    Abstract Background: No treatment other than platelet administration is known to protect against spontaneous hemorrhage in thrombocytopenic dogs.
    Objectives: Primary: determine if treatment with ε-aminocaproic acid (EACA) decreases the requirement for blood transfusions and improves outcome in dogs with severe thrombocytopenia. Secondary: find evidence of hyperfibrinolysis and determine the effect EACA administration on rapid (rTEG) and tissue plasminogen activator-spiked (tPA-rTEG) thromboelastography parameters.
    Animals: Twenty-seven dogs with severe thrombocytopenia were treated with EACA, and data from an additional 33 were obtained from the hospital database as historical control (HC) cohort.
    Methods: Single arm clinical trial with HCs. The EACA group dogs received EACA (100 mg/kg IV followed by a constant-rate infusion [CRI] of 400 mg/kg/24 hours). Thromboelastography before and during EACA infusion, hospitalization days, number of transfusions, and mortality were compared.
    Results: No difference was found in number of transfusions per dog (median, interquartile range; 1, 0-2.5 vs 0.9, 0-2; P = .5) and hospitalization days (4, 4-6 vs 4.5, 3.75-6; P = .83) between HC and EACA groups, respectively, and no difference in survival was identified by log-rank analysis (P = .15). Maximum amplitude on both rTEG and tPA-rTEG increased after EACA administration (rTEG baseline: 23.6, 9.6-38.9; post-EACA: 27.3, 19.8-43.2; P < .001; tPA-rTEG baseline: 23, 10.9-37.2; post-EACA: 24.7, 16.7-44.8; P < .002).
    Conclusions and clinical importance: Although EACA increased clot strength, there was no effect on outcome. Treatment with EACA at this dosage cannot be recommended as a routine treatment but may be considered for dogs with severe ongoing hemorrhage.

    Abstract Background: Childhood trauma (CT) are frequently reported by patients with bipolar disorder (BD), but it is unclear whether and how CT contribute to patients' cognitive and psychosocial impairments. We aimed to examine the impact of CT on cognition and psychosocial functioning in a large sample of 345 patients with BD and 183 healthy control participants (HC) using the Childhood Trauma Questionnaire, neurocognitive tests and ratings of mood symptoms and functioning.
    Results: Patients showed broad cognitive impairments across memory, attention and executive function and functional disability despite being in partial or full remission and had higher levels of CT than HC. Higher levels of CT correlated with impairments across almost all cognitive domains and lower psychosocial functioning across BD patients and HC. Of these, the associations between CT and poorer working memory and lower psychosocial functioning, respectively, prevailed after adjusting for clinical and demographical variables. Diagnosis of BD and estimated verbal intelligence did not moderate these associations. Analysis of CT sub-categories showed that working memory impairments were related particularly to childhood physical and emotional abuse, while psychosocial difficulties were related to physical and emotional neglect.
    Conclusions: CT may have negative implications for working memory and psychosocial functioning across both BD and healthy populations. If the findings are replicated, this would suggest that early interventions that reduce the frequency of CT in vulnerable families may aid children's cognitive and psychosocial development.

    Abstract The neurocognitive mechanism underlying negation processing remains controversial. While negation is suggested to modulate the access of word meaning, no such evidence has been observed in the event-related potential (ERP) literature on sentence processing. In the current study, we applied both univariate ERP and multivariate pattern analysis (MVPA) methods to examine the processing of sentence negation. We investigated two types of negative congruent/incongruent sentence pairs with truth-value evaluation (e.g., "A robin is a/not a bird") and without (e.g., "The woman reads a/no book"). In the N400 time window, ERPs consistently showed increased negativity for negative and incongruent conditions. MVPA, on the other hand, revealed nuanced interactions between polarity and congruency. In the later P600 time window, MVPA but not the ERPs revealed an effect of congruency, which may be functionally distinct from the N400 window. We further used cross-decoding to show that the cognitive processes underlying the N400 window for both affirmative and negative sentences are comparable, whereas in the P600 window, only for the truth sentences, negative sentences showed a distinct pattern from their affirmative counterparts. Our results thus speak for a more interactive, but nevertheless serial and biphasic, and potentially construction-specific processing account of negation. We also discuss the advantage of applying MVPA in addition to the classical univariate methods for a better understanding of the neurobiology of negation processing and language comprehension alike.

    Abstract Bipolar disorder (BD) is often accompanied by persistent cognitive impairment. However, screening for cognitive impairment in the clinic is challenged by a lack of consensus on screening procedures. This study assesses cognitive impairment prevalence and screening feasibility in alignment with the International Society for Bipolar Disorder Targeting Cognition Task Force recommendations. Between January 2022 and May 2023, 136 newly diagnosed BD outpatients were assessed with the Screen for Cognitive Impairment in Psychiatry after 15-20 months of specialised care at the Copenhagen Affective Disorder Clinic. Cognitive impairment patterns and associations with cognitive complaints, perceived stress, and functioning were examined. Most screened patients (73 %) achieved full or partial remission, with 51 % being cognitively normal, 38 % showing global impairments, and 11 % displaying selective impairments. Among remitted patients, 56 % were cognitively normal, while 31 % and 13 % exhibited global or selective impairments, respectively. Both objectively impaired patient groups reported more subjective cognitive difficulties than those who were cognitively normal. The globally impaired group also demonstrated poorer functioning, more depressive symptoms and lower quality of life than cognitively normal patients. Across all patients, lower cognitive performance correlated with more cognitive complaints, lower functioning, lower quality of life, and more depressive symptoms. Cognitive screenings were relatively easily implementable, involving only a 1.5 h session including mood ratings, feedback and cognitive strategy discussion. The study highlights the clinical relevance and feasibility of cognitive screenings in BD patients, emphasizing the need for tailored interventions given frequent cognitive impairment in clinically stable individuals.

    Abstract Background: Shared care between oncology specialists and general practice regarding the delivery of palliative care (PC) is necessary to meet the demands for a cohesive PC. The primary objective of this study is to investigate models of cross-sectorial integration between primary care and oncology specialists that have been developed to promote early and basic PC and factors influencing the process.
    Method: A scoping review was conducted using publications dated up until April 2023. Searches were conducted in MEDLINE, CINAHL, Embase, Web of Science and ProQuest Dissertations and Theses. Complementary searches were performed via reference lists and grey literature. Explicit early PC models aimed at patients with cancer aged ≥18 years with healthcare professionals from primary care and oncology constituted the inclusion criteria. The screening of the papers was performed independently by two reviewers. The reporting adheres to the extension for scoping reviews of the Preferred Reporting Items for Systematic Reviews and Meta-Analyses.
    Results: The search provided 5630 articles of which six met the eligibility criteria, each describing a different model of early and cross-sectorial, integrated PC. 12 active components were identified. Education of staff as well as good communication and cooperation skills are essential factors to succeed with integrated, early PC.
    Conclusion: Integration of PC between general practice and oncology specialists has potential. The components of basic PC have been established. Factors known to influence the process are trust, communication and a common goal. Further research is required into strategies for approaching different levels of integration.

    Abstract Imitation from observation (IfO) is a learning paradigm that consists of training autonomous agents in a Markov Decision Process (MDP) by observing expert demonstrations without access to its actions. These demonstrations could be sequences of environment states or raw visual observations of the environment. Recent work in IfO has focused on this problem in the case of observations of low-dimensional environment states, however, access to these highly-specific observations is unlikely in practice. In this paper, we adopt a challenging, but more realistic problem formulation, learning control policies that operate on a learned latent space with access only to visual demonstrations of an expert completing a task. We present BootIfOL, an IfO algorithm that aims to learn a reward function that takes an agent trajectory and compares it to an expert, providing rewards based on similarity to agent behavior and implicit goal. We consider this reward function to be a distance metric between trajectories of agent behavior and learn it via contrastive learning. The contrastive learning objective aims to closely represent expert trajectories and to distance them from non-expert trajectories. The set of non-expert trajectories used in contrastive learning is made progressively more complex by bootstrapping from roll-outs of the agent learned through RL using the current reward function. We evaluate our approach on a variety of control tasks showing that we can train effective policies using a limited number of demonstrative trajectories, greatly improving on prior approaches that consider raw observations.
