    Abstract Ulcerative colitis (UC) is a subtype of inflammatory bowel disease (IBD) and occurs mainly in the colon. The etiology of UC is rather complex and involves various pathological factors, including genetic susceptibility, dietary intakes, environment, and microbiota. In China, the Chang-Yan-Ning (CYN) formula has been utilized in the clinic to treat gastrointestinal disorders, but its pharmacological evidence remains elusive. The investigation was designed to explore the molecular and cellular mechanisms of CYN. Liquid Chromatography with tandem mass spectrometry (LC/MS) was performed to identify the key components in the formula; Network pharmacology analysis was executed to predict the potential targets of CYN; An experimental murine colitis model was established by utilizing 2% dextran sodium sulfate (DSS), and CYN was administered for 14 days.     Abstract Objectives: To investigate the technical performance of IDentifier DNA typing kit (YanHuang34) and evaluate its forensic application value.

    Results: IDentifier DNA typing kit (YanHuang34) had good species specificity, veracity, adaptability, inhibitor tolerance and balance. The sensibility was up to 0.062 5 ng. It was able to detect different types of samples, degraded samples and inhibitor mixed samples. Complete DNA typing could be obtained for samples with the mixture ratio less than 4∶1. The system efficiency of IDentifier DNA typing kit (YanHuang34) was very high, with TDP up to 1-1.08×10
    Conclusions: The IDentifier DNA typing kit (YanHuang34) is adaptive to the GB/T 37226-2018 requirements. It can be used for individual identification and paternity identification, and is suitable for application in the field of forensic science.

    Abstract Changes in hydrothermal conditions have significant effects on vegetation, but there is still a lack of understanding of how vegetation responds to land surface (surface temperature and soil moisture) and meteorological (temperature and precipitation) conditions in mountain regions. This study examined the trends of leaf area index (LAI) in the Yan Mountains over the last four decades using Global Land Surface Satellite (GLASS) data. The results showed a persistent increase of LAI (greening) over 20 % to 80 % of the study area in growing season, spring, summer and autumn. Anthropogenic activities caused the greening trend by crop management before 2000 and afforestation after 2000. The increasing rate of LAI varied with elevation, and the most significant increase occurred in areas between 300 and 900 m, and the lowest increase occurred in areas below 300 m. Moreover, we found that LAI was negatively correlated with land surface temperature and soil moisture, but positively correlated with precipitation and air temperature. The time-lag effect was found between hydrothermal factors and LAI in the past four decades. There was a time lag of 2–3 months between LAI changes and temperature/precipitation during the early and late stages of the growing season, and a time lag of 0–1 month during the middle stage. Specifically, there was no time lag in vegetation response to surface soil moisture, and a time lag of 2–3 months in vegetation response to land surface temperature from July to October. Our findings provide insights into how vegetation adapts to land surface and climatic hydrothermal conditions in mountain regions and can be used by governments to develop policies for ecological protection.

    Abstract Stevens-Johnson syndrome/toxic epidermal necrolysis (SJS/TEN) is a rare adverse cutaneous reaction with a low incidence and high mortality. Despite posing a serious threat to patients' health and lives, there is no high-quality evidence for a standard treatment regimen. Here we report the case of a 62-year-old man with stage IV pancreatic cancer who experienced immunotherapy-induced SJS/TEN. After consensus-based regular treatments at a local hospital, his symptoms became worse. Thus, he consented to receive Chinese herbal medicine (CHM) therapy. The affected parts of the patient were treated with the CHM Pi-Yan-Ning which was applied externally for 20 min twice a day. After 7 days of treatment, the dead skin began peeling away from the former lesions that had covered his hands, feet, and lips, indicating that skin had regenerated. After 12 days of treatment, the patient's skin was completely recovered. In this case, SJS/TEN was successfully treated with Pi-Yan-Ning, suggesting that there might be tremendous potential for the use of Pi-Yan-Ning in the treatment of severe skin reactions to drug treatments. Further basic investigations and clinical trials to explore the mechanism and efficacy are needed.

    Abstract Obesity and related metabolic disorders are associated with intestinal microbiota dysbiosis, disrupted intestinal barrier and chronic inflammation. Shen-Yan-Fang-Shuai formula (SYFSF) is a traditional Chinese herbal formula composed of Astragali Radix, Radix Angelicae Sinensis, Rheum Officinale Baill, and four other herbs. In this study, we identified that SYFSF treatment prevented weight gain, low-grade inflammation and insulin resistance in high-fat diet (HFD)-fed mice. SYFSF also substantially improved gut barrier function, reduced metabolic endotoxemia, as well as systemic inflammation. Sequencing of 16S rRNA genes obtained from fecal samples demonstrated that SYFSF attenuated HFD-induced gut dysbiosis, seen an decreased Firmicutes to Bacteroidetes ratios. Microbial richness and diversity were also higher in the SYFSF-treated HFD group. Furthermore, similar therapeutic effects and changes in gut microbiota profile caused by SYFSF could be replicated by fecal microbiota transfer (FMT). Taken together, our study highlights the efficacy of SYFSF in preventing obesity and related metabolic disorders. Its therapeutic effect is associated with the modulation of gut microbiota, as a prebiotic.

    Abstract Context: Qi-Wei-Xiao-Yan-Tang (XYT), composed of Radix et Rhizoma Rhei, Radix Astragali, Radix Wikstroemiae Indicae, Fructus Ligustri Lucidi, Poria and Radix Glycyrrhizae, has been widely used as an anti-inflammatory drug.
    Objective: The present study investigated the anti-inflammatory, antibacterial effects and serum pharmacochemistry of XYT.
    Methods: The dimethylbenzene-induced inflammation test, the acetic acid-induced vascular permeability test and the carrageenan-induced paw edema test were used to evaluate the anti-inflammatory activity of XYT (200, 100 and 50 mg/kg); minimal inhibitory concentration (MIC). Minimal bactericidal concentration (MBC) tests were used to evaluate the antibacterial activity of XYT. Additionally, serum pharmacochemistry was performed to study the biologically active substances.
    Results: All the tests for anti-inflammatory effects were shown active with these test systems; the anti-inflammatory effects at doses of 100 and 200 mg/kg were significant (p < 0.05); MIC and MBC tests indicated that XYT showed a broader antimicrobial spectrum and stronger toxicity to the tested microbes. Additionally, calycosin-7-glucoside, sennoside A, aloeemodin and rhein were detected as the predominant components in rat serum which may play the key role in the anti-inflammatory activities of XYT.
    Conclusions: This is the first report of the pharmacological activities and serum pharmacochemistry of XYT, and the first evidence of anti-inflammatory and antimicrobial properties of the extracts of XYT. The results of our work demonstrated that XYT has significant anti-inflammatory and antibacterial properties, and calycosin-7-glucoside, sennoside A aloeemodin and rhein may be the biologically active substances of XYT. XYT can be utilized as an effective and safe disease preventive or therapeutic agent.

    Abstract The organic-rich shales of the Chang 7 Member in the Yan’an Formation of the Yan’an area, Ordos Basin is a hot spot for lacustrine shale gas exploration. In this paper, taking the Chang 7 Member shale in the Yan’an area as an example, the main controlling factors of lacustrine shale gas accumulation and the prediction of “sweet spots” are systematically carried out. The results show that the Yanchang Formation shale has the complete gas generating conditions. Shale gas accumulation requires three necessary accumulation elements, namely gas source, reservoir and good preservation conditions. The dynamic hydrocarbon generation process of the Chang 7 shale reservoirs is established according to the thermal simulation experiments of hydrocarbon generation, and the mechanism of catalytic degradation and gas generation in the Chang 7 Member under the background of low thermal evolution degree is revealed. The enriched authigenic pyrite can catalyze the hydrocarbon generation of organic matter with low activation energy, thereby increasing the hydrocarbon generation rates in the low-mature-mature stage. Different types of pores at different scales (2–100 nm) form a multi-scale complex pore network. Free gas and dissolved gas are enriched in laminar micro-scale pores, and adsorbed gas is enriched in nano-scale pores of thick shales, and silty laminates can improve the physical properties of the reservoir. This is because the laminar structure has better hydrocarbon generation conditions and is favorable for the migration of oil and gas molecules. The thickness of the lacustrine shale in the Chang 7 Member is between 40 and 120 m, which has exceeded the effective hydrocarbon expulsion thickness limit (8–12 m). At the end of the Early Cretaceous, the excess pressure of the Chang 7 shale was above 3 MPa. At present, horizontal wells with a daily gas production of more than 50,000 cubic meters are distributed in areas with high excess pressures during the maximum burial depth.

    Abstract Hydrologic connectivity is related to the water-mediated transport of matter, energy, and organisms within or between elements of the hydrologic cycle. It reflects the hydrological consequences caused by topographic, land cover, and climatic factors, and is an important tool to characterize and predict the hydrological responses to climate and landscape change. In the Loess Plateau region, a large number of reservoirs have been constructed to trap sediment and storage water for drinking, irrigation, and industries. The land cover has been significantly reshaped in the past decades. These changes may alter the watershed hydrological connectivity. In this study, we mapped the spatial pattern of hydrological connectivity with consideration of reservoir impedances, mitigation of climate, and land cover in the Yan River watershed on the Loess Plateau by using the network index (NI) approach that is based on topographical wetness index. Three wetness indices were used, i.e., topographical wetness index (TWI), SAGA (System for Automated Geoscientific Analyses) wetness index (WIS), and wetness index adopted aridity index (AI) determined by precipitation and evapotranspiration (WIPE). In addition, the effective catchment area (ECA) was also employed to reveal the connectivity of reservoirs and river networks to water source areas. Results show that ECA of reservoirs and rivers account for 35% and 65%, respectively; the hydrological connectivity to the reservoir was lower than that to the river networks. The normalized hydrological connectivity revealed that the connectivity to river channels maintained the same distribution pattern but with a decreased range after construction of reservoirs. As revealed by comparing the spatial patterns of hydrological connectivity quantified by NI based on WIS and WIPE respectively, vegetation cover patterns had significantly alternated watershed hydrological connectivity. These results imply a decreased volume of flow in river channels after reservoir construction, but with same temporal period of flow dynamic. It is illustrated that the network index (NI) is suitable to quantify the hydrological connectivity and it is dynamic in the context of human intervention and climate change.

    Abstract Distributions of transverse momentum [Formula: see text] and the related angular variable [Formula: see text] of Drell–Yan lepton pairs are measured in 20.3 fb[Formula: see text] of proton–proton collisions at [Formula: see text] TeV with the ATLAS detector at the LHC. Measurements in electron-pair and muon-pair final states are corrected for detector effects and combined. Compared to previous measurements in proton–proton collisions at [Formula: see text] TeV, these new measurements benefit from a larger data sample and improved control of systematic uncertainties. Measurements are performed in bins of lepton-pair mass above, around and below the

    Abstract Li-Fei-Xiao-Yan prescription (LFXY) has been clinically used in China to treat inflammatory and infectious diseases including inflammatory lung diseases. The present study was aimed at evaluating the potential therapeutic effects and potential mechanisms of LFXY in a murine model of lipopolysaccharide- (LPS-) induced acute lung injury (ALI). In this study, the mice were orally pretreated with LFXY or dexamethasone (positive drug) before the intratracheal instillation of LPS. Our data indicated that pretreatment with LFXY enhanced the survival rate of ALI mice, reversed pulmonary edema and permeability, improved LPS-induced lung histopathology impairment, suppressed the excessive inflammatory responses
