    Abstract Objective: Diabetic retinopathy (DR) is a severe diabetic complication that leads to severe visual impairment or blindness. He-Ying-Qing-Re formula (HF), a traditional Chinese medicinal concoction, has been identified as an efficient therapy for DR with retinal vascular dysfunction for decades and has been experimentally reported to ameliorate retinal conditions in diabetic mice. This study endeavors to explore the therapeutic potential of HF with key ingredients in DR and its underlying novel mechanisms.
    Methods: Co-expression gene modules and hub genes were calculated by weighted gene co-expression network analysis (WGCNA) based on transcriptome sequencing data from high-glucose-treated adult retinal pigment epithelial cell line-19 (ARPE-19). The chromatographic fingerprint of HF was established by ultra-performance liquid chromatography coupled with high-resolution mass spectrometry (UPLC-Q-TOF-MS). The molecular affinity of the herbal compound was measured by molecular docking. Reactive oxygen species (ROS) was measured by a DCFDA/H2DCFDA assay. Apoptosis was detected using the TUNEL Assay Kit, while ELISA, Western blot, and real-time quantitative reverse transcription polymerase chain reaction (qRT-PCR) were used for detecting the cytokine, protein, and mRNA expressions, respectively.
    Results: Key compounds in HF were identified as luteolin, paeoniflorin, and nobiletin. For WGCNA, ME-salmon ("protein deacetylation") was negatively correlated with ME-purple ("oxidative impairment") in high-glucose-treated ARPE-19. Luteolin has a high affinity for SIRT1 and P53, as indicated by molecular docking. Luteolin has a hypoglycemic effect on type I diabetic mice. Moreover, HF and luteolin suppress oxidative stress production (ROS and MDA), inflammatory factor expression (IL-6, TNF-α, IL1-β, and MCP-1), and apoptosis, as shown in the
    Conclusion: Using HF and its active compound luteolin as therapeutic agents offers a promising approach to diabetic retinopathy treatment. It primarily suppressed protein acetylation and oxidative stress via the SIRT1/P53 pathway in retinal pigment epithelial cells.

    Abstract "Shi Ying Zi" powder is a traditional Chinese herbal formula for preventing and treating coccidiosis. In our previous studies, it showed anticoccidial effects and exhibited the potential to control Eimeria tenella infection. In this research, we evaluated the antioxidation and immune effect of "Shi Ying Zi" powder and its effective active ingredient osthole on coccidiosis-infected broilers to explore the mechanism of its anticoccidial effect. We analyzed changes in the antioxidant index, the pathological changes in cecum, immune index of serum and composition of cecal flora. The results showed that the use of "Shi Ying Zi" powder and osthole alleviated the pathological changes in the cecum, spleen and bursa of Fabricius, upregulated the spleen and bursal weigh index. "Shi Ying Zi" powder of 10 g/kg effectively rocovered the contents of interleukins and immunoglobulin in serum. Osthole increased the proportion of Firmicutes, Actino-bacteria and Lactobacillus in the cecum. In summary, "Shi Ying Zi" powder and osthole have anticoccidial effects, and they also can active the immunity, antioxidant functions and upregulate the beneficial bacteria population in Eimeria tenella-infected broilers.

    Abstract Objective: To establish a gas chromatography-mass spectrometry(GC-MS) method for determination of seven sugars and sugar alcohols in infants Ying Yang Bao nutritional supplements.
    Methods: The samples were extracted with pure water and diluted with 95% ethanol. After being dried by nitrogen, methoxyamine hydrochloride oxime was dissolved in pyridine and derivatized by MSTFA. The capillary column TG-5 Ms(30 m×0. 25 mm, 0. 25 μm) was used for determination by GC-MS.
    Results: The limits of detection(LODs)were 1. 0-3. 0 mg/g and the limits of quantification(LOQs)were 3. 3-10. 0 mg/g. The average recoveries of seven kinds of sugar and sugar alcohols were 86. 7%-96. 7%, and the relative standard deviation was less than 5. 1%(n=6). The contents of seven sugars and sugar alcohols in soybean matrix nutritional supplements were determined in the range of 0. 25-13. 70 g/100 g, which was consistent with the nutrition label of the products.
    Conclusion: The method is convenient, mild and fit for batch sample analysis.

    Abstract Ethnopharmacology relevance: He-Ying-Qing-Re Formula (HF) was empirically modified from Si-Miao-Yong-An Decoction (SD), which was recorded in the literature of Divine Doctor's Secret Transmission, and has been utilized for centuries to treat vasculopathy through clearing heat and accelerating bloodstream. HF has been used as an effective holistic treatment of diabetic retinopathy (DR) for decades and experimentally reported to ameliorate retinal condition in diabetic mice.
    Aim of the study: Our study aims to investigate the effect of HF in preventing sustained hyperglycemia and hyperlipidemia-associated retinal ganglion cell (RGC) cell death and its possible mechanism.
    Materials and methods: Chromatographic fingerprint of HF was obtained upon the UPLC-based analytic system; Diabetic retinopathy was established in streptozotocin (STZ) injection-induced hyperglycemic mice; Alterations of retinal structure was assayed by H&E staining. Expression of PSD-95 and CHOP in retinae was assessed by immunofluorescence; RGC cell line (mRGC) was used for in vitro study. Cell death was analyzed by flow cytometry; Intracellular reactive oxygen species (ROS) was measured by 2',7'-dichlorofluorescin diacetate (DCFDA); Apoptosis-related proteins and signaling were monitored with immunoblotting and colorimetric assay.
    Results: Chlorogenic acid, ferulic acid, and rutin were identified in HF. HF attenuates the loss of RGCs, thinning of inner retinal layers in diabetic mice. Furthermore, expressions of Brn3a and PSD-95 were restored while CHOP level was downregulated upon HF treatment. In vitro study, HF alleviates H
    Conclusion: HF administration was a valid therapeutic approach for DR treatment, oriented at the blockade of endoplasmic reticulum- and mitochondria-dependent oxidative stress-induced retinal neurodegeneration including RGC apoptosis.

    Abstract A novel indole-3-acetic acid-producing bacterium, designated TEGT-2T, was isolated from the roots of Sinopodophyllum hexandrum collected from the Qinling Mountains in shaanxi province, northwestern China, and was subjected to a taxonomic study by using a polyphasic approach. Cells of strain TEGT-2T were Gram-stain-positive, strictly aerobic, endospore-forming rods and motile by means of peritrichous flagella. Phylogenetic analyses based on 16S rRNA gene sequences showed that strain TEGT-2T was a member of the genus Paenibacillus, exhibiting the highest sequence similarity to Paenibacillus pectinilyticus KCTC 13222T (97.9 %), Paenibacillus frigoriresistens CCTCC AB 2011150T (97.3 %), Paenibacillus ferrarius CCTCC AB 2013369T (96.9 %) and Paenibacillus alginolyticus NBRC 15375T (96.5 %). The only menaquinone detected was MK-7, and the major fatty acid was anteiso-C15 : 0. The major polar lipids were diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, two unidentified aminophospholipids, two unidentified phospholipids, an unidentified aminolipid and two unidentified lipids. meso-Diaminopimelic acid was detected in the peptidoglycan. The DNA G+C content was 46.6 mol%. DNA-DNA relatedness values for strain TEGT-2T with respect to its closest phylogenetic relatives Paenibacilluspectinilyticus KCTC 13222T and Paenibacillus. frigoriresistens CCTCC AB 2011150T were lower than 40 %. Based on the phenotypic, phylogenetic and genotypic data, strain TEGT-2T is considered to represent a novel species of the genus Paenibacillus, for which the name Paenibacillus qinlingensis sp. nov. is proposed. The type strain is TEGT-2T (=CCTCC AB 2015258T=KCTC 33806T).

    Abstract Tipping's relevance vector machine (RVM) applies kernel methods to construct basis function networks using a least number of relevant basis functions. Compared to the well-known support vector machine (SVM), the RVM provides a better sparsity, and an automatic estimation of hyperparameters. However, the performance of the original RVM purely depends on the smoothness of the presumed prior of the connection weights and parameters. Consequently, the sparsity is actually still controlled by the choice of kernel functions and/or kernel parameters. This may lead to severe underfitting or overfitting in some cases. In this research, we explicitly involve the number of basis functions into the objective of the optimization procedure, and construct the RVM by maximizing the harmony function between "hypothetical" probability distribution in the forward training pathway and "true" probability distribution in the backward testing pathway, using Xu's Bayesian Ying-Yang (BYY) harmony learning technique. The experimental results have shown that our proposed methodology can achieve both the least complexity of structure and goodness of fit to data.

    Abstract A Gram-stain-positive, strictly aerobic, rod-shaped, motile and endospore-forming bacterial strain, designated TEGR-3T, was isolated from the roots of Sinopodophyllum hexandrum collected from the Qinling Mountains in Shaanxi Province, China. Strain TEGR-3T produced siderophores and hydrolysed aesculin, starch and CM-cellulose. Phylogenetic analyses based on 16S rRNA gene sequences showed that strain TEGR-3T was a member of the genus Paenibacillus, exhibiting the highest sequence similarity to Paenibacillus endophyticus LMG 27297T (97.3 %) and Paenibacillus castaneae DSM 19417T (97.3 %). MK-7 was the only menaquinone detected and anteiso-C15 : 0 and C16 : 0 were the major fatty acids. The major polar lipids were diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, two unidentified aminophospholipids, two unidentified phospholipids and an unidentified lipid. The cell-wall peptidoglycan contained meso-diaminopimelic acid as the diagnostic diamino acid. The DNA G+C content was 45.2 mol%. DNA-DNA relatedness values for strain TEGR-3T with respect to its closest phylogenetic relatives Paenibacillus endophyticus LMG 27297T and Paenibacillus castaneae DSM 19417T were lower than 40 %. Based on the phenotypic, phylogenetic and genotypic data, strain TEGR-3T is considered to represent a novel species of the genus Paenibacillus, for which the name Paenibacillus sinopodophylli sp. nov. is proposed. The type strain is TEGR-3T (=CCTCC AB 2016047T=KCTC 33807T).
