    Abstract Fire is an inherently evolutionary process, even though much more emphasis has been given to ecological responses of plants and their associated communities to fire. Here, we synthesize contributions to a Special Feature entitled ‘Fire as a dynamic ecological and evolutionary force’ and place them in a broader context of fire research. Topics covered in this Special Feature include a perspective on the impacts of novel fire regimes on differential forest mortality, discussions on new approaches to investigate vegetation‐fire feedbacks and resulting plant syndromes, synthesis of fire impacts on plant–fungal interactions, and a meta‐analysis of arthropod community responses to fire. We conclude by suggesting pathways forward to better understand the ecological and evolutionary consequences of fire. These include developing ecological and evolutionary databases for fire ecology, integrating hierarchical genetic structure or phylogenetic structure, and developing new experimental frameworks that limit context‐dependent outcomes.     Abstract Numerous mass spectrometry-based strategies ranging from hydrogen-deuterium exchange to ion mobility to native mass spectrometry have been developed to advance biophysical and structural characterization of protein conformations and determination of protein-ligand interactions. In this study, we focus on the use of ultraviolet photodissociation (UVPD) to examine the structure of human carbonic anhydrase II (hCAII) and its interactions with arylsulfonamide inhibitors. Carbonic anhydrase, which catalyzes the conversion of carbon dioxide to bicarbonate, has been the target of countless thermodynamic and kinetic studies owing to its well-characterized active site, binding cavity, and mechanism of inhibition by hundreds of ligands. Here, we showcase the application of UVPD for evaluating structural changes of hCAII upon ligand binding on the basis of variations in fragmentation of hCAII versus hCAII-arylsulfonamide complexes, particularly focusing on the hydrophobic pocket. To extend the coverage in the midregion of the protein sequence, a supercharging agent was added to the solutions to increase the charge states of the complexes. The three arylsulfonamides examined in this study largely shift the fragmentation patterns in similar ways, despite their differences in binding affinities.

    Abstract The focus of this study was to examine how clinicians conceptualized acculturation and societal stressors, such as discrimination among Latinx adolescent clients. This qualitative investigation of clinicians sought to understand the experiences of their work with Latinx adolescent clients in the current political and social climate. The primary participants were nineteen clinicians who responded to a web-based survey. Through thematic analysis, various themes emerged, including clinicians’ observation of pervasive ethnic/racial discrimination and negative valenced emotions such as anger, fear, and anxiety in their clients. Other themes included clinicians’ observations of Latinx adolescents’ acculturative stress and cultural pressures to maintain connection with their family traditions. Clinicians reported an overreliance on social media use among Latinx adolescents, particularly with the increased reporting of deportations among the Latinx community. The findings were discussed in the context of the current political climate. Implications for future directions and limitations were addressed in chapter six of this document.

    Abstract Sudden cardiac death (SCD) in young people is predominantly caused by genetic causes as cardiomyopathies. Hypertrophic cardiomyopathy is the most common genetic cardiovascular disease and is responsible for the major proportion of SCD in the young. The purpose of this study was to identify the genetic variants present in young SCD victims with HCM characteristics. From the Portuguese records of autopsies performed at the National Institute of Legal Medicine and Forensic Sciences, North Delegation, 16 young (16-50 years) SCD victims whose death was suspected to be a manifestation of HCM were selected. Using next-generation sequencing, the coding regions of 40 genes associated with HCM, candidates, or strongly related to HCM-phenocopies were investigated. The victims included in this study were all males, with a mean age of 33.4 ± 11.7 years, left ventricle mean thickness of 21.5 ± 6.28 mm, and the majority of deaths occurred during sleep (36%). A pathogenic or likely pathogenic variant was identified in six out of 16 (37.5%) victims, in the most common HCM genes (MYBPC3 and MYH7). Our results indicate that molecular autopsy of SCD victims contributes to a more precise identification of a cause of death, and this can be used in the prevention of SCD cases through family screening of first relatives who may carry the same pathogenic variant.

    Abstract Introduction: Work-related musculoskeletal disorders result from the overuse of the musculoskeletal system and insufficient time for the structures to recover. They are generally characterized by chronic pain, paresthesia, feeling of heaviness and fatigue, especially in the upper extremities, concomitantly or not, with an insidious onset.
    Objectives: To characterize musculoskeletal complaints and occupational risks in workers with work-related musculoskeletal disorders.
    Methods: A cross-sectional observational study of 60 participants in a Workers' Health Reference Center with clinical and imaging diagnosis of work-related musculoskeletal disorders. The instrument used contained 30 questions about individual factors, occupational risks, and musculoskeletal abnormalities. The results were analyzed descriptively, and the chi-square test was used to assess associations with a significance level at p < 0.05. Data analysis was performed using BioEstat 5®.
    Results: Most participants were men (66.7%) working in the industrial sector. The most common complaint was pain (100%) in the shoulders (43.8%) and lumbar spine (22%), and the most common abnormalities were tendinopathies and intervertebral disc disorders. The following risk factors were identified: 8-hour workday (80%); repetitive gestures (86.7%); twisting (58.3%); bending (61.7%); standing (66.7%); manual work (96.7%); and 10-30 kg of weight handled (35%).
    Conclusions: A large number of workers exposed to biomechanical and organizational risks report musculoskeletal pain. Employers should check working conditions and adjust them, investing in health promotion and protection actions to effectively reduce the occurrence of these disorders.
