    Abstract Immunohistochemistry and all techniques that use antibodies and fluorescence are widespread, essential and irreplaceable tools used in both research laboratory settings and diagnostic pathology laboratories. The field was born approximately 80 years ago, with the idea that antibodies could be tagged with fluorescent substances and used to detect antigens in cells and microorganisms, and has vertiginously evolved since; these advances have come in all aspects of the methodology, tissue fixation, generation of antibodies, monoclonal antibodies, signal amplification, antigen retrieval, signal amplification, microscopy and have become increasingly sophisticated, from in situ hybridization, in situ proximity ligation assay, flow cytometry, comet assay, to multiplexing and green fluorescent protein reconstitution, yielding Nobel Prizes along the way and generating invaluable scientific and diagnostic advances as well as timeless beautiful images.

    Abstract Immunohistochemistry (IHC) offers a valuable method for determining the spatial distribution of proteins in cells and tissues. Fixation of tissues prior IHC enables their long-term stability and preserves tissue morphology; however, downstream analysis of protein localization within fixed samples can be complicated by cross-links formed between proteins during formalin fixation which mask target epitopes. Antigen Retrieval (AR) is a process introduced to reverse such cross-links, improving the sensitivity of antibody-based protein detection, and can be performed using protease- or heat-based approaches. Even following AR, low abundance target proteins may require additional amplification for sensitive visualization. The development of amplification approaches such as the use of biotinylated secondary antibodies with avidin-biotin complex and tyramide signal amplification greatly improve the sensitivity of IHC, enabling a wider range of epitopes to be detected when coupled with AR.

    Abstract Lately, there has been an increasing demand for materials that could improve tissue regenerative therapies and provide antimicrobial effects. Similarly, there is a growing need to develop or modify biomaterials for the diagnosis and treatment of different pathologies. In this scenario, hydroxyapatite (HAp) appears as a bioceramic with extended functionalities. Nevertheless, there are certain disadvantages related to the mechanical properties and lack of antimicrobial capacity. To circumvent them, the doping of HAp with a variety of cationic ions is emerging as a good alterative due to the different biological roles of each ion. Among many elements, lanthanides are understudied despite their great potential in the biomedical field. For this reason, the present review focuses on the biological benefits of lanthanides and how their incorporation into HAp can alter its morphology and physical properties. A comprehensive section of the applications of lanthanides-substituted HAp nanoparticles (HAp NPs) is presented to unveil the potential biomedical uses of these systems. Finally, the need to study the tolerable and non-toxic percentages of substitution with these elements is highlighted.
