    Abstract Background: Electronic cigarettes (e-cigarettes) are a consumer product whose benefits and risks are currently debated. Advocates of the "tobacco harm reduction" strategy emphasize their potential as an aid to smoking cessation, while advocates of the precautionary principle emphasize their risks instead. There have been only a few studies to date on the prevalence of e-cigarette use in Germany.
    Methods: In May 2016, in collaboration with Forsa, an opinion research firm, we carried out a survey among 4002 randomly chosen persons aged 14 and older, asking them about their consumption of e-cigarettes with and without nicotine, reasons for using e-cigarettes, plans for future use, estimation of danger compared to that of tobacco products, smoking behavior, and sociodemographic features.
    Results: 1.4% of the respondents used e-cigarettes regularly, and a further 2.2% had used them regularly in the past. 11.8% had at least tried them, including 32.7% of smokers and 2.3% of persons who had never smoked. 24.5% of ex-smokers who had quit smoking after 2010 had used e-cigarettes at least once. 20.7% of the respondents considered electronic cigarettes less dangerous than conventional cigarettes, 46.3% equally dangerous, and 16.1% more dangerous. An extrapolation of these data to the general population suggests that about one million persons in Germany use e-cigarettes regularly and another 1.55 million have done so in the past.
    Conclusion: The consumption of electronic cigarettes in Germany is not very widespread, but it is not negligible either. Nearly 1 in 8 Germans has tried e-cigarettes at least once. Regular consumers of e-cigarettes are almost exclusively smokers and ex-smokers.

    Abstract The disruption of ionic and H-bond interactions between the cytosolic ends of transmembrane helices TM3 and TM6 of class-A (rhodopsin-like) G protein-coupled receptors (GPCRs) is a hallmark for their activation by chemical or physical stimuli. In the bovine photoreceptor rhodopsin, this is accompanied by proton uptake at Glu(134) in the class-conserved D(E)RY motif. Studies on TM3 model peptides proposed a crucial role of the lipid bilayer in linking protonation to stabilization of an active state-like conformation. However, the molecular details of this linkage could not be resolved and have been addressed in this study by molecular dynamics (MD) simulations on TM3 model peptides in a bilayer of 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC). We show that protonation of the conserved glutamic acid alters the peptide insertion depth in the membrane, its side-chain rotamer preferences, and stabilizes the C-terminal helical structure. These factors contribute to the rise of the side-chain pKa (> 6) and to reduced polarity around the TM3 C terminus as confirmed by fluorescence spectroscopy. Helix stabilization requires the protonated carboxyl group; unexpectedly, this stabilization could not be evoked with an amide in MD simulations. Additionally, time-resolved Fourier transform infrared (FTIR) spectroscopy of TM3 model peptides revealed a different kinetics for lipid ester carbonyl hydration, suggesting that the carboxyl is linked to more extended H-bond clusters than an amide. Remarkably, this was seen as well in DOPC-reconstituted Glu(134)- and Gln(134)-containing bovine opsin mutants and demonstrates that the D(E)RY motif is a hydrated microdomain. The function of the D(E)RY motif as a proton switch is suggested to be based on the reorganization of the H-bond network at the membrane interface.

    Abstract Information transfer in the brain requires a homeostatic control of neuronal excitability. Therefore, a functional balance between excitatory and inhibitory systems is established during development. This review contains recent information about the molecular mechanisms orchestrating the establishment and maintenance of this excitation-inhibition (E-I) balance, and it reviews examples of deregulation of inhibitory and excitatory systems at a molecular, network and disease level of investigation.

    Abstract ABSTRACT Urban forest fragments surrounded by housing matrix are a source of richness and can function as a conservation site in the relation between the living beings and the environment. The present study have as aim to describe the community structure of tree and arborescent species in a urban forest fragment in the municipality of Sério, Rio Grande do Sul (RS) state, inserted in the Atlantic Forest biome, phytoecological formation Montane Deciduous Forest. The floristic inventory was produced through walking method, reaching the total fragment area, in monthly field incursions. For sampling, 40 sampling units (UA’s) of 100 m² were scattered, with distance of 20 meters in between them. Inside the UAs, it was obtained the height and circumference at the chest height (CAP) of all specimens which have CAP equal or higher than 15 cm. It was estimated the parameters of dominancy, density, frequency, absolute and relative, and the importance value index (IVI). The floristic inventory found 59 trees and arborescent species belonging to 48 genera and 30 families. In the quantitative inventory, 535 tree specimens belonging to 42 genera and 27 families were sampled. Among the species, three are exotic and three are in the threatened species list of the state of Rio Grande do Sul. The absolute density was estimated in 1,338 individuals/ha -1 . Actinostemon concolor showed the denser values (140 ind.ha -1 ), followed by Alchornea triplinervia e Eugenia rostrifolia , both with 108 ind.ha -1 . The species with the great IVI’s were Alchornea triplinervia , Vernonanthura discolor , Actinostemon concolor , Eugenia rostrifolia and Araucaria angustifolia. The estimated diversity found through the Shanon Index (Hʼ) was 3.35, and the equability of Pielou (Jʼ) was 0.85, denoting a consistent species diversity. The fragment can be seen as an important shelter for the regional wild fauna and flora.
