    Abstract Ebola virus (EBOV) causes severe hemorrhagic fever in humans and is lethal in a large percentage of those infected. The EBOV matrix protein viral protein 40 kDa (VP40) is a peripheral binding protein that forms a shell beneath the lipid bilayer in virions and virus-like particles (VLPs). VP40 is required for virus assembly and budding from the host cell plasma membrane. VP40 is a dimer that can rearrange into oligomers at the plasma membrane interface, but it is unclear how these structures form and how they are stabilized. We therefore investigated the ability of VP40 to form stable oligomers using in vitro and cellular assays. We characterized two lysine-rich regions in the VP40 C-terminal domain (CTD) that bind phosphatidylinositol-4,5-bisphosphate (PI(4,5)P

    Abstract An important paradigm shift towards the circular economy is to prioritize waste prevention, reuse, recycling, and recovery before disposal is necessary. In this context, a sustainable protocol of converting waste pea peel (wPP) into low-cost carbon nanomaterials for sensing and conversion of p-nitrophenol (p-NP) into value-added paracetamol is being reported. Two fractions of the carbonaceous nanomaterials were obtained after the hydrothermal treatment (HT) of wPP, firstly an aqueous portion containing water-soluble carbon dots (wPP-CDs) and a solid residue, which was converted into carbonized biochar (wPP-BC). Blue-colored fluorescent wPP-CDs displayed excitation-dependent and pH-independent properties with a quantum yield (QY) of 8.82 %, which were exploited for the fluorescence sensing of p-NP with 4.20 μM limit of detection. Pyrolyzed biochar acting as an efficient catalyst effectively reduces p-NP to p-aminophenol (p-AP) in just 16 min with a 0.237 min

    Abstract Background: Most Parkinson's disease (PD) loci have shown low prevalence in the Indian population, highlighting the need for further research.
    Objective: The aim of this study was to characterize a novel phosphatase tensin homolog-induced serine/threonine kinase 1 (PINK1) mutation causing PD in an Indian family.
    Methods: Exome sequencing of a well-characterized Indian family with PD. A novel PINK1 mutation was studied by in silico modeling using AlphaFold2, expression of mutant PINK1 in human cells depleted of functional endogenous PINK1, followed by quantitative image analysis and biochemical assessment.
    Results: We identified a homozygous chr1:20648535-20648535 T>C on GRCh38 (p.F385S) mutation in exon 6 of PINK1, which was absent in 1029 genomes from India and in other known databases. PINK1 F385S lies within the highly conserved Deutsche Forschungsgemeinschaft (DFG) motif, destabilizes its active state, and impairs phosphorylation of ubiquitin at serine 65 and proper engagement of parkin upon mitochondrial depolarization.
    Conclusions: We characterized a novel nonconservative mutation in the DFG motif of PINK1, which causes loss of its ubiquitin kinase activity and inhibition of mitophagy. © 2024 The Authors.     Abstract We present measurements of the cross section and double-helicity asymmetry A_{LL} of direct-photon production in p[over →]+p[over →] collisions at sqrt[s]=510  GeV. The measurements have been performed at midrapidity (|η|<0.25) with the PHENIX detector at the Relativistic Heavy Ion Collider. At relativistic energies, direct photons are dominantly produced from the initial quark-gluon hard scattering and do not interact via the strong force at leading order. Therefore, at sqrt[s]=510  GeV, where leading-order-effects dominate, these measurements provide clean and direct access to the gluon helicity in the polarized proton in the gluon-momentum-fraction range 0.02<x<0.08, with direct sensitivity to the sign of the gluon contribution.<br />
