    Abstract Acute Myeloid Leukemia (AML) is a malignant blood cancer with a high mortality rate. Neutrophil extracellular traps (NETs) influence various tumor outcomes. However, NET-related genes (NRGs) in AML had not yet received much attention. This study focuses on the role of NRGs in AML and their interaction with the immunological microenvironment. The gene expression and clinical data of patients with AML were downloaded from the TCGA-LAML and GEO cohorts. We identified 148 NRGs through the published article. Univariate Cox regression was used to analyze the association of NRGs with overall survival (OS). The least absolute shrinkage and selection operator were utilized to assess the predictive efficacy of NRGs. Kaplan-Meier plots visualized survival estimates. ROC curves assessed the prognostic value of NRG-based features. A nomogram, integrating clinical information and prognostic scores of patients, was constructed using multivariate logistic regression and Cox proportional hazards regression models. Twenty-seven NRGs were found to significantly impact patient OS. Six NRGs-CFTR, ENO1, PARVB, DDIT4, MPO, LDLR-were notable for their strong predictive ability regarding patient survival. The ROC values for 1-, 3-, and 5-year survival rates were 0.794, 0.781, and 0.911, respectively. In the training set (TCGA-LAML), patients in the high NRG risk group showed a poorer prognosis (p < 0.001), which was validated in two external datasets (GSE71014 and GSE106291). The 6-NRG signature and corresponding nomograms exhibit superior predictive accuracy, offering insights for pre-immune response evaluation and guiding future immuno-oncology treatments and drug selection for AML patients.

    Abstract Fyn has been proven to be involved in various cell behaviors and pathophysiological processes. However, the expression and roles of Fyn in trophoblasts remain unclear. Here, we aimed to evaluate the participation of Fyn in trophoblast behavior and function, and the related mechanisms were briefly explored. Fyn expression in the HTR-8/SVneo, JEG-3, and JAR cell lines was evaluated by immunofluorescence, quantitative real-time PCR and western blotting. Fyn expression in human hydatidiform moles was also determined by immunohistochemistry and western blot. To explore the effects of Fyn, HTR-8/SVneo and JEG-3 cells were transfected with Fyn shRNA or overexpression plasmid or treated with the Fyn activity inhibitor SU6656 or ERK1/2 inhibitor U0126. The migration, proliferation, and apoptosis of trophoblast cells were assessed using transwell assays, flow cytometry, and cell counting kit-8 assays, respectively. The production of primary inflammatory cytokines, HLA-G and active matrix metallopeptidase (MMP) 2/9, and the phosphorylation of ERK1/2 and STAT3 were evaluated by ELISA, western blot, or gelatin zymography. The results showed that Fyn was expressed by trophoblast cells, mainly in the cytoplasm and membrane. Fyn expression and activity levels both increased in order from HTR-8/SVneo and JAR to JEG-3. The overexpression of Fyn promoted the proliferation and migration of trophoblast cells and inhibited their apoptosis, while the opposite effects were observed for Fyn knockdown and inhibition.     Abstract Understanding the combination of conservation tillage and straw mulching on water utilization is important for developing water management strategies in arid and semiarid areas. Few studies have investigated the effects of different tillage treatments on precipitation efficiency in winter wheat (Triticum aestivum L.)‐summer maize (Zea mays L.) cropping systems. A 3‐year field experiment with five treatments, that is, conventional tillage (CT), no‐tillage, no‐tillage with straw mulching (NT + S), subsoiling tillage, and subsoiling tillage with straw mulching (ST + S), was conducted. The objective was to investigate how conservation tillage with straw mulching affected soil water storage (SWC), precipitation storage efficiency (PSE), water consumption, grain yield, water use efficiency (WUE), and precipitation use efficiency (PUE) in a winter wheat‐summer maize cropping system. The results indicated that the ST + S treatment increased SWC by 10.1% and 8.3% in 2014–2015 and 2015–2016, respectively. The average PSE over the 3 years under the ST + S treatment was significantly higher, by 9.1% and 6.6%, than that under the CT treatment during the respective wheat and maize seasons. Compared with CT, the average annual yield under the NT + S treatment and ST + S treatment increased by 8.5% and 14.4%, respectively. In the winter wheat‐summer maize system, the NT + S and ST + S treatments significantly increased the annual WUE and PUE. The annual average PUE under the ST + S treatment was significantly higher, by 14.1%, than that under CT over the 3 years. Therefore, the ST + S treatment was more beneficial than the other treatments for increasing yield, WUE and PUE in sandy loam soil on the North China Plain.

    Abstract Abstract Background Macrophages phenotypic deviation and immune imbalance play vital roles in pregnancy-associated diseases such as spontaneous miscarriage. Trophoblasts regulate phenotypic changes in macrophages, however, their underlying mechanism during pregnancy remains unclear. Therefore, this study aimed to elucidate the potential function of trophoblast-derived miRNAs (miR-410-5p) in macrophage polarization during pregnancy. Methods Patient decidual macrophage tissue samples in spontaneous abortion group and normal pregnancy group (those who had induced abortion for non-medical reasons) were collected at the Reproductive Medicine Center of Renmin Hospital of Wuhan University from April to December 2021. Furthermore, placental villi and decidua tissue samples were collected from patients who had experienced a spontaneous miscarriage and normal pregnant women for validation and subsequent experiments at the Shenzhen Zhongshan Obstetrics & Gynecology Hospital (formerly Shenzhen Zhongshan Urology Hospital), from March 2021 to September 2022. As an animal model, 36 female mice were randomly divided into six groups as follows: naive-control, lipopolysaccharide-model, agomir-negative control prevention, agomir-410-5p prevention, agomir-negative control treatment, and agomir-410-5p treatment groups. We analyzed the miR-410-5p expression in abortion tissue and plasma samples; and supplemented miR-410-5p to evaluate embryonic absorption in vivo. The main source of miR-410-5p at the maternal–fetal interface was analyzed, and the possible target gene, signal transducer and activator of transcription (STAT) 1, of miR-410-5p was predicted. The effect of miR-410-5p and STAT1 regulation on macrophage phenotype, oxidative metabolism, and mitochondrial membrane potential was analyzed in vitro.     Abstract Extracorporeal membrane oxygenation (ECMO) is a life support system used in the treatment of severe respiratory and circulatory failure. High shear stress caused by the high rotational speed of centrifugal blood pumps can cause hemolysis and platelet activation, which are among the major factors leading to the complications of the ECMO system. In this study, a novel blood pump named rotary displacement blood pump (RDBP), which can considerably reduce rotational speed and shear stress while ensuring the normal pressure flow relationship, was proposed. We employed computational fluid dynamics (CFD) analysis to investigate the performance of RDBP under adult ECMO support operating conditions (5 L/min with 350 mmHg). The efficiency and H-Q curves of the RDBP were calculated to evaluate its hydraulic performance, and pressure, flow patterns, and shear stress distribution were analyzed to estimate the hemodynamic characteristics in the pump. In addition, the modified index of hemolysis (MIH) was calculated for the RDBP based on a Eulerian approach. The hydraulic efficiency of the RDBP was 47.28%. The velocity distribution of flow field in the pump was relatively uniform. Most of the liquid (more than 75%) in the pump was exposed to low scale shear stress (<1 Pa), which was close to normal physiological conditions. The gap area was the main distribution location of high scale shear stress. The high wall shear stress (>9 Pa) volume fraction of the RDBP was small and located in the boundary areas between the rotor's edge and the housing. The MIH value of the RDBP was 9.87 ± 0.93 (mean ± SD). The RDBP can achieve better hydraulic efficiency and hemodynamic performance at lower rotational speed. The design of this novel pump is expected to provide a new direction for developing a blood pump for ECMO.
