    Abstract The mangrove ecosystem plays a crucial role in preserving the biodiversity of plants, animals, and microorganisms that are essential for materials cycles. However, the exploration of endophytic fungi isolated from mangroves, particulary in Santa Catarina (SC, Brazil), remains limited. Therefore, the purpose of this study was to assess the biodiversity of endophytic fungi found in Avicennia schaueriana, Laguncularia racemosa, Rhizophora mangle, and Spartina alterniflora from two mangroves on the Island of Santa Catarina: one impacted by anthropic action (Itacorubi mangrove) and the other environmentally preserved (Ratones mangrove). Samplings were carried out between January 2020 and May 2021. Fungi were isolated from leaves, stems, and roots, identified, and clustered into groups through morphological characteristics. Further, a representative strain of each group was identified through ITS1 sequencing. A total of 373 isolates were obtained from plant tissues, of which 96 and 277 isolates were obtained from Itacorubi and Ratones mangroves, respectively. Molecular identification showed that the endophytic fungal community comprised at least 19 genera. The data on fungal community diversity revealed comparable diversity indices for genera in both mangroves. However, we observed differences in the total frequency of fungal genera between impacted (27.38%) and non-impacted (72.62%) mangroves. These findings suggest that anthropic activities in and around the Santa Catarina mangroves have had negative impact on the frequency of endophytic fungi. This emphasizes the reinforcing the significance of preserving these environments to ensure the maintenance of fungal community diversity.

    Abstract Mining activities generate large quantities of wastes that significantly alter the biogeochemistry and ecological structure of entire river basins. Microbial communities that develop in these areas present a variety of survival and adaptation mechanisms. Knowing this diversity at the molecular level is strategic both for understanding adaptive processes and for identifying genomes with potential use in bioremediation and bioprospecting. In this work, prokaryotic and eukaryotic communities were evaluated by meta-taxonomics (16S and 18S amplicons) in sediments and water bodies impacted by acid mine drainage in an important coal mining area in southern Brazil. Five sampling stations were defined on a gradient of impacts (pH 2.7-4.25). Taxon diversity was directly proportional to pH, being greater in sediments than in water. The dominant prokaryotic phyla in the samples were Proteobacteria, Actinobacteria, Acidobacteria, OD1, Nitrospirae, and Euryarchaeota, and among the eukaryotes, algae (Ochrophyta, Chlorophyta, Cryptophyceae), fungi (Basidiomycota, Ascomycota, and Cryptomycota), and protists (Ciliophora, Heterolobosea, Cercozoa). The prokaryotic genera Leptospirillum, Acidithiobacillus, Acidiphilium, Thiomonas, Thermogymnomonas, and Acidobacterium, and the eukaryotic genera Pterocystis and Poteriospumella were associated with more acidic conditions and higher metal concentrations, while the prokaryotic genera Sediminibacterium, Gallionella Geothrix, and Geobacter were more abundant in transitional environments.

    Abstract Mycobacterium tuberculosis (M.tb), the pathogen responsible for tuberculosis (TB) poses as the major cause of death among infectious diseases. The knowledge about the molecular diversity of M.tb enables the implementation of more effective surveillance and control measures and, nowadays, Whole Genome Sequencing (WGS) holds the potential to produce high-resolution epidemiological data in a high-throughput manner. Florianópolis, the state capital of Santa Catarina (SC) in south Brazil, shows a high TB incidence (46.0/100,000). Here we carried out a WGS-based evaluation of the M.tb strain diversity, drug-resistance and ongoing transmission in the capital metropolitan region. Resistance to isoniazid, rifampicin, streptomycin was identified respectively in 4.0% (n = 6), 2.0% (n = 3) and 1.3% (n = 2) of the 151 studied strains by WGS. Besides, resistance to pyrazinamide and ethambutol was detected in 0.7% (n = 1) and reistance to ethionamide and fluoroquinolone (FQ) in 1.3% (n = 2), while a single (0.7%) multidrug-resistant (MDR) strain was identified. SNP-based typing classified all isolates into M.tb Lineage 4, with high proportion of sublineages LAM (60.3%), T (16.4%) and Haarlem (7.9%). The average core-genome distance between isolates was 420.3 SNPs, with 43.7% of all isolates grouped across 22 genomic clusters thereby showing the presence of important ongoing TB transmission events. Most clusters were geographically distributed across the study setting which highlights the need for an urgent interruption of these large transmission chains. The data conveyed by this study shows the presence of important and uncontrolled TB transmission in the metropolitan area and provides precise data to support TB control measures in this region.

    Abstract Dados apontam um elevado número de animais errantes no Brasil. Os abrigos trabalham na tentativa de conter o aumento de animais abandonados, fornecendo um local adequado para permanência temporária e promovendo ações de conscientização da população local. O propósito deste estudo foi avaliar o perfil populacional de animais na instituição na tentativa de compreender as características de abandono e influência do abrigo na comunidade. Para elaboração deste trabalho foram utilizados dados pertencentes a uma organização não governamental da cidade de Joinville, Santa Catarina. Foram solicitadas informações referentes a características dos animais abrigados, como espécie, sexo, idade, status vacinal e reprodutivo. Os dados foram planilhados e submetidos à análise estatística. Os resultados mostraram um predomínio da população de cães em relação aos gatos. Também pode ser observado um reduzido número de animais com idade inferior a 12 meses em ambas as espécies. Quanto ao sexo, houve diferença apenas em caninos, onde o número de machos foi superior ao de fêmeas. De acordo com o protocolo da instituição, todos os animais foram submetidos a vacinação assim que admitidos e encaminhados a castração no momento que estiverem aptos para o procedimento cirúrgico. Campanhas de adoção e o incentivo à guarda responsável são mecanismos utilizados pela ONG para conscientizar a população e encontrar um lar para os animais abrigados aumentando a sua capacidade de acolhimento. A diferença na proporção de animais entre as espécies, sexo e idade tem relação direta com as características buscadas pelos adotantes. Os dados evidenciam a importância dos abrigos na diminuição dos animais errantes e promoção do bem-estar animal, sendo irrefutável a necessidade do incentivo e manutenção dessas instituições. Também é fundamental a elaboração de políticas públicas que assegurem o acesso a serviço veterinário em todas as localidades para a população impossibilitada de custear o tratamento de seus animais de estimação.

    Abstract Mycobacterium tuberculosis has a complex cell wall containing mycolic acids (MA), which play an important role in pathogenesis, virulence, and survival by protecting the cell against harsh environments. Studies have shown that genes encoding enzymes involved in MA synthesis are essential to mycobacterial functionality. Here, we used whole-genome sequencing to evaluate mutations in genes related to MA metabolism in M. tuberculosis isolates from pulmonary tuberculosis patients of the Florianópolis Metropolitan Area, Santa Catarina, Brazil, and assessed associations with clinical, epidemiological, and genotypic data. The mutations Rv3057c Asp112Ala (104/151), Rv3720 His70Arg (104/151), and Rv3802c Val50Phe (105/151) were identified in about 69% of the isolates and were related to the LAM lineage. SIT 216/LAM5 (13.2%, 20/151) had the highest frequency and presented the mutations accD2 Lys23Glu, kasA Gly269Ser, mmaA4 Asn165Ser, otsB1 Asp617Asn, Rv3057c Asp112Ala, Rv3720 His70Arg, Rv3802c Val50Phe, and tgs4 Ala216Glu. All SIT 73/T isolates (6.6%, 10/151) showed a characteristic and exclusive gene mutation pattern: amiD Rv3376 3790075G > A, fbpA-aftB 4266941G > A, echA11 Asn220fs, and otsB2 Ser110Arg. SITs 20/LAM1, 64/LAM6, 50/H3, 137/X2, and 119/X1 were also related to specific mutations. SITs from the LAM lineage differed in mutation profile from those of the T, Haarlem, and X lineages. Isolates from patients who had treatment failure showed mutations that do not seem to have a pattern related to this outcome. It was possible to identify a broad repertoire of single-nucleotide polymorphisms in genes related to MA metabolism in M. tuberculosis isolates. This study also described, for the first time, the variability between different SITs/sublineages of Lineage 4 circulating in Florianópolis Metropolitan Area.

    Abstract Objetivo: Estudar a ocorrência e identificar tendências na distribuição das cesarianas ocorridas no Estado de Santa Catarina de acordo com os grupos de classificação de Robson, entre 2014 e 2019. Método: Estudo transversal que incluiu todos os registros de microdados de partos e nascimentos do Sistema de Informações sobre Nascidos Vivos do Ministério da Saúde no período supracitado. Foram realizadas análises de tendência temporal da ocorrência de cesarianas em cada um dos 10 grupos de Robson e suas respectivas taxas e contribuições. Essas foram ainda analisadas quanto a distribuição entre as sete macrorregiões de saúde do estado. Resultados: A taxa geral de cesariana foi de 58,3% (IC95%: 58,1; 58,4), variando de 60,5% em 2014 a 57,4% em 2019. Teve destaque a macrorregião da Grande Oeste com a maior taxa geral de cesariana (67,9%). O grupo 5 (mulheres com cesariana prévia) e o grupo 2 (nulíparas com cesariana eletiva) apresentaram as maiores taxas individuais e contribuições para a taxa geral de cesariana. Enquanto o grupo 5 demonstrou tendência ascendente, o grupo 2 reduziu sua contribuição na taxa geral de cesariana no período. Conclusão: Observou-se redução discreta na ocorrência das cesarianas ao longo dos anos. Mulheres dos grupos 2 e 5 representam os principais grupos-alvo para intervenções visando a redução de cesarianas desnecessárias no Estado.

    Abstract Fascioliasis is a food-borne parasitic disease that affects a range of animals, including humans caused by Fasciola hepatica. The present study aimed to determine the spatial distribution of bovine fasciolosis and to assess the correlation between the high Positivity Index (PI) and climate data and land altitude, from 2004 to 2008 and 2010 in Santa Catarina (SC), Brazil. Condemned livers of slaughtered animals were obtained from 198 out of 293 municipalities and from 518.635 animals, exclusively from SC. There was a statistically significant difference (P < 0.001) between the prevalence of F. hepatica and land altitude ( ρ ^ s = -0.43). The highest PI (above 10.1%) was observed in cities at 500 to 600 m (P < 0.01; ρ ^ s = -0.47) of altitude. There was no correlation between fascioliasis and rainfall in SC. It was determined that weather conditions in the past decade did not impose any limitation to the occurrence of the parasite, making it a disease of permanent clinical importance. These findings are essential to regions with similar geographical and climate conditions (i.e. altitude), when considering long-term control measurements, where animals and humans can be infected.

    Abstract Objective: to investigate the time trend of prevalence of hypertension, diabetes mellitus, and both diseases combined, from 2004 to 2011; and to analyze their association with sociodemographic factors in adults registered in the Brazilian National Health System (SUS) in Florianópolis-SC, Brazil, in 2011.
    Methods: time-series study using secondary data from the Primary Health Care Information System of adults (≥20 years) at SUS; Prais-Winsten regression was used.
    Results: from 2004 to 2011, prevalences of hypertension (7 to 13.5%), diabetes (2.2 to 4.2%) and both diseases combined (1.2 to 2.9%) increased; after adjustment, the occurrence of the diseases was higher among women (except for the association of hypertension with diabetes), the elderly, less educated and residents in the Mainland and South sanitary districts.
    Conclusion: the occurrence of the investigated diseases doubled from 2004 to 2011. However, the prevalences found are below the ones described in the Brazilian scientific literature.

    Abstract Feijoa [Acca sellowaiana (O. Berg) Burret], also known as pineapple guava or goiabeira‐serrana, is a species of the Myrtaceae family native to southern Brazil and Uruguay. The genetic diversity of feijoa's natural populations has been studied and is relatively well‐documented; however, the genetic diversity of feijoa accessions in the largest active germplasm bank (AGB) in Brazil, and is still poorly documented. Characterizing and conserving pineapple guava accessions present in the AGB is essential for developing breeding programs and conserving the diversity of the species. Thus, the objective of this study was to characterize the genetic diversity of all accessions using eight microsatellite markers. The AGB collection includes accessions from 26 municipalities in Brazil, New Zealand, the United States, Israel, and Uruguay; selected clones, and selections of directed crossings, totaling 313 accessions. Genotyping 309 of these accessions with eight microsatellite markers revealed 119 alleles. Polymorphism information content (PIC) values varied between .466 (ASE40) and .890 (ASE34) with an overall mean of .718. This genotyping confirms the existence of two main types or biotypes in the species, denoted as the Uruguay type and Brazil type. Also, a significant proportion of the feijoa diversity, which is absent from the AGB‐Feijoa collection, is present in germplasm maintained by indigenous people, quilombola communities, federal conservation units, participatory genetic breeding, and areas of natural occurrence, thus reinforcing the need to introduce new accessions into São Joaquim's AGB‐Feijoa.
