    Abstract Evidence for the singly Cabibbo suppressed decay $\Lambda_c^+\to p\pi^0$ is reported for the first time with a statistical significance of $3.7\sigma$ based on 6.0 $\mathrm{fb}^{-1}$ of $e^+e^-$ collision data collected at center-of-mass energies between 4.600 and 4.843 GeV with the BESIII detector at the BEPCII collider. The absolute branching fraction of $\Lambda_c^+\to p\pi^0$ is measured to be $(1.56^{+0.72}_{-0.58}\pm0.20)\times 10^{-4}$, which distinctly exceeds the upper limit measured by Belle experiment. Combining with the branching fraction of $\Lambda_c^+\to n\pi^+$, $(6.6\pm1.3)\times10^{-4}$, the ratio of the branching fractions of $\Lambda_c^+\to n\pi^+$ and $\Lambda_c^+\to p\pi^0$ is calculated to be $3.2^{+2.2}_{-1.2}$. As an important input for the theoretical models describing the decay mechanisms of charmed baryons, our result indicates that the non-factorizable contributions play an essential role and their interference with the factorizable contributions should not be significant. In addition, the absolute branching fraction of $\Lambda_c^+\to p\eta$ is measured to be $(1.63\pm0.31_{\rm stat}\pm0.11_{\rm syst}) \times10^{-3}$, which is consistent with previous measurements.

    Abstract The prompt production of the charm baryon Λ_{c}^{+} and the Λ_{c}^{+}/D^{0} production ratios were measured at midrapidity with the ALICE detector in pp and p-Pb collisions at sqrt[s_{NN}]=5.02  TeV. These new measurements show a clear decrease of the Λ_{c}^{+}/D^{0} ratio with increasing transverse momentum (p_{T}) in both collision systems in the range 2<p_{T}<12  GeV/c, exhibiting similarities with the light-flavor baryon-to-meson ratios p/π and Λ/K_{S}^{0}. At low p_{T}, predictions that include additional color-reconnection mechanisms beyond the leading-color approximation, assume the existence of additional higher-mass charm-baryon states, or include hadronization via coalescence can describe the data, while predictions driven by charm-quark fragmentation processes measured in e^{+}e^{-} and e^{-}p collisions significantly underestimate the data. The results presented in this Letter provide significant evidence that the established assumption of universality (colliding-system independence) of parton-to-hadron fragmentation is not sufficient to describe charm-baryon production in hadronic collisions at LHC energies.

    Abstract Cyclin D2 (CCND2) stabilization underpins a range of macrocephaly-associated disorders through mutation of CCND2 or activating mutations in upstream genes encoding PI3K-AKT pathway components. Here, we describe three individuals with overlapping macrocephaly-associated phenotypes who carry the same recurrent de novo c.179G>A (p.Arg60Gln) variant in Myc-associated factor X (MAX). The mutation, located in the b-HLH-LZ domain, causes increased intracellular CCND2 through increased transcription but it does not cause stabilization of CCND2.     Abstract Using a sample of $1.06\times10^{8}~\psi(3686)$ events collected with the BESIII detector at BEPCII, the decays $\eta_{c}(2S)\to p\bar{p}$ and $h_{c}\to p\bar{p}$ are searched for, where $\eta_c(2S)$ and $h_c$ are reconstructed in the decay chains $\psi(3686)\to\gamma\eta_{c}(2S)$, $\eta_{c}(2S)\to p\bar{p}$ and $\psi(3686)\to\pi^{0}h_{c}$, $h_{c}\to p\bar{p}$, respectively. No significant signals are observed. The upper limits of the product branching fractions are determined to be $\mathcal{B}(\psi(3686)\to\gamma\eta_c(2S))\times\mathcal{B}(\eta_{c}(2S)\to p\bar{p})<1.4\times10^{-6}$ and $\mathcal{B}(\psi(3686)\to\pi^0h_c)\times\mathcal{B}(h_{c}\to p\bar{p})<1.3\times10^{-7}$ at the 90% C.L. The branching fractions for $\chi_{cJ}\to p\bar{p}$ $(J=0,~1,~2)$ are also measured to be $(24.5\pm0.8\pm1.3,~8.6\pm0.5\pm0.5,~8.4\pm0.5\pm0.5)\times10^{-5}$, which are the world's most precise measurements.

    Abstract Using 567 $pb^{-1}$ of data collected with the BESIII detector at a center-of-mass energy of $\sqrt{s}=$ 4.599 $GeV$, near the $\Lambda_{c}^{+}\Lambda_{c}^{-}$ threshold, we study the singly Cabibbo-suppressed decays $\Lambda_c^{+}\to p\pi^{+}\pi^{-}$ and $\Lambda_c^{+}\to pK^{+}K^{-}$. By normalizing with respect to the Cabibbo-favored decay $\Lambda_c^{+}\to pK^{-}\pi^{+}$, we obtain ratios of branching fractions: $\frac{\mathcal{B}(\Lambda_c^{+}\to p\pi^{+}\pi^{-})}{\mathcal{B}(\Lambda_c^{+}\to pK^{-}\pi^{+})}$ = $(6.70 \pm 0.48 \pm 0.25)\%$, $\frac{\mathcal{B}(\Lambda_c^{+}\to p\phi)}{\mathcal{B}(\Lambda_c^{+}\to pK^{-}\pi^{+})}$ = $(1.81 \pm 0.33 \pm 0.13)\%$, and $\frac{\mathcal{B}(\Lambda_c^{+}\to pK^{+}K^{-}_{\text{non-}\phi})}{\mathcal{B}(\Lambda_c^{+}\to pK^{-}\pi^{+})}$ = $(9.36 \pm 2.22 \pm 0.71)\times10^{-3}$, where the uncertainties are statistical and systematic, respectively. The absolute branching fractions are also presented. Among these measurements, the decay $\Lambda_c^{+}\to p\pi^{+}\pi^{-}$ is observed for the first time, and the precision of the branching fraction for $\Lambda_c^{+}\to pK^{+}K^{-}_{\text{non-}\phi}$ and $\Lambda_c^{+}\to p\phi$ is significantly improved.

    Abstract Patients with genetically determined deficiency of complement component 5 are usually diagnosed because of recurrent invasive Neisseria meningitidis infections. Approximately 40 individual cases have been diagnosed worldwide. Nevertheless, reports of the responsible genetic defects have been sporadic, and we know of no previous reports of C5 deficiency being associated with a number of independent meningococcal disease cases in particular communities. Here we describe C5 deficiency in seven unrelated Western Cape, South African families. Three different C5 mutations c.55C>T:p.Q19X, c.754G>A:p.A252T and c.4426C>T:p.R1476X were diagnosed in index cases from two families who had both presented with recurrent meningococcal disease. p.Q19X and p.R1476X have already been described in North American Black families and more recently p.Q19X in a Saudi family. However, p.A252T was only reported in SNP databases and was not associated with disease until the present study was undertaken in the Western Cape, South Africa. We tested for p.A252T in 140 patients presenting with meningococcal disease in the Cape Town area, and found seven individuals in five families who were homozygous for the mutation p.A252T. Very low serum C5 protein levels (0.1-4%) and correspondingly low in vitro functional activity were found in all homozygous individuals. Allele frequencies of p.A252T in the Black African and Cape Coloured communities were 3% and 0.66% and estimated homozygosities are 1/1100 and 1/22,500 respectively. In 2012 we reported association between p.A252T and meningococcal disease. Molecular modelling of p.A252T has indicated an area of molecular stress in the C5 molecule which may provide a mechanism for the very low level in the circulation. This report includes seven affected families indicating that C5D is not rare in South Africa.

    Abstract Purpose: To use (13) C magnetic resonance spectroscopy (MRS) and (31) P MRS to develop a direct assay for regional [HCO3-] in the human brain and to define brain pH and physiological response of [HCO3-] to fasting.
    Materials and methods: Seven healthy subjects underwent MRS examinations on a 1.5T MRI scanner. Subjects were well fed with repeated examinations performed after 4 and 12 hours of fasting. Proton noise decoupling (13) C MRS were acquired using pulse and acquired acquisition while (31) P MRS were acquired using a 2D chemical shift imaging method with relaxation time (TR) of 2 seconds.
    Results: Fasting brain bicarbonate concentrations (6.7 ± 2.5 mM for 12-hour fasting, P = 0.002 and 8.3 ± 2.1 mM for 4-hour fasting, P = 0.015) are significantly reduced compared to fed state (11.6 ± 1.3 mM). However, no significant difference in brain pH was observed, confirming the critical role of pCO2 in intracerebral pH homeostasis.
    Conclusion: We demonstrated that the intracellular HCO3- in human brain is readily modified by diet but appears to have no measurable effect on cerebral pH. Natural abundance (13) C can provide useful information relevant to human brain pH homeostasis by providing information for HCO3-.
