfaced derivatives of the endoplasmic reticulum become refashioned into the Golgi membranes as the Golgi membranes are being refashioned into those that delimit secretory vacuoles. The variations encountered in the two hepatomas are described. The electron microscope literature dealing with the relations of the Golgi apparatus to secretory granules, on the one hand, and the endoplasmic reticulum, on the other, is reviewed briefly.
