Abstract Stripe rust, caused by Puccinia striiformis f. sp. tritici , is a serious wheat fungal disease, causing significant annual yield losses worldwide. The Chinese wheat cultivar Chuanyu 16 has shown good adult-plant resistance (APR) to stripe rust in Sichuan province, a hotspot for stripe rust epidemics. Chuanyu 16 was crossed with Chuanyu 12 and Chuanmai 32. Two populations, each with 140 recombinant inbred lines (RILs), were developed by single-seed descent, and used for quantitative trait locus (QTL) mapping. Field trials were conducted in Chengdu and Yaan from 2005 to 2008, providing stripe rust reaction data for six environments. Seven hundred and thirty one simple sequence repeat (SSR) markers were screened for association with stripe rust reaction, initially through bulked segregant analysis (BSA). Three QTLs for stripe rust resistance derived from Chuanyu 16 were detected in the first cross. They were detected by inclusive composite interval mapping (ICIM) and designated QYr.caas-1BL.1 , QYr.caas-1BL.2 and QYr.caas-2AS . They explained 6.0 - 12.8%, 4.5 - 5.8% and 14.9 - 43.0%, respectively, of the phenotypic variance across environments. One digenic epistatic QTL between QYr.caas-1BL.2 and QYr.caas-2AS explained 4.3 - 10.4% of the phenotypic variance. QYr.caas-2AS was also detected in Chuanmai 32/Chuanyu 16, explaining 27.9 - 57.2% of the phenotypic variance across six environments. This QTL showed a major effect against stripe rust in Chuanyu 16, and was located in a similar position to Yr17. Specific markers indicated the presence of a segment from chromosome 2N of Triticum ventricosum that carries Yr17 . Despite the lack of evidence for Yr17 in Chuanyu 16 based on pedigree, and inconsistencies in stripe rust response relative to a near-isogenic reference stock with the gene, we concluded that QYr.caas-2AS is Yr17 . QYr .caas-1BL.1 and QYr.caas-1BL.2 showed minor effects for APR against stripe rust. QYr.caas-1BL.1 is probably a new gene for APR to stripe rust.

Abstract Stripe rust, caused by Puccinia striiformis f. sp. tritici, is a devastating wheat disease worldwide. The Chinese wheat cultivar Chuanmai 32 has shown stable resistance to stripe rust for 10 yr in Sichuan Province, a hotspot for stripe rust epidemics. The objective of the present study was to map quantitative trait loci (QTL) for adult-plant resistance (APR) to stripe rust in a population of 140 recombinant inbred lines (RILs) derived from Chuanmai 32/Chuanyu 12. Field trials were conducted in Chengdu and Yaan, Sichuan, from 2005 to 2008, providing stripe rust reaction data for 6 environments. 797 simple sequence repeat (SSR) markers were screened for association with stripe rust reaction, initially through bulked segregant analysis (BSA). Based on the mean disease values averaged across environments, the broad-sense heritability of maximum disease severity (MDS) was 0.75. Two QTLs for stripe rust resistance were detected by composite interval mapping (CIM). They were designated QYr.caas-3BL and QYr.caas-3BS and explained from 6.6 to 20.1%, respectively, of the phenotypic variance across environments. QYr.caas-3BL came from Chuanmai 32; QYr.caas-3BS with lower effect was from the susceptible parent Chuanyu 12. Both QTLs appear to be new.

Abstract There has been a shift in focus toward environmentally and economically sustainable forms of economic growth known as High-quality economic development (HQED). However, this study analyzes the impact of tourism development (TD) and renewable energy consumption on HQED in 30 provinces of China, while covering the time period from 2007 to 2021. TD and HQED has been measured with help of Global Moran Index. This study has used dynamic spatial Durbin model (SDM) to measure the dynamic impact of TD index and renewable energy consumption on HQED along with green finance, foreign direct investment and investment in education. The findings from empirical analysis shows that TD has negative impact on HQED and in more developed regions, the relationship is positive, while in the less developed western part of China, the U-shape has been reversed. Central and northeastern China have a U-shaped connection, while it has been noticed the interaction term of TD and renewable energy endorses HQED. In addition, renewable energy consumption, green finance and increase in education investment have positive and significant impact on HQED while foreign direct investment has negative impact on HQED in China. Therefore, in the light of this study policymakers should focus on the quality of tourism industry, green finance for renewable energy supply and enhancing education investment in China to attain the goal of HQED.

Abstract Natural rubber (NR) is extensively utilized in numerous industries, such as aerospace, military, and transportation, because of its exceptional elasticity and all-around mechanical qualities. However, commercial NR made using various techniques typically has distinct mechanical characteristics. For instance, whole field latex rubber (SCR-WF) cured with accelerator 2-Mercaptobenzothiazole exhibits poor mechanical properties. This work attempts to enhance the mechanical property of SCR-WF via the addition of lanthanum stearate (LaSt). The influence of LaSt on strain-induced crystallization (SIC) and the mechanical properties of SCR-WF were investigated. The results of crosslinking density measured by the equilibrium swelling method demonstrate that the presence of LaSt significantly increases the crosslinking density of SCR-WF with lower loading of LaSt. The results of the mechanical properties show that the introduction of LaSt can enhance the tensile strength and fracture toughness of SCR-WF. To reveal the mechanism of LaSt improving the mechanical properties of SCR-WF, synchrotron radiation wide-angle X-ray diffraction (WAXD) experiments were used to investigate the SIC behaviors of SCR-WF. We found that the LaSt leads to higher crystallinity of SIC for the strain higher than 3.5. The tube model indicates the contribution of LaSt in both crosslinking and topological constraints. This work may provide an instruction for developing SCR-WF with superior mechanical properties.

Abstract Spatial transcriptomics analysis allows the examination of the biological characteristics and spatial distribution of individual lung cells at a single-cell resolution. However, due to the presence of cavities in the alveoli of the lungs, it is challenging to section them for spatial transcriptomics experiments. Here, we present a protocol for acquiring high-quality fresh mouse lung spatial transcriptomics data. We describe steps for lung perfusion, acquiring frozen slices, collecting cDNA from lung sections, and data analysis. For complete details on the use and execution of this protocol, please refer to Jiang et al.

Abstract Background: Systemic lupus erythematosus (SLE) patients have a higher risk of acute myocardial infarction (AMI) compared to the general population. However, the underlying common mechanism of this association is not fully understood. This study aims to investigate the molecular mechanism of this complication.
Methods: Gene expression profiles of SLE (GSE50772) and AMI (GSE66360) were obtained from the Gene Expression Omnibus (GEO) database. Common differentially expressed genes (DEGs) in SLE and AMI were identified, and functional annotation, protein-protein interaction (PPI) network analysis, module construction, and hub gene identification were performed. Additionally, transcription factor (TF)-gene regulatory network and TF-miRNA regulatory network were constructed for the hub genes.
Results: 70 common DEGs (7 downregulated genes and 63 upregulated genes) were identified and were mostly enriched in signaling pathways such as the IL-17 signaling pathway, TNF signaling pathway, lipid metabolism, and atherosclerosis. Using cytoHubba, 12 significant hub genes were identified, including IL1B, TNF, FOS, CXCL8, JUN, PTGS2, FN1, EGR1, CXCL1, DUSP1, MMP9, and ZFP36.
Conclusions: This study reveals a common pathogenesis of SLE and AMI and provides new perspectives for further mechanism research. The identified common pathways and hub genes may have important clinical implications for the prevention and treatment of AMI in SLE patients.

Abstract Objective: This study aimed to establish a combined method of radiomics and deep learning (DL) in magnetic resonance imaging (MRI) to predict lymph node metastasis (LNM) preoperatively in patients with squamous cell carcinoma of the tongue.
Study design: In total, MR images of 196 patients with lingual squamous cell carcinoma were divided into training (n = 156) and test (n = 40) cohorts. Radiomics and DL features were extracted from MR images and selected to construct machine learning models. A DL radiomics nomogram was established via multivariate logistic regression by incorporating the radiomics signature, the DL signature, and MRI-reported LN status.
Results: Nine radiomics and 3 DL features were selected. In the radiomics test cohort, the multilayer perceptron model performed best with an area under the receiver operating characteristic curve (AUC) of 0.747, but in the DL cohort, the best model (logistic regression) performed less well (AUC = 0.655). The DL radiomics nomogram showed good calibration and performance with an AUC of 0.934 (outstanding discrimination ability) in the training cohort and 0.757 (acceptable discrimination ability) in the test cohort. The decision curve analysis demonstrated that the nomogram could offer more net benefit than a single radiomics or DL signature.
Conclusion: The DL radiomics nomogram exhibited promising performance in predicting LNM, which facilitates personalized treatment of tongue cancer.
