    Abstract In the past, different researchers have conducted studies on incentives and how they are linked to employee motivation, influencing emerging economies. This study addresses two gaps as outlined in previous studies. One research gap exists in examining employee loyalty and employee engagement in relation to the business cycle. The other gap is observed in the recommendation that future researchers use different moderators between incentives, the health of employees, and job performance with population health. This focus was explored in the present study by identifying the responses of hospitals and physicians to the business cycle to examine the impact of incentives on job performance and health of workers in public and private sector hospitals in Shandong, Eastern China. Data were collected in the form of questionnaires that consisted of close-ended questions. These questionnaires were then filled out by 171 doctors and 149 nurses working in both public and private sectors in Shandong, Eastern China. The results showed that there is a relation between different variables. Some variables have more impact on other variables such as transformational leadership, which has a significant impact on the job performance and business cycle, whereas monetary incentives also impact job performance and population health, but this impact was lower than that of transformational leadership in terms of how job performance influences emerging economies.

    Abstract Liquid biopsy, the technique used to shed light on diseases via liquid samples, has displayed various advantages, including minimal invasiveness, low risk, and ease of multiple sampling for dynamic monitoring, and has drawn extensive attention from multidisciplinary fields in the past decade. With the rapid development of microfluidics, it has been possible to manipulate targets of interest including cells, microorganisms, and exosomes at a single number level, which dramatically promotes the characterization and analysis of disease-related markers, and thus improves the capability of liquid biopsy. However, when lab-ready techniques transfer into hospital-applicable tools, they still face a big challenge in processing raw clinical specimens, which are usually of a large volume and consist of rare targets drowned in complex backgrounds. Efforts toward the sample preparation of clinical specimens (i.e., recovering/concentrating the rare targets among complex backgrounds from large-volume liquids) are required to bridge the gap between the proof-of-concept demonstrations and practical applications. The throughput, sensitivity, and purity (TSP performance criteria) in sample preparation, i.e., the volume speed in processing liquid samples and the efficiencies of recovering rare targets and depleting the backgrounds, are three key factors requiring careful consideration when implementing microfluidic-based liquid biopsy for clinical practices. Platforms based on a single microfluidic module (single-modal microfluidics) can hardly fulfill all the aforementioned TSP performance criteria in clinical practices, which puts forward an urgent need to combine/couple multiple microfluidic modules into one working system (i.e., multi-modal microfluidics, M

    Abstract The Rice–Fish Symbiotic System in the Longxian village is the first Globally Important Agricultural Heritage System (GIAHS) in Asia. This paper examined the value transformation mechanism of the Longxian Rice–Fish Symbiotic System from the micro perspective of material cycle and energy flow and quantified its economic value using the gross ecosystem product (GEP). Using the InVEST model and ArcGIS tools, the GEP was estimated to be approximately CNY 116.51 million (approx. CNY 252,624 per hectare per year). The ecosystem regulating service value occupied more than 80% of the GEP. Within the regulating ecosystem services, the climate regulation value was the highest (CNY 55.72 million) followed by flood storage value (CNY 32.77 million). These two categories contributed 94.44% of the total value of regulating ecosystem services. The paper recommends employing GEP to evaluate the value of commodities and services provided by an ecosystem. The methodology and approach of the research can serve as an important reference for other GIAHS programs and provides a scientific and rigorous framework for accessing the economic value of agricultural heritage systems.

    Abstract The ecological environment is fragile in arid regions, and the direct and indirect impacts of continuing urbanization on vegetation growth in cities still need to be studied in depth. In this study, we focused on four provincial capital cities (Urumqi, Lanzhou, Yinchuan, and Hohhot) in arid regions of China. We used continuous 30 m land cover and vegetation greenness (VG) data from 1990 to 2021 to extract the impact of urbanization on vegetation growth by separating the impact of natural conditions. Our results showed that the study area’s urban area (UA) had expanded rapidly at a rate of 39.2 km ^2 /a, increasing by 3.39 times between 1990 and 2021. While urban expansion occurred rapidly, the overall VG of the study area also increased (slope = 2.16 × 10 ^−3 ), with the enhancement of VG increasing gradually from west to east. The VG and its trend in the UA were significantly higher than those in the natural vegetation area (NA). In addition, the duration of the urban vegetation growth season was longer than that of the NA, which also confirmed the positive impact of urbanization on the vegetation growth period. Furthermore, we found that the positive impact of urbanization on vegetation had continuously strengthened over time. In 1990 and 2021, the VG areas had 83% and 87% above the ‘zero-impact line’ in the UA, respectively, indicating a very significant impact of urbanization on vegetation growth in arid regions with fragile natural conditions. Our study identified the long-term dynamic trends in urbanization and VG in arid regions and clarified the non-linear relationship between the two concurrent growth factors. This has significant implications for correctly understanding the impact of urbanization on vegetation in arid regions and can provide a scientific reference for the ecological construction of urban environments in the region.

    Abstract Nanobodies, which represent the next generation of antibodies due to their unique properties, face a significant limitation in their poor physical adsorption on solid supports. In this study, we successfully discovered polystyrene binding nanobodies from a synthetic nanobody library. Notably, bivalent nanobody B2 exhibited high affinity for polystyrene (0.7 nM for ELISA saturation binding analysis and 15.6 nM for isothermal titration calorimetry), displaying a pH-dependent behavior. Remarkably, hydrophobic and electrostatic interactions contribute minimally to the binding process. Molecular modeling provided insights into the interaction between B2 and polystyrene, revealing that the Trp51 residue within the CDR2 loop formed an aromatic H-bond with polystyrene at a distance of 2.74 Å, thus explaining the observed reduction in B2 affinity caused by Trp51 mutations. To explore B2's potential in protein immobilization, we constructed a bispecific nanobody by fusing B2 to an anticarcinoembryonic antigen nanobody 11C12, which cannot be immobilized on polystyrene through passive adsorption. Remarkably, the fusion construct achieved effective immobilization on polystyrene within 5 min by passing the need for periplasmic protein purification despite its low expression level. Moreover, the fusion construct demonstrated excellent linearity in the chemiluminescent enzyme immunoassay. For the first time, this study reports a simplified and seamless platform for the oriented immobilization of nanobody. Importantly, the entire process eliminated the need for protein purification, enabling efficient and rapid immobilization of fusion proteins directly from crude cell extracts, even when the expression level was low. Our developed process dramatically reduced the processing time from 2.5 days to just 5 min.

    Abstract Urban vegetation experiences multiple natural and human impacts during urbanization, including land conversion, local environmental factors, and human management, which may bring positive or negative impacts on vegetation gross primary productivity (GPP) at multiple scales. In this study, we analyzed the spatial-temporal changes of GPP and three urbanization factors: land urbanization (impervious surface coverage), population urbanization (Population), and economic urbanization Gross domestic product (GDP) at city-district-grid scales in Beijing during 2000–2018. Overall, both GPP and three urbanization factors showed an increased trend. The relationships between GPP and urbanization factors exhibit diverse characteristics at multiple scales: unlike the linear relationship observed at city scale, the relationships at district and grid scales all demonstrated nonlinear relationship, even a U shape between GPP and population/GDP. Furthermore, the positive impact of urbanization on GPP increased and offset the negative impact of land conversion from 9.9% in 2000 to 35% in 2018, indicating that urban management and climate during urbanization effectively promote vegetation photosynthesis and neutralize the negative impact of urban area expansion. Our findings highlight the increased growth offset by urbanization on vegetation and the importance of analysis at a finer scale. Understanding these urbanization types’ impact on vegetation is pivotal in formulating comprehensive strategies that foster sustainable urban development and preserve ecological balance.

    Abstract Tumor microenvironment is the general term for all non-cancer components and their metabolites in tumor tissue. These components include the extracellular matrix, fibroblasts, immune cells, and endothelial cells. In the early stages of tumors, the tumor microenvironment has a tumor suppressor function. As the tumor progresses, tumor immune tolerance is induced under the action of various factors, such that the tumor suppressor microenvironment is continuously transformed into a tumor-promoting microenvironment, which promotes tumor immune escape. Eventually, tumor cells manifest the characteristics of malignant proliferation, invasion, metastasis, and drug resistance. In recent years, stress effects of the extracellular matrix, metabolic and phenotypic changes of innate immune cells (such as neutrophils, mast cells), and adaptive immune cells in the tumor microenvironment have been revealed to mediate the emerging mechanisms of immune tolerance, providing us with a large number of emerging therapeutic targets to relieve tumor immune tolerance. Gastric cancer is one of the most common digestive tract malignancies worldwide, whose mortality rate remains high. According to latest guidelines, the first-line chemotherapy of advanced gastric cancer is the traditional platinum and fluorouracil therapy, while immunotherapy for gastric cancer is extremely limited, including only Human epidermal growth factor receptor 2 (HER-2) and programmed death ligand 1 (PD-L1) targeted drugs, whose benefits are limited. Clinical experiments confirmed that cytotoxic T-lymphocyte-associated protein 4 (CTLA-4), vascular endothelial growth factor receptor (VEGFR) and other targeted drugs alone or in combination with other drugs have limited efficacy in patients with advanced gastric cancer, far less than in lung cancer, colon cancer, and other tumors. The failure of immunotherapy is mainly related to the induction of immune tolerance in the tumor microenvironment of gastric cancer. Therefore, solving the immune tolerance of tumors is key to the success of gastric cancer immunotherapy. In this study, we summarize the latest mechanisms of various components of the tumor microenvironment in gastric cancer for inducing immune tolerance and promoting the formation of the malignant phenotype of gastric cancer, as well as the research progress of targeting the tumor microenvironment to overcome immune tolerance in the treatment of gastric cancer.

    Abstract Liquid biopsy, an important enabling technology for early diagnosis and dynamic monitoring of cancer, has drawn extensive attention in the past decade. With the rapid developments of microtechnology, it has been possible to manipulate cells at the single-cell level, which dramatically improves the liquid biopsy capability. As the microtechnology-enabled liquid biopsy matures from proof-of-concept demonstrations towards practical applications, a main challenge it is facing now is to process clinical samples which are usually of a large volume while containing very rare targeted cells in complex backgrounds. Therefore, a high-throughput liquid biopsy which is capable of processing liquid samples with a large volume in a reasonable time along with a high recovery rate of rare targeted cells from complex clinical liquids is in high demand. Moreover, the purity, viability and release feasibility of recovered targeted cells are the other three key impact factors requiring careful considerations. To date, among the developed techniques, micropore-type filtration has been acknowledged as the most promising solution to address the aforementioned challenges in practical applications. However, the presently reported studies about micropore-type filtration are mostly based on trial and error for device designs aiming at different cancer types, which requires lots of efforts. Therefore, there is an urgent need to investigate and elaborate the fundamental theories of micropore-type filtration and key features that influence the working performances in the liquid biopsy of real clinical samples to promote the application efficacy in practical applications. In this review, the state of the art of microtechnology-enabled filtration is systematically and comprehensively summarized. Four key features of the filtration, including throughput, purity, viability and release feasibility of the captured targeted cells, are elaborated to provide the guidelines for filter designs. The recent progress in the filtration mode modulation and sample standardization to improve the filtration performance of real clinical samples is also discussed. Finally, this review concludes with prospective views for future developments of filtration-based liquid biopsy to promote its application efficacy in clinical practice.
