    Abstract Within the islets of Langerhans, beta cells orchestrate synchronized insulin secretion, a pivotal aspect of metabolic homeostasis. Despite the inherent heterogeneity and multimodal activity of individual cells, intercellular coupling acts as a homogenizing force, enabling coordinated responses through the propagation of intercellular waves. Disruptions in this coordination are implicated in irregular insulin secretion, a hallmark of diabetes. Recently, innovative approaches, such as integrating multicellular calcium imaging with network analysis, have emerged for a quantitative assessment of the cellular activity in islets. However, different groups use distinct experimental preparations, microscopic techniques, apply different methods to process the measured signals and use various methods to derive functional connectivity patterns. This makes comparisons between findings and their integration into a bigger picture difficult and has led to disputes in functional connectivity interpretations. To address these issues, we present here a systematic analysis of how different approaches influence the network representation of islet activity. Our findings show that the choice of methods used to construct networks is not crucial, although care is needed when combining data from different islets. Conversely, the conclusions drawn from network analysis can be heavily affected by the pre-processing of the time series, the type of the oscillatory component in the signals, and by the experimental preparation. Our tutorial-like investigation aims to resolve interpretational issues, reconcile conflicting views, advance functional implications, and encourage researchers to adopt connectivity analysis. As we conclude, we outline challenges for future research, emphasizing the broader applicability of our conclusions to other tissues exhibiting complex multicellular dynamics.

    Abstract A solid methodology for trials testing the side effects of PPPs on the large earth bumblebee, Bombus terrestris dalmatinus, from the laboratory to large-scale field studies is presented with a stepwise approach. The study designs may serve various purposes, such as practical help regarding commercial pollination, safeguarding biodiversity or for risk assessments in the registration process under GLP. In order to achieve high uniformity between bumblebee colonies, specially standardized R&D colonies (IPM Impact-Koppert) are used and adapted to the different test designs. The initial step is the study under laboratory conditions, through simulating the three possible means of bumblebee exposure to the compound: acute contact, oral by sugar water and/or by pollen. The products are mainly tested according to the maximum field recommended concentration (MFRC), but a sequential dilution testing scheme may be applied to the oral sugar water exposure, if triggered. In the next step, simulating a more field-realistic but still controlled exposure scenario, R&D colonies can be tested under semi-field conditions in a tunnel set-up. The test design can be customised according to the specific requirements. The final step, if needed, is to monitor bumblebees in a field study where colonies are exposed to the product in a common agricultural landscape. In all study types, the following assessment parameters are recorded at regular intervals during the experimental phase: the presence/vitality of the mother queen, colony strength, colony weight, brood volume, the number of queen-brood cells, and the number of newly-formed queens (gynes). In a final assessment of the colonies, the reproduction rate of the control and treated colonies expressed as number of newly formed queens and, if possible, drones and the colony strength as the number of worker bees is compared at the end of the experiment.     Abstract Desertification is defined as land degradation occurring in the global drylands. It is one of the global problems targeted under the Sustainable Development Goals (SDG 15). The aim of this article is to review the history of desertification and to evaluate the scientific evidence for desertification spread and severity. First quantitative estimates of the global extent and severity of desertification were dramatic and resulted in the establishment of the UN Convention to Combat Desertification (UNCCD) in 1994. UNCCD’s task is to mitigate the negative impacts of desertification in drylands. Since the late 1990s, science has become increasingly critical towards the role of desertification in sustainable land use and food production. Many of the dramatic global assessments of desertification in the 1970s and 1980s were heavily criticized by scientists working in drylands. The used methodologies and the lack of ground-based evidence gave rise to critical reflections on desertification. Some even called desertification a myth. Later desertification assessments relied on remote sensing imagery and mapped vegetation changes in drylands. No examples of large areas completely degraded were found in the scientific literature. In science, desertification is now perceived as a local feature that certainly exists but is not as devastating as was earlier believed. However, the policy arena continues to stress the severity of the problem. Claims that millions of hectares of once productive land are annually lost due to desertification are regularly made. This highlights the disconnection between science and policy, and there is an urgent need for better dialogue in order to achieve SDG 15.

    Abstract Recently, residues of 4-acetaminophen (paracetamol) have been found in urine of slaughter animals. To investigate its possible origin, a 7 week animal experiment was performed with 5 groups of 4 calves fed different amounts of roughage, and treated or not with 4-acetaminophen. Group 1 (control): new-born calves were fed, after the first days of colostrum, solely dairy based calf milk replacer (CMR). The animals from the other groups were 2–3 weeks of age at the start of the experiment and were fed the same CMR. Group 2 was administered oral Pracetam® 30 mg acetaminophen/kg Body Weight (BW) in the CMR, 5 days a week at alternate weeks, with maximum roughage. Group 3 was fed legal minimum roughage. Group 4 received the maximum roughage. Group 5 was administered human paracetamol pills 30 mg/kg BW once a week at weeks 2–6 with maximum roughage. The animals were housed in separate pens on slatted rubber floors without bedding. The controls were housed in a different room than the other animals. Urine was sampled once in the acclimatization period and for 5 days after treatment. At week 2, 4 and 6 at respectively 1, 2 and 3 days after treatment, one animal from groups 2 and 5 was sacrificed. An additional animal from group 4 was sacrificed at week 4, the rest of the animals at day 43. Urine, muscle, liver and kidney were analysed for residues of 4-acetaminophen and its metabolites. It appeared that controls and the low and high roughage animals had almost no 4-acetaminophen (<3 μg/L) in the urine and low levels of metabolites, whereas high roughage had at slaughter higher levels of metabolites than low roughage. Pracetam® treated animals showed high levels 4-acetaminophen (up to 130.000 μg/L) and metabolites during treatment, and lower levels (2800 μg/L) at slaughter. In the paracetamol treated animals 4-acetaminophen and metabolites were detectable during treatment (up to 130.000 μg/L) and until 7 days after treatment (110 μg/L). 4-Acetaminophen and metabolites were only present in tissues of treated animals, ...
