Abstract Approximately 15% of colorectal cancer (CRC) cases present with high levels of microsatellite instability (MSI-H). Bulk RNA-sequencing approaches have been employed to elucidate transcriptional differences between MSI-H and microsatellite stable (MSS) CRC tumors. These approaches are frequently confounded by the complex cellular heterogeneity of tumors. We performed single-cell deconvolution of bulk RNA-sequencing on The Cancer Genome Atlas colon adenocarcinoma (TCGA-COAD) dataset. Cell composition within each dataset was estimated using CIBERSORTx. Cell composition differences were analyzed using linear regression. Significant differences in abundance were observed for 13 of 19 cell types between MSI-H and MSS/MSI-L tumors in TCGA-COAD. This included a novel finding of increased enteroendocrine (

Abstract The social work grand challenge to ensure healthy youth development necessarily involves a focus on violence prevention, including the prevention of sexual and dating violence during adolescence. The experience of sexual and dating violence is associated with numerous and often long-lasting detrimental mental, physical, and social outcomes, many of which this grand challenge seeks to prevent. Although evidence shows that gender is a critical axis of identity to consider in violence prevention research and practice efforts, gender is not a central lens applied in the field's approach to such issues within this grand challenge. First, this article articulates a rationale for infusing a gender analysis into understanding sexual and dating violence and conceptualizing effective violence prevention strategies. Second, authors describe gender-transformative (GT) approaches to sexual and dating violence prevention, a promising practice for both ensuring the healthy development of youths and reducing violence perpetration by focusing on engaging boys and young men. Third, authors discuss the potential for GT strategies to be used in prevention efforts targeting adolescent social issues more broadly. Last, authors call for practitioners and researchers focused on healthy youth development to apply a gender analysis in their efforts and articulate concrete ways to do so.

Abstract Numerous demographic factors have been associated with colorectal cancer (CRC) risk. To better define biological mechanisms underlying these associations, we performed RNA sequencing of stem-cell-enriched organoids derived from the healthy colons of seven European Americans and eight African Americans. A weighted gene co-expression network analysis was performed following RNA sequencing. Module-trait relationships were determined through the association testing of each module and five CRC risk factors (age, body mass index, sex, smoking history, and race). Only modules that displayed a significantly positive correlation for gene significance and module membership were considered for further investigation. In total, 16 modules were associated with known CRC risk factors (

Abstract Reports of declines in abundance and biomass of insects and other invertebrates from around the world have raised concerns about food limitation that could have profound impacts for insectivorous species. Food availability can clearly affect species; however, there is considerable variation among studies in whether this effect is evident, and thus a lack of clarity over the generality of the relationship. To understand how decreased food availability due to invertebrate declines will affect bird populations, we conducted a systematic review and used meta-analytic structural equation modelling, which allowed us to treat our core variables of interest as latent variables estimated by the diverse ways in which researchers measure fecundity and chick body condition. We found a moderate positive effect of food availability on chick body condition and a strong positive effect on reproductive success. We also found a negative relationship between chick body condition and reproductive success. Our results demonstrate that food is generally a limiting factor for breeding songbirds. Our analysis also provides evidence for a consistent trade-off between chick body condition and reproductive success, demonstrating the complexity of trophic dynamics important for these vital rates.
