Abstract A combined convolutional autoencoder-recurrent neural network machine learning model is presented to analyse and forecast the dynamics and low-order statistics of the local convective heat flux field in a two-dimensional turbulent Rayleigh-B\'{e}nard convection flow at Prandtl number ${\rm Pr}=7$ and Rayleigh number ${\rm Ra}=10^7$. Two recurrent neural networks are applied for the temporal advancement of flow data in the reduced latent data space, a reservoir computing model in the form of an echo state network and a recurrent gated unit. Thereby, the present work exploits the modular combination of three different machine learning algorithms to build a fully data-driven and reduced model for the dynamics of the turbulent heat transfer in a complex thermally driven flow. The convolutional autoencoder with 12 hidden layers is able to reduce the dimensionality of the turbulence data to about 0.2 \% of their original size. Our results indicate a fairly good accuracy in the first- and second-order statistics of the convective heat flux. The algorithm is also able to reproduce the intermittent plume-mixing dynamics at the upper edges of the thermal boundary layers with some deviations. The same holds for the probability density function of the local convective heat flux with differences in the far tails. Furthermore, we demonstrate the noise resilience of the framework which suggests the present model might be applicable as a reduced dynamical model that delivers transport fluxes and their variations to the coarse grid cells of larger-scale computational models, such as global circulation models for the atmosphere and ocean.

Abstract Livestock data on antimicrobial resistance (AMR) are commonly collected from bacterial populations of clinical and non-clinical isolates. In contrast to data on non-clinical isolates from livestock, data on clinical isolates are not harmonized in Europe. The Normalized Resistance Interpretation (NRI) method was applied to overcome the lack of harmonization of laboratory methods and interpretation rules between monitoring systems. Statistical analyses were performed to identify associations between the isolate type (clinical vs. non-clinical) and resistance to four antimicrobials (ampicillin, tetracycline, gentamicin, and nalidixic acid) per animal category in Germany and France. Additional statistical analyses comparing clinical and non-clinical isolates were performed with the available data on the same antimicrobial panel and animal categories from the UK and Norway. Higher resistance prevalence was found in clinical isolates compared to non-clinical isolates from calves to all antimicrobials included in Germany and France. It was also found for gentamicin in broilers from France. In contrast, in broilers and turkeys from Germany and France and in broilers from the UK, a higher resistance level to ampicillin and tetracycline in non-clinical isolates was encountered. This was also found in resistance to gentamicin in isolates from turkeys in Germany. Resistance differed within countries and across years, which was partially in line with differences in antimicrobial use patterns. Differences in AMR between clinical and non-clinical isolates of Escherichia coli are associated with animal category (broiler, calf, and turkey) and specific antimicrobials. The NRI method allowed comparing results of non-harmonized AMR systems and might be useful until international harmonization is achieved.

Abstract Two proteins of the Escherichia coli membrane protein complex, CsgG and CsgF, are studied as proteinaceous nanopores for DNA sequencing. It is highly desirable to control the DNA as it moves through the pores, this requires characterisation of DNA translocation and subsequent optimization of the pores. In order to inform protein engineering to improve the pores, we have conducted a series of molecular dynamics simulations to characterise the mechanical strength and conformational dynamics of CsgG and the CsgG-CsgF complex and how these impact ssDNA, water and ion movement. We find that the barrel of CsgG is more susceptible to damage from external electric fields compared to the protein vestibule. Furthermore, the presence of CsgF within the CsgG-CsgF complex enables the complex to withstand higher electric fields. We find that the eyelet loops of CsgG play a key role in both slowing the translocation rate of DNA and modulating the conductance of the pore. CsgF also impacts the DNA translocation rate, but to a lesser degree than CsgG.

Abstract Using 226 x 10(6) BB[overline] events recorded on the Upsilon(4S) resonance with the BABAR detector at the SLAC e(+)e(-) PEP-II storage rings, we reconstruct B(-) ---> D(*0)e(-)nu[overline](e) decays using the decay chain D(*0) --> D(0)pi(0) and D(0)-->K(-)pi(+). From the dependence of their differential rate on w, the dot product of the four velocities of B(-) and D(*0), and using the form factor description by Caprini et al. with the parameters F(1) and rhoA(1)(2), we obtain the results rhoA(1)(2) = 1.16+/-0.06+/-0.08, F(1)|V(cb)|=(35.9+/-0.6+/-1.4) x 10(-3), and B(B(-) --> D(*0)e(-)nu[overline](e))=(5.56+/-0.08+/-0.41)%.
