Abstract Objective: This study aims to explore the clinical diagnosis and treatment methods of bladder malakoplakia (MUB) to enhance the understanding of the disease.
Methods: A retrospective analysis of the diagnosis and treatment processes of three cases of MUB treated in our department was conducted. Relevant literature from both domestic and international sources was reviewed to provide a comprehensive analysis.
Results: All three patients underwent transurethral resection of bladder lesions combined with antibiotic therapy, and two of them received transurethral instillation of gemcitabine. There were two cases with two recurrences each, and one case with four recurrences, with the latter also concurrently presenting with unilateral ureteral malakoplakia. Postoperative pathology confirmed MUB in all three cases. Close follow-up revealed no significant recurrence in the patients.
Conclusion: The effective diagnosis rate is increased by conducting multiple deep, repetitive, and randomly selected live tissue examinations. The definitive diagnosis of MUB relies on pathological histological examination. Treatment involving a combination of antibiotics and transurethral resection of bladder lesions proves to be effective. Exploring the use of bladder instillation of gemcitabine widens the spectrum of MUB treatment methods.

Abstract Background: T cell exhaustion (TEX) signifies a condition of T cell disorder which implicate the therapeutic benefits and prognostic significance in patients with cancer. However, its role in the Head and Neck Squamous Carcinoma (HNSCC) remains incompletely understood.
Methods: The detailed data of HNSCC samples were obtained from The Cancer Genome Atlas (TCGA) database and two Gene Expression Omnibus (GEO) datasets. We computed the expression scores of four TEX-related pathways and detected gene modules closely linked to these pathways, indicating prognostic significance. Following this, regression analyses were performed to select eight genes for the development of a predictive signature. The predictive capacity of this signature was evaluated. Additionally, we examined the relationships between TEX-related signature risk scores and the effectiveness of immunotherapy as well as drug sensitivity.
Results: A novel prognostic model, comprising eight TEX-related genes, was established for patients with HNSCC. The prognostic value was further confirmed using additional GEO datasets: GSE65858 and GSE27020. This signature enables the stratification of patients into high- and low- risk groups, each showing distinct survival outcomes and responsiveness to immunotherapy. The low-risk group demonstrated improved prognosis and enhanced efficacy of immunotherapy. In addition, AZD6482, TAF1, Ribociclib, LGK974, PF4708671 and other drugs showed increased sensitivity in the high-risk group based on drug sensitivity values, offering tailored therapeutic recommendations for individuals with various risks profiles.
Conclusion: In conclusion, we developed a novel T cell exhaustion-associated signature, which holds considerable predictive value for both the prognosis of patients with HNSCC and the effectiveness of tumor immunotherapy.

Abstract Tieguanyin is one of the top ten most popular teas and the representative of oolong tea in China. In this study, a rapid and non-destructive method is developed to detect adulterated tea and its degree. Benshan is used as the adulterated tea, which is about 0%, 10%, 20%, 30%, 40%, and 50% of the total weight of tea samples, mixed with Tieguanyin. Taking the fluorescence spectra from 475 to 1000 nm, we then established the 2-and 6-class discriminant models. The 2-class discriminant models had the best evaluation index when using SG-CARS-SVM, which can reach a 100.00% overall accuracy, 100.00% specificity, 100% sensitivity, and the least time was 1.2088 s, which can accurately identify pure and adulterated tea; among the 6-class discriminant models (0% (pure Tieguanyin), 10, 20, 30, 40, and 50%), with the increasing difficulty of adulteration, SNV-RF-SVM had the best evaluation index, the highest overall accuracy reached 94.27%, and the least time was 0.00698 s. In general, the results indicated that the two classification methods explored in this study can obtain the best effects. The fluorescence hyperspectral technology has a broad scope and feasibility in the non-destructive detection of adulterated tea and other fields.

Abstract Numerous pieces of evidence have indicated that microRNA (miRNA) plays a crucial role in a series of significant biological processes and is closely related to complex disease. However, the traditional biological experimental methods used to verify disease-related miRNAs are inefficient and expensive. Thus, it is necessary to design some excellent approaches to improve efficiency. In this work, a novel method (CFSAEMDA) is proposed for the prediction of unknown miRNA–disease associations (MDAs). Specifically, we first capture the interactive features of miRNA and disease by integrating multi-source information. Then, the stacked autoencoder is applied for obtaining the underlying feature representation. Finally, the modified cascade forest model is employed to complete the final prediction. The experimental results present that the AUC value obtained by our method is 97.67%. The performance of CFSAEMDA is superior to several of the latest methods. In addition, case studies conducted on lung neoplasms, breast neoplasms and hepatocellular carcinoma further show that the CFSAEMDA method may be regarded as a utility approach to infer unknown disease–miRNA relationships.

Abstract [Objective] As an important measure to stimulate traditional development drivers, the role of digital transition in promoting resource-based enterprises to achieve high-quality economic development needs to be examined. [Methods] This study used the sample data of Chinese resource-based enterprises from 2011 to 2019 and text analysis techniques to depict digital transition, and empirically examined the impact of digital transition on total factor productivity of resource-based enterprises and mechanism based on panel models and threshold models. [Results] The research found that: (1) Digital transition significantly improved total factor productivity of resource-based enterprises in the four aspects of mining, production and manufacturing, business management, and raw material transportation, and this conclusion is still valid after a series of robustness tests. (2) Digital transition had a positive impact on total factor productivity of resource-based enterprises by promoting specialized division of labor. Digital transition promoted specialized division of labor within the enterprises to maximize the comparative advantages of various production factors and improve the total factor productivity of resource-based enterprises. It also broke through time and space constraints from outside the enterprises, reduced transaction costs and search costs, encouraged more enterprises to participate in specialized division of labor, and further promoted the growth of total factor productivity of resource-based enterprises. (3) The promoting effect of digital transition on total factor productivity of resource-based enterprises presented a dynamic nonlinear trend of increasing marginal effect. Under the threshold of specialized division of labor, digital transition had a more significant promoting effect on total factor productivity of resource-based enterprises, indicating that specialized division of labor can strengthen this effect and accelerate the improvement of total factor productivity of resource-based enterprises. ...

Abstract The antitumor drug candidate X-05 is being developed as an innovative anti-lung cancer drug candidate due to its excellent antitumour activity. A Caco-2 cell permeability study and solubility study confirmed that X-05 belonged to BCS class or compounds. Therefore, the main challenge is to develop appropriate preparations for preclinical studies and further clinical phase research. By evaluating the preliminary results of kinetic solubility in biorelevant media and the structural analysis of X-05 and polymers, three polymers PVP K30, PVP VA 64 and HPMCAS, which may have intermolecular interactions with X-05, were chosen to select the optimal carrier for X-05 to prepare amorphous solid dispersions (ASDs). ASD X-05-PVP VA 64 was selected as the optimal polymer by evaluating its kinetic solubility in biorelevant media and solid stability. The physical and chemical properties of ASD X-05-PVP VA 64 remain stable when the drug loading is as high as 50%. The drug-polymer interactions of ASD X-05-PVP VA 64 were studied by ultraviolet spectrophotometry, nuclear magnetic resonance spectrometry, infrared and Raman spectrophotometry, and the results indicated that the intermolecular hydrogen bond interaction between the drug and polymer was the foundation of the solubilization and stabilization of X-05 in PVP VA 64.
