    Abstract This book reviews the challenges and opportunities posed by flow chemistry in drug discovery, and offers a handy reference tool for medicinal chemists interested in the synthesis of biologically active compounds. Prepared by expert contributors, the respective chapters cover not only fundamental methodologies and reactions, such as the application of catalysis, especially biocatalysis and organocatalysis; and non-conventional activation techniques, from photochemistry to electrochemistry; but also the development of new process windows, processes and reactions in drug synthesis. Particular attention is given to automatization and library synthesis, which are of great importance in the pharmaceutical industry. Readers will also find coverage on selected topics of general interest, such as how flow chemistry is contributing to drug discovery R&D in developing countries, and the green character of this enabling technology, for example in the production of raw materials for the pharmaceutical industry from waste. Given its scope, the book appeals to medicinal chemistry researchers working in academia and industry alike, as well as professionals involved in scale-up and drug development.

    Author's details Antonio de la Hoz is professor in Organic Chemistry in the University of Castilla-La Mancha. He obtained his Ph. D. from the Universidad Complutense in Madrid in 1986. After his postdoctoral research in 1987 with Prof. Mikael Begtrup at the Danmarks Tekniske Høskole, he joined the Faculty of Chemistry of the Universidad de Castilla-La Mancha in Ciudad Real in 1988. In 1993, he worked under the supervision Prof. André Loupy in the Université de Paris-Sud in Microwave Assisted Organic Chemistry. Prof. de la Hoz has authored over 220 scientific publications and he has been a foundation member of the Spanish Green Chemistry Network. His current research interests focus on Green methodologies, microwave activation, mechanochemistry, flow methodologies and solvent-free reactions, and the applications of heterocyclic compounds in material and supramolecular chemistry. Ángel Díaz-Ortiz Ángel Díaz-Ortiz obtained his PhD in Chemistry from the Complutense University of Madrid (Spain) in 1988

    Abstract The reaction of hydrazines with α,β-unsaturated carbonyl compounds to afford 2-pyrazolines was studied using a dissymmetric chalcone (phenyl/p-tolyl) and three hydrazines, hydrazine itself, phenylhydrazine and thiosemicarbazide. Several products were identified, and some reaction paths established thanks to the evolution of ¹H and ¹³C NMR spectra with time. Theoretical calculations on energies and chemical shifts were of paramount importance to ascertain the structure of some products. For important steps, the transition states were calculated while IRCs proved necessary to find some unexpected intermediates.

    Abstract A new series of donor–acceptor–donor (D–A–D) structures derived from arylethynyl 1 H -benzo[ d ]imidazole was synthesized and processed into single crystals with the goal of testing such crystals’ ability to act as optical waveguides. Some crystals displayed luminescence in the 550–600 nm range and optical waveguiding behavior with optical loss coefficients around 10 −2 dB/μm, which indicated a notable light transport. The crystalline structure, confirmed by X-ray diffraction, contains internal channels that are important for light propagation, as we previously reported. The combination of a 1D assembly, a single crystal structure, and notable light emission properties with low losses from self-absorption made 1 H -benzo[ d ]imidazole derivatives appealing compounds for optical waveguide applications.

    Abstract Objective: To determine if potential predictors for invasive mechanical ventilation (IMV) are also determinants for mortality in COVID-19-associated acute respiratory distress syndrome (C-ARDS).
    Design: Single center highly detailed longitudinal observational study.
    Setting: Tertiary hospital ICU: two first COVID-19 pandemic waves, Madrid, Spain.
    Patients or participants: 280 patients with C-ARDS, not requiring IMV on admission.
    Interventions: None.
    Main variables of interest: Target: endotracheal intubation and IMV, mortality.
    Predictors: demographics, hourly evolution of oxygenation, clinical data, and laboratory results.
    Results: The time between symptom onset and ICU admission, the APACHE II score, the ROX index, and procalcitonin levels in blood were potential predictors related to both IMV and mortality. The ROX index was the most significant predictor associated with IMV, while APACHE II, LDH, and DaysSympICU were the most with mortality.
    Conclusions: According to the results of the analysis, there are significant predictors linked with IMV and mortality in C-ARDS patients, including the time between symptom onset and ICU admission, the severity of the COVID-19 waves, and several clinical and laboratory measures. These findings may help clinicians to better identify patients at risk for IMV and mortality and improve their management.
