    Abstract 12/15-lipoxygenase (12/15-LOX) plays an essential role in oxidative conversion of polyunsaturated fatty acids into various bioactive lipid molecules. Although 12/15-LOX's role in the pathophysiology of various human diseases has been well studied, its role in weight gain is controversial and poorly clarified. Here, we demonstrated the role of 12/15-LOX in high-fat diet (HFD)-induced weight gain in a mouse model.     Abstract A diastereoselective N-heterocyclic carbene-catalysed reaction between enal and 3-cyano-2-imino-2H-chromene is described for accessing a new type of C<sub>4</sub>-functionalized 2-amino-3-cyano-4H-chromene. A wide variety of enals and iminochromenes afforded the desired homoenolate addition product 2-amino-4H-chromenes in good yields and with moderate to good diastereoselectivities.

    Abstract We obtain several lower bounds on the $\textsf{Max-Cut}$ of $d$-degenerate $H$-free graphs. Let $f(m,d,H)$ denote the smallest $\textsf{Max-Cut}$ of an $H$-free $d$-degenerate graph on $m$ edges. We show that $f(m,d,K_r)\ge \left(\frac{1}{2} + d^{-1+\Omega(r^{-1})}\right)m$, generalizing a recent work of Carlson, Kolla, and Trevisan. We also give bounds on $f(m,d,H)$ when $H$ is a cycle, odd wheel, or a complete bipartite graph with at most 4 vertices on one side. We also show stronger bounds on $f(m,d,K_r)$ assuming a conjecture of Alon, Bollabas, Krivelevich, and Sudakov (2003). We conjecture that $f(m,d,K_r)= \left( \frac{1}{2} + \Theta_r(d^{-1/2}) \right)m$ for every $r\ge 3$, and show that this conjecture implies the ABKS conjecture.

    Abstract In this paper, we deal with the power regulation of tethered-wing systems and demonstrate advantages of variable-pitch control in mitigating the dynamic mechanical loads and power fluctuations. The proposed scheme is based on a strategy that maximizes the energy capture during low-speed wind and prevents overloads during the high-speed wind. To realize this strategy, we use a tether reeling-speed controller to track the optimal generator speed during low-speed wind and a MIMO speed-force controller for power limitation during high-speed wind. The controllers are synthesized using H-infinity method and are based on a linear parameter varying (LPV) system that expresses the flexible dynamics of system as a function of the tether's length and force. Using this method, the controllers are made robust with respect to dynamic and parametric uncertainties and the wing's pitch angle activity is minimized during high-speed wind. We carry out extensive simulations to demonstrate the controllers' performance. These include the implementation of the scheme in a detailed 3-dimensional tethered-wing system simulator with a realistic turbulent wind field.

    Abstract Hydroperoxyalkylperoxy (OOQOOH) radical isomerization is an important low-temperature chain branching reaction within the mechanism of hydrocarbon oxidation. This isomerization may proceed via the migration of the α-hydrogen to the hydroperoxide group. In this work, a combination of high level composite methods-CBS-QB3, G3, and G4-is used to determine the high-pressure-limit rate parameters for the title reaction. Rate rules for H-migration reactions proceeding through 5-, 6-, 7-, and 8-membered ring transitions states are determined. Migrations from primary, secondary and tertiary carbon sites to the peroxy group are considered. Chirality is also investigated by considering two diastereomers for reactants and transition states with two chiral centers. This is important since chirality may influence the energy barrier of the reaction as well as the rotational energy barriers of hindered rotors in chemical species and transition states. The effect of chirality and hydrogen bonding interactions in the investigated energies and rate constants is studied. The results show that while the energy difference between two diastereomers ranges from 0.1-3.2 kcal/mol, chirality hardly affects the kinetics, except at low temperatures (atmospheric conditions) or when two chiral centers are present in the reactant. Regarding the effect of the H-migration ring size, it is found that in most cases, the 1,5 and 1,6 H-migration reactions have similar rates at low temperatures (below ∼830 K) since the 1,6 H-migration proceeds via a cyclohexane-like transition state similar to that of the 1,5 H-migration.

    Abstract We recently developed a screening system capable of identifying and evaluating inhibitors of the Hepatitis B virus (HBV) ribonuclease H (RNaseH), which is the only HBV enzyme not targeted by current anti-HBV therapies. Inhibiting the HBV RNaseH blocks synthesis of the positive-polarity DNA strand, causing early termination of negative-polarity DNA synthesis and accumulation of RNA:DNA heteroduplexes. We previously reported inhibition of HBV replication by N-hydroxyisoquinolinediones (HID) and N-hydroxypyridinediones (HPD) in human hepatoma cells. Here, we report results from our ongoing efforts to develop more potent anti-HBV RNaseH inhibitors in the HID/HPD compound classes. We synthesized and screened additional HIDs and HPDs for preferential suppression of positive-polarity DNA in cells replicating HBV.
