    Abstract Objective: To observe the long-term effects of total hysterectomy on urinary function, evaluate the effects of preoperative nutritional status, urinary occult infection, and surgical factors on the induction of postoperative stress urinary incontinence (SUI), and explore the incidence and risk factors of SUI.
    Study design: From January 2017 to December 2017, 164 patients with benign non-prolapsing diseases who underwent a laparoscopic total hysterectomy in the First People's Hospital of Taicang were selected as the analysis objects. The International Incontinence Standard Questionnaire for Female Lower Urinary Tract Symptoms (ICIQ-FLUTS) and Pelvic Floor Impact Questionnaire-short version 20 (PFDI-20) were used for telephone follow-up to subjectively assess the urinary function of patients, collect their medical records, and statistically analyze the number of postoperative SUI cases. Logistic multivariate analysis was used to analyze the influencing factors of postoperative female SUI, presented as adjusted odds ratios with 95% confidence intervals.
    Results: Only 97 out of 164 patients completed the ICIQ-FLUTS and PFDI-20 questionnaires. Among these participants, 28 patients (28.86%) were diagnosed with SUI (study group), while 69 patients (71.13%) were classified as women without SUI (control group). The age, menopause, parity ≥ 2 times, Body mass index (BMI) ≥ 28 kg/m
    Conclusions: Hysterectomy for benign non-prolapse diseases has a long-term potential impact on the urinary system of patients, and the risk of postoperative SUI increases. The main risk factors of SUI are parity, menopausal status, obesity, preoperative nutritional status, and occult infection of the urinary system.

    Abstract Analysis of cell types is recognized as a major task in current single-cell genotyping and phenotyping. In neuroscience, 3-D neuron morphologies are often reconstructed from multi-dimensional microscopic images. Recent studies indicate that neurons could form very complicated distributions in the feature space, and thus they can be explored using manifold analysis. We have developed manifold classification toolkit software to replace the conventional clustering analysis to discover cell subtypes from three state-of-the-art collections of single neurons' 3-D morphologies that reconstructed from images. We have gathered 9208 3-D spatially registered whole mouse brain neurons from three datasets with the highest quality to date generated by the single neuron morphology community. To explore manifold distribution, our method uses minimum spanning tree-based principal skeletons to approximate locally linear embeddings, to explore the morphological feature spaces, which correspond to dendritic arbors, axonal arbors or both categories of arborization patterns of neurons. We show manifold classification is a suitable approach for a majority of often referred cell types, each of which was also discovered to contain multiple subtypes. Our results show an initial effort to employ manifold classification but not traditional clustering analysis as an alternative framework for analyzing 3-D neuron morphologies reconstructed from 3-D microscopic images.

    Abstract Background: Cervical cancer (CC) is highly lethal and aggressive with an increasing trend of mortality for females. Molecular characterization-based methods hold great promise for improving the diagnostic accuracy and for predicting treatment response.
    Methods: The mRNAs expression data of CC patients and cellular senescence-related genes were obtained from the Cancer Genome Atlas (TCGA) and CellAge databases, respectively. Differentially expressed genes (DEGs) of senescence related genes between tumor and normal tissues were used for Least absolute shrinkage and selection operator (LASSO) regression to construct a prognostic model. Univariate and LASSO regression analyses were applied to establish a predictive nomogram. The performance of the nomogram were evaluated by Kaplan-Meier curve, receiver operating characteristic (ROC), Harrell's concordance index (C-index), and calibration curve. GSE44001 and GSE52903 were used for external validation.
    Results: We established a cellular senescence-related genes-based stratified model, and a multivariable-based nomogram, which could accurately predict the prognosis of CC patients in the TCGA database. The Kaplan-Meier curve indicated that patients in the low-risk group had considerably better overall survival (OS,
    Conclusions: Our results suggested a six-senescence related signature and established a prognostic nomogram that reliably predicted the overall survival for CC. These findings may be beneficial to personalized treatment and medical decision-making.
