    Abstract Nodulation results of a host-specific association between rhizobia and leguminous plants. However, it has been observed a rich and diverse bacterial community inside nodules. This study assessed the bacterial community in nodule of cowpea grown in sites with distinct soil properties to address the hypothesis that these differences could influence the bacterial community inside the nodules. Soil samples and nodules of cowpea were collected in six different sites from Northeastern, Brazil. There was a different bacterial community structure and composition comparing bulk soil and nodules, with lower diversity in the last. However, the bacterial communities inside the nodules were not different comparing all sites. The bacterial communities found inside the nodules belonged to Proteobacteria (Bradyrhizobium and Rhizobium with 97% and 2.63%, respectively), Actinobacteria (Conexibacter and Amycolatopsis with 0.047% and 0.015%, respectively), and Firmicutes (Bacillus and Salipaludibacillus with 0.042% and 0.040%, respectively). Our analysis also showed that bulk soil and nodules presented 99.3% and 0.03% of exclusive OTUs, respectively, while these pools shared 0.4% of the bacterial groups. These results indicate that cowpea selected a specific bacterial group that colonizes the nodules, and it is not influenced by soil properties.

    Abstract A DNA vaccine expressing dengue-4 virus premembrane (prM) and envelope (E) genes was produced by inserting these genes into a mammalian expression plasmid (pCI). Following a thorough screening, including confirmation of protein expression in vitro, a recombinant clone expressing these genes was selected and used to immunize BALB/c mice. After 3 immunizations all the animals produced detectable levels of neutralizing antibodies against dengue-4 virus. The cytokines levels and T cell proliferation, detected ex vivo from the spleen of the immunized mice, showed that our construction induced substantial immune stimulation after three doses. Even though the antibody levels, induced by our DNA vaccine, were lower than those obtained in mice immunized with dengue-4 virus the levels of protection were high with this vaccine. This observation is further supported by the fact that 80% of the vaccine immunized group was protected against lethal challenge. In conclusion, we developed a DNA vaccine employing the genes of the prM and E proteins from dengue-4 virus that protects mice against this virus.

    Abstract To investigate the prevalence of Campylobacter and Salmonella in poultry carcasses in state of Rio de Janeiro, Brazil, 60 samples from 6 slaughterhouses were collected over a period of 6 months. A total of 82 Campylobacter isolates were obtained from twenty seven (45%) positive chicken carcasses, including 44 isolates (53.66%) of Campylobacter jejuni and 38 (46.34%) of Campylobacter coli. The identification of all strains was confirmed by PCR. Salmonella was isolated from 4 (6.67%) carcasses by conventional method and was detected in 5 (8.33%) of 60 chicken carcasses by PCR. Two Salmonella Albany and two Salmonella Typhimurium were identified. Antimicrobial susceptibility testing was primarily done by the disk diffusion method and later by assessing minimum inhibitory concentrations (MICs) against all the isolates. All the Campylobacter isolates were resistant to ciprofloxacin and enrofloxacin. It was observed high MIC values for enrofloxacin (64 μg/mL) in one C. jejuni and two C. coli strains, and for ciprofloxacin (≥128 μg/mL) in one C. jejuni and three C. coli strains. No Salmonella isolate was resistant to these antibiotics by both methods. These findings reveal a broad extent of fluoroquinolone resistance in Campylobacter isolates from chicken carcasses in Brazil and underline the need for prudent use of these antibiotics in poultry production to minimize the spread of fluoroquinolone resistant Campylobacter.

    Abstract To identify generational differences in the dietary patterns of Brazilian adults born between 1934 and 1975. A cross-sectional study from the baseline of the multicentre Brazilian Longitudinal Study of Adult Health (ELSA-Brasil) cohort. Year of birth was categorized into three birth generations: Traditionalists (born between 1934 and 1945); Baby Boomers (born between 1946 and 1964); and Generation X (born between 1965 and 1975). Food consumption was investigated using an FFQ. Latent class analysis (LCA) was used to identify data-driven dietary patterns. Brazil. Individuals (n 15 069) aged 35–74 years. A three-class model was generated from the LCA for each birth generation. Generation X presented higher energy intakes (kJ/kcal) from soft drinks (377·4/90·2) and sweets (1262·3/301·7) and lower energy intakes from fruit (1502·5/359·1) and vegetables (311·3/74·4) than Baby Boomers (283·7/67·8, 1047·7/250·4, 1756·0/419·7 and 365·3/87·3, respectively) and Traditionalists (186·2/44·5, 518·8/124·0, 1947·7/465·5 and 404·6/96·7, respectively). For Baby Boomers and Generation X, we found food patterns with similar structures: mixed pattern (22·7 and 29·7 %, respectively), prudent pattern (43·5 and 34·9 %, respectively) and processed pattern (33·8 and 35·4 %, respectively). Among Traditionalists, we could also identify mixed (30·9 %) and prudent (21·8 %) patterns, and a third pattern, named restricted dietary pattern (47·3 %). The younger generation presented higher frequencies of consuming a pattern characterized by a low nutritional diet, compared with other generations, indicating that they may age with a greater burden of chronic diseases. It is important to develop public health interventions promoting healthy foods, focusing on the youngest generations.

    Abstract The Neotropical region hosts 4225 freshwater fish species, ranking first among the world's most diverse regions for freshwater fishes. Our NEOTROPICAL FRESHWATER FISHES data set is the first to produce a large‐scale Neotropical freshwater fish inventory, covering the entire Neotropical region from Mexico and the Caribbean in the north to the southern limits in Argentina, Paraguay, Chile, and Uruguay. We compiled 185,787 distribution records, with unique georeferenced coordinates, for the 4225 species, represented by occurrence and abundance data. The number of species for the most numerous orders are as follows: Characiformes (1289), Siluriformes (1384), Cichliformes (354), Cyprinodontiformes (245), and Gymnotiformes (135). The most recorded species was the characid Astyanax fasciatus (4696 records). We registered 116,802 distribution records for native species, compared to 1802 distribution records for nonnative species. The main aim of the NEOTROPICAL FRESHWATER FISHES data set was to make these occurrence and abundance data accessible for international researchers to develop ecological and macroecological studies, from local to regional scales, with focal fish species, families, or orders. We anticipate that the NEOTROPICAL FRESHWATER FISHES data set will be valuable for studies on a wide range of ecological processes, such as trophic cascades, fishery pressure, the effects of habitat loss and fragmentation, and the impacts of species invasion and climate change.     Abstract A DNA vaccine expressing dengue-4 virus premembrane (prM) and envelope (E) genes was produced by inserting these genes into a mammalian expression plasmid (pCI). Following a thorough screening, including confirmation of protein expression in vitro, a recombinant clone expressing these genes was selected and used to immunize BALB/c mice. After 3 immunizations all the animals produced detectable levels of neutralizing antibodies against dengue-4 virus. The cytokines levels and T cell proliferation, detected ex vivo from the spleen of the immunized mice, showed that our construction induced substantial immune stimulation after three doses. Even though the antibody levels, induced by our DNA vaccine, were lower than those obtained in mice immunized with dengue-4 virus the levels of protection were high with this vaccine. This observation is further supported by the fact that 80% of the vaccine immunized group was protected against lethal challenge. In conclusion, we developed a DNA vaccine employing the genes of the prM and E proteins from dengue-4 virus that protects mice against this virus.
