    Abstract Traditional Chinese medicine (TCM) has been used to treat triple-negative breast cancer (TNBC), a breast cancer subtype with poor prognosis. Clinical studies have verified that the Sanyingfang formula (SYF), a TCM prescription, has obvious effects on inhibiting breast cancer recurrence and metastasis, prolonging patient survival, and reducing clinical symptoms. However, its active ingredients and molecular mechanisms are still unclear. In this study, the active ingredients of each herbal medicine composing SYF and their target proteins are obtained from the Traditional Chinese Medicine Systems Pharmacology database. Breast cancer-related genes are obtained from the GeneCards database. Major targets and pathways related to SYF treatment in breast cancer are identified by analyzing the above data. By conducting molecular docking analysis, we find that the active ingredients quercetin and luteolin bind well to the key targets KDR1, PPARG, SOD1, and VCAM1.

    Abstract Esophageal squamous cell carcinoma (ESCC) is one of the deadliest malignancies worldwide. Ying Yang 1 (YY1), a ubiquitously expressed GLI-Krüppel zinc finger transcription factor, plays a regulatory role in a variety of fundamental biological processes, such as embryonic development, growth, apoptosis, differentiation and oncogenic transformation. The purpose of this study was to investigate the expression of YY1 in normal and cancerous esophageal tissues and its function in ESCC development. We found that the expression of YY1 mRNA was significantly increased in the tumor tissues, compared with the para-tissues or normal esophageal tissues. The increased expression of YY1 in tumor samples was further confirmed by immunohistochemistry. Furthermore, the overexpression of YY1 conferred radioresistance to the ESCC TE-1 cells and resulted in markedly reduced cell proliferation. Accordingly, the small interfering RNA-mediated silencing of YY1 expression in TE-1 cells resulted in increased proliferation by enhancing the binding of P21 to Cyclin D1 and CDK4, a protein complex known to mediate cell cycle progression. Moreover, besides P21, heme oxygenase-1 (HO-1) was identified as a YY1 downstream effector, as YY1 stimulated HO-1 expression in esophageal cancer cells. YY1 mediated biological function through transcription of HO-1. Forced expression of HO-1 could moderately suppress proliferation of TE-1 cells. The expression of YY1 significantly correlated with that of HO-1 in ESCC tissues. Taken together, we demonstrated overexpression of YY1 in esophageal carcinoma and identified HO-1 as its target.

    Abstract Given the finite resources of seed production, a trade-off between seed weight and number was expected. However, it still remains unclear how the seed weight-number trade-off changes if there are some mechanisms to enhance resource acquirements during the fruit developments. Sinopodophyllum hexandrum presents furled leaves in flowering time but unfurled leaves in fruiting time, which is considered to enhance the photosynthates to fruit development. To determine the relationship between seed weight and number of S. hexandrum, we examined the altitudinal variations and the relationship between seed weight and number in the fruits from 27 populations of in 2009 and 2010. Our results showed that, along the altitudinal gradient, seed weight decreased significantly but seed number increased insignificantly, indicating that the selective pressure on seed production might come from seed dispersal in high altitude populations. There was no significant relationship between seed weight and number in most populations, but an inverse relationship was found between seed weight and number when the data from all populations were pooled, indicating the seed weight-number trade-off and resource limitation of seed production in S. hexandrum on species level. We suggested that the two unfurled leaves of S. hexandrum might contribute to the resources allocated to the fruit development via enhancing photosynthates, which might mask the seed weight-number trade-off on population level to some degree.
