    Abstract Background: Yin-Chen-Hao-Tang (YCHT) consists of three aqueous extracts from Artemisia capillaris, Gardenia sp., and prepared Rheum rhabarbarum (rhubarb) (3:2:1). YCHT is characterized by its anti-inflammatory properties in liver regulation and relief of jaundice. We aimed to study the effects and mechanisms of action of YCHT on biliary obstructive cirrhosis.
    Materials and methods: Secondary biliary fibrosis was induced in rats by bile duct ligation (BDL) and scission. One week after BDL, rats were randomly divided into a saline-treated BDL or YCHT-treated BDL group for 4 weeks. Liver function and hepatic hydroxyproline (Hyp) content were assessed. Types I and IV collagen (Col-IV), laminin, fibronectin, alpha smooth muscle actin (α-SMA), and proliferating cell nuclear antigen protein and messenger ribonucleic acid (mRNA) expression were assessed with immunohistochemistry and real-time polymerase chain reaction.
    Results: In the YCHT-treated BDL group, serum total bilirubin, total bile acids, aspartate aminotransferase, alanine aminotransferase, and γ-glutamyl transferase were lower than those in the sham-operated BDL group. The proliferation of bile ducts in hepatic tissues and the Hyp content and Col deposition were also significantly lower than those in control rats. In addition, α-SMA and Col-IV staining was less obvious, and mRNA expression of Procol-α1 (IV), platelet derived growth factor subunit B (PDGF)-B, connective tissue growth factor, and transforming growth factor-beta in proliferative biliary epithelial cells (BECs) in the YCHT-treated BDL group was significantly lower than those in controls.
    Conclusions: YCHT effectively reduces the formation of biliary obstructive cirrhosis mainly via inhibition of BEC proliferation by down-regulation of PDGF-B mRNA expression, inhibition of BEC profibrogenic paracrines, and the epithelial-mesenchymal transition pathological process.

    Abstract Yin Chen Hao Tang (YCHT) is one of the most frequently used prescriptions in the long history of traditional Chinese medicine practice. The prescription contains three Chinese herbs, namely Artemisia capillaries Thunb. (Compositae), Rheum officinale Baillon (Polygonaceae), and Gardenia jasminoids Ellis (Rubiaceae), and has been widely used to treat acute hepatitis with jaundice. In this study, the in vitro anti-HSV-1 and HSV-2 activities of the water extract of YCHT were investigated. Results showed that YCHT water extract inhibited both HSV-1 and HSV-2 infections. However, the inhibition was more effective against HSV-2 than against HSV-1. The IC(50) and IC(90) values of YCHT water extract against HSV-1 infection were in the range of 142.5-150.1 and 191.3-393.9 microg/ml, and against HSV-2 infection they were in the range of 19.6-29.4 and 42.2-97.7 microg/ml, respectively. The water extract of YCHT showed no cytotoxic effect at a concentration of 500 microg/ml or below, and had a CC(50) value of 850.7+/-1.7 microg/ml. The prescription was found to diminish HSV-2 infectivity in a dose-dependent manner, and the activity was influenced by the incubation periods and the incubation temperatures. Concurrent addition of virus with YCHT or pre-treatment of the virus with the prescription extract both protected the cells from infection. In summary, the water extract of YCHT was concluded to inhibit infections by HSV-1 and HSV-2 and this effect was likely mediated through direct inactivation of the virus infectivity.

    Abstract The environment is a key element that affects many aspects of our society, including the economy, education and talents. In this article, the main purpose is to provide statistical models, algorithms and quantitative evidence regarding environmental effect evaluation (EEE). To accomplish this investigation, I first establish a theoretical EEE model and then apply a quantile-type path-modeling algorithm in the developed EEE model at different quantile levels. In the real-data analysis, this article investigates hypotheses regarding this theoretical EEE model and illustrates the statistical performances of quantile-type path-modeling EEE estimators through bootstraps. The results mainly illustrate that the environment has indispensable impacts on the economy, education and science and technology talent directly and has indirect effects on scientific infrastructure and science and technology output. Compared with the existing classical path-modeling algorithm, quantile-type path-modeling EEE estimators make full use of quantile regression and then overcome the classical exploration of only average effects. Both the quantile-type EEE model and quantile-type path-modeling algorithm capture changes in the relations among constructs and between the constructs and observed variables, and this helps to analyze the entire distribution of the outcome variables in this EEE model.
