    Abstract Aim of the study: To evaluate the therapeutic effect of SYNC in diarrhea irritable bowel syndrome (IBS-D) and explore its underlying mechanism through transcriptomic sequencing (RNA-Seq).
    Materials and methods: A rat model of IBS-D was constructed to elucidate the effects of SYNC. Abdominal withdrawal reflex (AWR), fecal water content (FWC), and recording body weight were calculated to assess visceral sensitivity in rats. Histopathological changes in the colon and alterations in mast cell (MC) count were determined. Immunohistochemistry was employed to assess mast cell tryptase (MCT) expression in rat colons. Serum levels of corticotropin-releasing Hormone (CRH), interleukin-6 (IL-6), calcitonin gene-related peptide (CGRP), and 5-hydroxytryptamine (5-HT) were quantified using ELISA. RNA-Seq of colon tissue was performed, followed by Gene Ontology (GO), Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analyses. Western blot analysis was conducted to quantify the expression levels of key proteins in the Nr4a3 pathway in the colon and hypothalamus tissues of rats.
    Results: SYNC alleviated visceral hypersensitivity and mood disorders in rats with IBS-D. Moreover, it was positively correlated with its dosage and the observed effects, such as the enhancement of the colon's mucosal lining condition and reduction in the number and activation of MCs within the model group. SYNC reduced the expression levels of factors related to the brain-gut axis and inflammatory markers in the bloodstream. RNA-Seq analysis indicated that SYNC down-regulated the expression of Nr4a3 and PI3K. These SYNC-targeted genes primarily played roles in immune regulation and inflammatory responses, correlating with the modulation of Nr4a3 and the PI3K/AKT pathway. Western blot analysis further confirmed SYNC's influence on inflammation-related MC activation by downregulating key proteins in the Nr4a3/PI3K pathway.
    Conclusions: SYNC inhibited mast cell activation and attenuated visceral hypersensitivity in the colon tissues of IBS-D rats. These effects were mediated by the Nr4a3/PI3K signaling pathway.

    Abstract The Chang’E 5 (CE-5) samples represent the youngest mare basalt ever known and provide an access into the late lunar evolution. Recent studies have revealed that CE-5 basalts are the most evolved lunar basalts, yet controversy remains over the nature of their mantle sources. Here we combine Fe and Mg isotope analyses with a comprehensive study of petrology and mineralogy on two CE-5 basalt clasts. These two clasts have a very low Mg# (∼29) and show similar Mg isotope compositions to Apollo low-Ti mare basalts as well as intermediate TiO _2 and Fe isotope compositions between low-Ti and high-Ti mare basalts. Fractional crystallization or evaporation during impact cannot produce such geochemical signatures that otherwise indicate a hybrid mantle source that incorporates both early- and late-stage lunar magma ocean (LMO) cumulates. Such a hybrid mantle source would be also compatible with the KREEP-like Rare Earth Elements pattern of CE-5 basalts. Overall, our new Fe–Mg isotope data highlight the role of late LMO cumulate for the generation of young lunar volcanism.

    Abstract Colitis-associated colorectal cancer (CAC) is a specific type of colorectal cancer (CRC) with high mortality and morbidity, the chronic inflammation in the intestinal mucosal is the characteristic of CAC. Chang Qing formula (CQF) is a Chinese herbal formula used clinically for the treatment of CAC with remarkable clinical efficacy, but its mechanism remains unclear. In the present work, Combined network pharmacology and transcriptomics were used to analyze the potential active ingredients and elucidate molecular mechanism of CQF in treating CAC. Firstly, the constituents migrating to blood of CQF were analyzed and identified by UPLC-Q-TOF-MS/MS, and core genes and pathways were screened by network pharmacology analysis. Encyclopedia of Genes and Genomes (KEGG) analysis showed that the IL-17 signaling pathway involved in CAC may be closely associated with the potential mechanismof action of CQF. Subsequently, the results from animal studies indicated that CQF profoundly reduced tumor numbers and tumor size in AOM/DSS mice. The RNA-seq data was analysed utilizing Ingenuity Pathway Analysis (IPA), and the results supported the idea that CQF exerts a tumour-suppressive effect via the IL-17 signalling pathway. Further studies demonstrated that CQF significantly reduced IL-17A levels, which in turn inhibited NF-κB/IL-6/STAT3 signaling cascade, suppressed MMP9 expression and promoted tumor cell apoptosis. In conclusion, the current study demonstrated that CQF remarkably improved inflammatory tumor microenvironment, and hindered the transformation of inflammation into cancer. These findings may help to design future strategies for the treatment of CAC.

    Abstract To foster an ecological civilization and ensure sustainable development of population resources, a critical focus of China's land spatial planning initiatives is the complex interplay among the prudent utilization of regional natural resources, stable socio-economic growth, and ecological environment preservation and governance. This interplay is vital for improving the population's quality of life, enhancing national resilience, and fostering the development of an ecological civilization. Within this context, this research focuses on the Chang-Ji-Tu region, employing a hybrid InVEST-Bayesian Belief Network-PLUS model. This approach facilitates a comprehensive assessment of habitat quality (HQ), carbon sequestration (CS), soil conservation (SC), crop production (CP), and total ecosystem services (TES) spanning 2005 to 2020, to optimize spatial structures based on these assessments. The findings indicate significant insights: (1) temporally, both HQ and CS exhibit an initial ascent followed by a subsequent decline, while SC demonstrates a rise, subsequent decrease, and then a gradual increase. CP shows a consistent increase, and TES initially decreases before experiencing a rise. Spatially, regions exhibiting high CP are primarily located in the northwest, in contrast to the southeast, demonstrating lower values. Conversely, HQ, CS, and SC exhibit lower values in the northwest and higher values in the southeast. TES exhibits considerable variability in the northwest and a more equilibrated distribution in the southeast. (2) A positive correlation is observed between land use/cover changes (LUCC) and HQ, CS, and SC, while a negative correlation is noted with CP and TES. (3) In exploring potential scenarios for economic development (ED), natural development (ND), and ecological protection (EP) in 2035, our study categorizes TES into distinct zones: ecological protection prohibited zones, ecological buffer prohibited zones, construction and development suitable zones, and restricted zones for arable land protection. This study, grounded in the current ecosystem status, thoroughly analyzes spatial structural optimization, offering crucial insights for future land spatial planning and ecological restoration.

    Abstract Background: Sect. Tuberculata belongs to Camellia, and its members are characterized by a wrinkled pericarp and united filaments. All the plants in this group, which are endemic to China, are highly valuable for exploring the evolution of Camellia and have great potential for use as an oil source. However, due to the complex and diverse phenotypes of these species and the difficulty of investigating them in the field, their complex evolutionary history and interspecific definitions have remained largely unelucidated.
    Results: Therefore, we newly sequenced and annotated 12 chloroplast (cp) genomes and retrieved the published cp genome of Camellia anlungensis Chang in sect. Tuberculata. In this study, comparative analysis of the cp genomes of the thirteen sect. Tuberculata species revealed a typical quadripartite structure characterized by a total sequence length ranging from 156,587 bp to 157,068 bp. The cp.genome arrangement is highly conserved and moderately differentiated. A total of 130 to 136 genes specific to the three types were identified by annotation, including protein-coding genes (coding sequences (CDSs)) (87-91), tRNA genes (35-37), and rRNA genes (8). The total observed frequency ranged from 23,045 (C. lipingensis) to 26,557 (C. anlungensis). IR region boundaries were analyzed to show that the ycf1 gene of C. anlungensis is located in the IRb region, while the remaining species are present only in the IRa region. Sequence variation in the SSC region is greater than that in the IR region, and most protein-coding genes have high codon preferences. Comparative analyses revealed six hotspot regions (tRNA-Thr(GGT)-psbD, psbE-petL, ycf15-tRNA-Leu(CAA), ndhF-rpl32, ndhD, and trnL(CAA)-ycf15) in the cp genomes that could serve as potential molecular markers. In addition, the results of phylogenetic tree construction based on the cp genomes showed that the thirteen sect. Tuberculata species formed a monophyletic group and were divided into two evolutionarily independent clades, confirming the independence of the section.
    Conclusions: In summary, we obtained the cp genomes of thirteen sect. Tuberculata plants and performed the first comparative analysis of this group. These results will help us better characterize the plants in this section, deepen our understanding of their genetic characteristics and phylogenetic relationships, and lay the theoretical foundation for their accurate classification, elucidation of their evolutionary changes, and rational development and utilization of this section in the future.

    Abstract In the present study, purine alkaloid analysis and transcriptome of Camellia gymnogyna Hung T. Chang (Theaceae) from Dayao Mountain were performed by high-performance liquid chromatography (HPLC) and RNA-Seq, respectively. The results showed that the major purine alkaloids accumulated in Camellia gymnogyna Hung T. Chang (Theaceae) were theobromine together with a small amount of theacrine and caffeine. Through polymerase chain reaction (PCR), three types of cDNA encoding N-methyltransferases were isolated from the leaves of Camellia gymnogyna Hung T. Chang (Theaceae) and designated GCS1, GCS2, and GCS3. We subsequently expressed GCS1, GCS2, and GCS3 in Escherichia coli and incubated lysates of the bacterial cells with a variety of xanthine substrates in the presence of S-adenosyl-L-methionine as the methyl donor. We found that the recombinant GCS1 proteins catalyzed 1,3,7-trimethyluric acid to produce theacrine, the recombinant GCS3 proteins catalyzed 7-methylxanthine to produce theobromine, while the recombinant GCS2 proteins did not catalyze any xanthine derivatives. Simultaneous analysis of the expressions of GCS1, GCS2, GCS3, and a caffeine synthase gene (TCS1) in Camellia gymnogyna Hung T. Chang (Theaceae) and other tea plants provided a reference for further research on the functions of these genes.

    Abstract Urbanization is a major source of carbon emissions. A quantitative study on the dynamic relationship between urbanization and its morphological characteristics and carbon emissions is crucial for formulating urban carbon emission reduction policies. Based on the carbon metabolism model, the carbon emissions at the country level in Chang-Zhu-Tan from 1995 to 2020 were calculated. The Tapio decoupling model was used to explore the decoupling relationship between the carbon emissions of Chang-Zhu-Tan and urban land, and a geographically and temporally weighted regression(GTWR) model was used to analyze the impact mechanism of urban spatial morphology on carbon emissions. The following conclusions were drawn:① carbon emissions at the county level in the study area formed a clustered distribution centered on the city jurisdiction and showed a trend of diffusion from year to year. Compared with those in 1995, there were seven new high carbon emission districts in 2020, all of which belonged to Changsha. ② From 1995-2020, the research area as a whole changed from mainly strong decoupling to mainly dilated negative decoupling, and the spatial decoupling state fluctuated back and forth between the decoupling and negative decoupling. By 2020, except for the seven regions with the uncoupling state regressing, all of them reached the uncoupling state or were close to the uncoupling state. ③ Urban patch area(CA), urban patch number(NP), and patch combination degree(COHESION) were positively correlated with urban carbon emissions, whereas landscape shape index(LSI), maximum patch index(LPI), and Euclidean distance mean(ENN_MN) were negatively correlated with urban carbon emissions, and the impact of different urban form indicators on carbon emissions had significant spatial heterogeneity.
