    Abstract Abstract In recent years, coal and gas outburst disasters are still occurring and difficult to prevent, seriously endangering the safety of coal mine production. It is well known that the transporting and crushing of outburst coal is the main pathway of energy dissipation during the coal and gas outburst process. However, a consensus regarding how much gas involves in outburst and affects energy dissipation is still lacking. Quantitative study on the gas effect on migration and fragmentation characteristics of outburst coal in restricted roadway space can improve the energy model and guide the prevention and control of gas outburst. In this paper, an improved visual coal and gas outburst dynamic effect simulation experiment system was used to conduct outburst simulation experiments at different gas pressure conditions. The results showed that the movement of outburst coal in the roadway has experienced various flow patterns. In the initial stage of the outburst, under low gas pressure condition, the motion of the outburst coal was dominated by stratified flow. However, as the gas pressure increases, the initial acceleration increases, and the outburst coal mainly move forward rapidly in the form of plug flow. The average velocity at 0.3, 0.5, and 0.8 MPa gas pressure condition were 6.75, 22.22 and 35.81 m/s, respectively. Gas also has a crushing effect on outburst coal. With increasing gas pressure, the number of coal powder particles of the same mass increased significantly, and the range of the particle size distribution of the particles decreaed, and the median particle size decreased. As the gas pressure increases, the outburst intensity gradually increases, and the total energy involved in the outburst work also increases. However, the energy dissipation pathways are different. At 0.3 MPa, the energy dissipation is dominated by crushing energy, which is about six times the ejection energy. It is well known that the transporting and crushing of outburst coal is the main pathway of energy dissipation during the coal and gas outburst process. As the gas pressure increased to 0.8 MPa, the proportion of the ejection energy gradually increases to about twice that of the crushing energy. Under the experimental conditions, 2.71-13.43% of the adsorbed gas involves in the outburst (AGIO) through rapid desorption, and the proportion increases with increasing gas pressure. This paper improves the energy model of coal and gas outburst, which is applicable to risk assessment and prevention of outburst disasters.

    Abstract Perioperative neurocognitive impairment (PND) is a common medical complication in the postoperative period. General anesthesia through volatile anesthetics poses a high risk of POCD. Moreover, the developing brain is especially vulnerable to anesthesia-induced neurotoxicity. Therefore, finding a practical approach to prevent or alleviate neonatal isoflurane (ISO) exposure-induced brain injury and cognitive decline is essential for reducing medical complications following major surgery during the early postnatal period. Using a repeated neonatal ISO exposure-induced PND rat model, we investigated the effects of methylene blue (MB) pretreatment on repeated neonatal isoflurane exposure-induced brain injury and memory loss. Intraperitoneal injection of low-dose MB (1 mg/kg) was conducted three times 24 h before each ISO exposure. The Barnes maze and novel objection test were conducted to assess learning and memory. Immunofluorescence staining, F-Jade C staining, TUNEL staining, and Western blot analysis were performed to determine mitochondrial fragmentation, neuronal injury, degeneration, and apoptosis. Evans blue extravasation assay, total antioxidant capacity assay, MDA assay kit, and related inflammatory assay kits were used to test blood-brain barrier (BBB) disruption, antioxidant capacity, and neuroinflammation. Behavioral tests revealed that MB pretreatment significantly ameliorated ISO exposure-induced cognitive deficits. In addition, MB pretreatment alleviates neuronal injury, apoptosis, and degeneration. Furthermore, the BBB integrity was preserved by MB pretreatment. Additional studies revealed that ISO-induced excessive mitochondrial fragmentation, oxidative stress, and neuroinflammation were significantly attenuated by MB pretreatment in the PND rat model. Our findings suggest that MB pretreatment alleviates ISO exposure-induced brain injury and memory loss for the first time, supporting MB pretreatment as a promising approach to protect the brain against neonatal ISO exposure-induced postoperative cognitive dysfunction.

    Abstract Abstract Background Aberrant expression and activation of circular RNAs (circRNAs) are closely associated with various cancers. The role of circ_MAPK9 (hsa_circ_0001566) in cancer progression remains unknown. This study aims to investigate the function, mechanism and clinical significance of circ_MAPK9 in hepatocellular carcinoma (HCC). Methods Circ_MAPK9 expression on the microarray of tumor from clinical HCC patients was detected by in situ hybridization (ISH). Circ_MAPK9 knockdown was achieved with siRNAs in SMMC-7721 and SK-Hep1 HCC cell lines. The biological function of circ_MAPK9 was verified in vitro by CCK8 test, colony formation assay, transwell assay, PI-Annexin V staining, and in vivo by xenograft tumor in nude mice. Fluorescent in situ hybridization (FISH), subcellular fractionation assay, a dual-luciferase reporter assay and rescue experiments were employed for further mechanistic investigation. Results The expression of circ_MAPK9 was significantly up-regulated in HCC tissues and cells, which was found to be associated with poor prognosis. Patients with high expression of circ_MAPK9 had a shorter overall survival and disease-free survival in comparison to those with low circ_MAPK9 expression. Functional assays showed that circ_MAPK9 knockdown suppressed cellular proliferation, migration, invasion and tumor growth in vivo, and promoted apoptosis in HCC cells. Moreover, we found that circ_MAPK9 knockdown could inhibit aerobic glycolysis by decreasing the production of adenosine triphosphate (ATP) and lactic acid, which was mediated by lactate dehydrogenase (LDHA). Mechanistically, circ_MAPK9 functioned as ceRNA via sponging miR-642b-3p and alleviated the inhibitory effect of miR-642b-3p on its target signal transducer and activator of transcription 3 (STAT3) and LDHA, thereby leading to STAT3 activation and LDHA expression.     Abstract Glycosylation, a fundamental biological process, involves the attachment of glycans to proteins, lipids, and RNA, and it plays a crucial role in various biological pathways. It is of great significance to obtain the precise spatial distribution of glycosylation modifications at the cellular and tissue levels. Here, we introduce LectoScape, an innovative method enabling detailed imaging of tissue glycomes with up to 1 μm resolution through image mass cytometry (IMC). This method utilizes 12 distinct, nonoverlapping lectins selected via microarray technology, enabling the multiplexed detection of a wide array of glycans. Furthermore, we developed an efficient labeling strategy for these lectins. Crucially, our approach facilitates the concurrent imaging of diverse glycan motifs, including N-glycan and O-glycan, surpassing the capabilities of existing technologies. Using LectoScape, we have successfully delineated unique glycan structures in various cell types, enhancing our understanding of the glycan distribution across human tissues. Our method has identified specific glycan markers, such as α2,3-sialylated Galβ1, 3GalNAc in O-glycan, and terminal GalNAc, as diagnostic indicators for cervical intraepithelial neoplasia. This highlights the potential of LectoScape in cancer diagnostics through the detection of abnormal glycosylation patterns.

    Abstract Deep learning techniques have been successfully applied in handwriting recognition. Oracle bone inscriptions (OBI) are the earliest hieroglyphs in China and valuable resources for studying the etymology of Chinese characters. OBI are of important historical and cultural value in China; thus, textual research surrounding the characters of OBI is a huge challenge for archaeologists. In this work, we built a dataset named OBI-100, which contains 100 classes of oracle bone inscriptions collected from two OBI dictionaries. The dataset includes more than 128,000 character samples related to the natural environment, humans, animals, plants, etc. In addition, we propose improved models based on three typical deep convolutional network structures to recognize the OBI-100 dataset. By modifying the parameters, adjusting the network structures, and adopting optimization strategies, we demonstrate experimentally that these models perform fairly well in OBI recognition. For the 100-category OBI classification task, the optimal model achieves an accuracy of 99.5%, which shows competitive performance compared with other state-of-the-art approaches. We hope that this work can provide a valuable tool for character recognition of OBI.

    Abstract Alzheimer's disease, one of the most common forms of dementia, is characterized by a slow progression of cognitive impairment and neuronal loss. Currently, approved treatments for AD are hindered by various side effects and limited efficacy. Despite considerable research, practical treatments for AD have not been developed. Increasing evidence shows that glial cells, especially microglia and astrocytes, are essential in the initiation and progression of AD. During AD progression, activated resident microglia increases the ability of resting astrocytes to transform into reactive astrocytes, promoting neurodegeneration. Extensive clinical and molecular studies show the involvement of microglia and astrocyte-mediated neuroinflammation in AD pathology, indicating that microglia and astrocytes may be potential therapeutic targets for AD. This review will summarize the significant and recent advances of microglia and astrocytes in the pathogenesis of AD in three parts. First, we will review the typical pathological changes of AD and discuss microglia and astrocytes in terms of function and phenotypic changes. Second, we will describe microglia and astrocytes' physiological and pathological role in AD. These roles include the inflammatory response, "eat me" and "don't eat me" signals, Aβ seeding, propagation, clearance, synapse loss, synaptic pruning, remyelination, and demyelination. Last, we will review the pharmacological and non-pharmacological therapies targeting microglia and astrocytes in AD. We conclude that microglia and astrocytes are essential in the initiation and development of AD. Therefore, understanding the new role of microglia and astrocytes in AD progression is critical for future AD studies and clinical trials. Moreover, pharmacological, and non-pharmacological therapies targeting microglia and astrocytes, with specific studies investigating microglia and astrocyte-mediated neuronal damage and repair, may be a promising research direction for future studies regarding AD treatment and prevention.
