    Abstract Objective: To evaluate methanolic, ethanolic, acetone and aqueous extracts from different parts of Eurycoma longifolia (E. longifolia) (leave, stem, and root) for antibacterial activity against Gram-positive and Gram-negative bacteria and to utilize the leaves and stem parts rather than the root, which is already used for male sexual enhancement in Malaysia.
    Methods: The study took place in the Laboratory of Molecular Biology of Biotechnology Engineering Department, Malaysia between January 2005 and June 2006. Methanolic, ethanolic, acetone and aqueous extracts of leaves, stems and roots of E. longifolia were investigated for their antibacterial properties using Agar-well diffusion method.
    Results: The alcoholic and acetone extracts of the leaves and stem extracts were active on both Gram-positive and Gram-negative bacteria except against 2 strains of Gram-negative bacteria (Escherichia coli and Salmonella typhi). The root extracts had no antibacterial activity against Gram-positive and Gram-negative bacteria tested. Aqueous leaves extract showed antibacterial activity against Staphylococcus aureus and Serratia marscesens.
    Conclusion: The alcoholic and acetone extracts from leaves and stems of E. longifolia contain potent antibacterial agent(s). This plant can serve as a potential source of antibacterial compounds.

    Abstract The production of phytase in a recombinant E.coliusing thepBAD expression system wasoptimized using response surface methodology with full-factorial faced centered central composite design. The ampicilin and arabinose concentrationin the cultivation media and the incubation temperature were optimized in order to maximize phytase productionusing 23 central composite experimental design. With this design the number of actual experiment performed could be reduced while allowing eludidation of possible interactions among thesefactors. The most significant parameter was shown to be the linear and quadratic effect of the incubation temperature. OPtim al conditionsfor phytase production were determinedto be100 μg/ml ampicilin, 0.2 % arabinose and an incubation temperature of37ºC.The production of phytase in the recombinant E. coliwas scaledup to 100ml and 1000ml.

    Abstract Using a combination of High-Performance Ion Chromatography analysis and kinetic studies, the pathway of myo-inositol hexakisphosphate dephosphorylation by a phytase from a Malaysian waste-water bacterium was established. The data demonstrate that the phytase preferably dephosphorylates myo-inositol hexakisphosphate in a stereospecific way by sequential removal of phosphate groups via D-I(1,2,3,4,5)P(5), D-I(2,3,4,5)P(4), D-I(2,3,4)P(3), D-I(2,3)P(2) to finally I(2)P. It was estimated that more than 90% of phytate hydrolysis occurs via D-I(1,2,3,4,5)P(5). Thus, the phytase from the Malaysian waste-water bacterium has to be considered a 6-phytase (E.C. 3.1.3.26). A second pathway of minor importance could be proposed which is in accordance with the results obtained from analysis of the dephosphorylation products formed by the action of the phytase under investigation on myo-inositol hexakisphosphate. It proceeds via D/L-I(1,2,4,5,6)P(5), D/L-I(1,2,4,5)P(4), D/L-I(1,2,4)P(3), D/L-I(2,4)P(2) to finally I(2)P.

    Abstract Homogenizer and ultrasonicator that we call as mechanical methods were used in the lab scale production of alcohol free reduced glutathione (alcohol free GSH) from baker’s yeast. The study was done to determine the effects of specific parameters of the methods and the pH of the medium on the production of the GSH. The cell disruptions were run at a wide range of those parameters at the suitable common parameters of the methods. The disrupted cells suspension was then centrifuged at 12,000 rpm for 20 min and the GSH content in the supernatant was analyzed by taking the absorbance (A) value of the solution by spectrophotometer at 412 nm. The results obtained showed that the production of GSH was affected by the adjusted specific parameters of each method and the yield of production by the homogenizing and ultrasonic method was found 3.3 and 4.1 times, respectively, better than the yield of production by the unadjusted parameters. Ultrasonic method which produced 23.2 mg/L product was found 1.3 times better than the homogenizing method (18.5 mg/L) in producing the GSH. The result obtained from the lab scale study of the above mentioned mechanical methods showed that the implementation of them in the large scale production of the alcohol free GSH is possible.

    Abstract The purpose of this study was to determine the effect of operating parameters such as the yeast concentration, processing temperature and isolation time on the isolation of GSH using Homogenizer Method. The disrupted cells solution was centrifuged at 12,000 rpm for 20 min and the GSH content in the supernatant was analyzed by taking the optical density (OD) value of the solution using Spectrophotometer at 412 nm. Yeast solution with the concentration of 3, 6, 9 and 12 wt.% was disrupted at their original temperature for 30 sec to study the effect of yeast concentration. The result obtained shows that the optimum value of yeast concentration is 9 wt.% where 10.98 μmol mL -1 GSH was isolated. To study the effect of processing temperature, isolation process of the 9 wt.% yeast solution was done at various values of temperature such as 19, 22, 25, 28, 30 and 40�C for 30 sec. The results indicated that, the optimum GSH isolation temperature is 22�C with the isolation of 17.93 μmol mL -1 GSH. Then, to study the effect of isolation time, the isolation process of sample with the concentration of 9 wt.% was done at 22�C for 15, 30, 45 and 60 sec. The result indicated that the optimum isolation time for the Ultrasonic process is 30 sec.
