Abstract Multi-center evaluations of pediatric patients with juvenile systemic sclerosis (jSSc) have suggested that the pathogenesis of jSSc may differ from that of systemic sclerosis (SSc) in adult patients. Therefore, we undertook to identify abnormalities in the T lymphocytes of jSSc patients and to determine if they differed from the abnormalities reported in the T lymphocytes of adult SSc patients. We identified decreases in the frequency of resting regulatory T lymphocytes and an increased frequency of CD45RA expressing effector memory (EMRA) CD4 T lymphocytes, which were characterized by an increased frequency of CCR7 protein expressing cells. Neither the increases in the EMRA subpopulation nor the increased CCR7 protein expression have been reported in adult SSc patients. The decrease in resting regulatory T lymphocytes in jSSc patients may permit the expansion of the disease initiating CD4 T lymphocytes present in the CCR7 expressing EMRA CD4 T lymphocyte subpopulation.

Abstract Hemophagocytic lymphohistiocytosis (HLH) is a syndrome resulting from uncontrolled hyper-inflammation, excessive immune system activation, and elevated levels of inflammatory cytokines. HLH can be caused by the inability to downregulate activated macrophages by natural killer (NK) and CD8 cytotoxic T cells through a process reliant on perforin and granzyme B to initiate apoptosis. Homozygous genetic mutations in this process result in primary HLH (pHLH), a disorder that can lead to multi-system organ failure and death in infancy. Heterozygous, dominant-negative, or monoallelic hypomorphic mutations in these same genes can cause a similar syndrome in the presence of an immune trigger, leading to secondary HLH (sHLH). A genetic mutation in a potential novel HLH-associated gene, dedicator of cytokinesis 2 (

Abstract The synthesis and characterization of the mononuclear chromium(II) terphenyl substituted primary amido-complexes Cr{N(H)Ar(Pr(i)(6))}(2) (Ar(Pr(i)(6)) = C(6)H(3)-2,6-(C(6)H(2)-2,4,6-(i)Pr(3))(2) (1), Cr{N(H)Ar(Pr(i)(4))}(2) (Ar(Pr(i)(4)) = C(6)H(3)-2,6-(C(6)H(3)-2,6-(i)Pr(2))(2) (2), Cr{N(H)Ar(Me(6))}(2) (Ar(Me(6)) = C(6)H(3)-2,6-(C(6)H(2)-2,4,6-Me(3))(2) (4), and the Lewis base adduct Cr{N(H)Ar(Me(6))}(2)(THF) (3) are described. Reaction of the terphenyl primary amido lithium derivatives Li{N(H)Ar(Pr(i)(6))} and Li{N(H)Ar(Pr(i)(4))} with CrCl(2)(THF)(2) in a 2:1 ratio afforded complexes 1 and 2, which are extremely rare examples of two coordinate chromium and the first stable chromium amides to have linear coordinated high-spin Cr(2+). The reaction of the less crowded terphenyl primary amido lithium salt Li{N(H)Ar(Me(6))} with CrCl(2)(THF)(2) gave the tetrahydrofuran (THF) complex 3, which has a distorted T-shaped metal coordination. Desolvation of 3 at about 70 °C gave 4 which has a formally two-coordinate chromous ion with a very strongly bent core geometry (N-Cr-N= 121.49(13)°) with secondary Cr--C(aryl ring) interactions of 2.338(4) Å to the ligand. Magnetometry studies showed that the two linear chromium species 1 and 2 have ambient temperature magnetic moments of about 4.20 μ(B) and 4.33 μ(B) which are lower than the spin-only value of 4.90 μ(B) typically observed for six coordinate Cr(2+). The bent complex 4 has a similar room temperature magnetic moment of about 4.36 μ(B). These studies suggest that the two-coordinate chromium complexes have significant spin-orbit coupling effects which lead to moments lower than the spin only value of 4.90 μ(B) because λ (the spin orbit coupling parameter) is positive. The three-coordinated complex 3 had a magnetic moment of 3.79 μ(B).

Abstract The origins of the extraordinarily large internal hyperfine field (62.4 T) for the three-dimensional (weak) ferromagnetically ordered ground state of alpha-Fe(OETAP) are discussed semiquantitatively in terms of existing physical theory. In particular, the classical Fermi-contact contribution to the internal field is found to be highly enhanced by a very large orbital contribution and a significant dipolar term of the same sign. A rationale for the unexpected ordering of this S = 1 non-Kramers system is also presented.

Abstract Mossbauer, EPR, magnetic susceptibility, and DFT studies of the unusual two-coordinate iron(II) amide Fe[N(t-Bu)(2)](2) show that it retains a linear N-Fe-N framework due to the nonbonding delta nature of the (xy, x(2)-y(2)) orbitals. The resulting near-degenerate ground state gives rise to a large magnetic moment and a remarkably large internal hyperfine field. The results confirm that extraordinary orbital magnetic effects can arise in linear transition metal complexes in which orbital degeneracies are not broken by Jahn-Teller or Renner-Teller distortions.
