    Abstract The Timepix3 readout ASIC chip is a hybrid pixelated radiation detector, designed at CERN, which contains a 256 px × 256 px matrix. Each of the 65,536 radiation-sensitive pixels can record an incoming particle, its energy deposition or time of arrival and measure them simultaneously. Since the detector is suitable for a wide range of applications from particle physics, national security and medicine to space science, it can be used in a wide range of temperatures. Until now, it has to be calibrated every time to the operating point of the application. This paper studies the possibility of energy measurement with Timepix3 equipped with a 500 m thick silicon sensor and MiniPIX readout interface in the temperatures between 10 ∘C and 70 ∘C with only one calibration. The detector has been irradiated by X-ray fluorescence photons in the energy range from 8 keV to 57 keV, and 31 keV to 81 keV photons from the

    Abstract An understanding of the structural organisation and chemistry of the cell walls in woody tissues is crucial from the perspective of plant mechanical strength, water transportability, as well as subsequent commercial utilisation of the wood. Poplar trees (Populus sp.), grown on two reclamation substrates ("Humus" and "Sand") under the extreme soil conditions of an external coal mining spoil heap of the lignite mine in Bełchatów (Central Poland), were examined. Conventional parameters - tree-ring width (TRW) and wood density (WD) resolved annually (years 2008-2017) were corroborated by a novel approach of Raman spectroscopic analysis. Annually resolved Raman spectroscopic data representing the lignin-to-cellulose ratio (Li/Ce) enabled to estimate trends of lignification. The above traits were obtained for the three poplar genotypes: H-275, Grandis, and Androscoggin to assess the suitability of their plantation on the reclaimed heap. Our results show a significant effect of genotype on TRW, WD, and the Raman Li/Ce, while the effect of the soil substrate was less pronounced. The highest Li/Ce was identified in the H-275 genotype grown on a substrate with hummus. H-275 also showed higher TRW values compared to the other genotypes. WD was significantly higher in Grandis and Androscoggin genotypes grown on the "Sand" substrate. Associations between tree-ring parameters and climatic variables (temperature and precipitation) were mostly low and not statistically significant. Our findings from individual tree rings indicate that the genotype is the crucial factor influencing the lignification of poplar trees grown on post-mining lands.

    Abstract Pleural empyema is a serious condition leading to a significant burden on health care systems due to protracted hospitalisations. Treatment ranges from non-surgical interventions such as antibiotic therapy and chest tube placement to thoracoscopic or open surgery. Various risk factors which impact outcomes have been investigated. The RAPID (renal, age, purulence, infection source, and dietary factors) score is a clinical risk score which identifies patients at risk of death and may be used to formulate individual treatment strategies accordingly. All patients undergoing surgical interventions for empyema at a major tertiary medical centre in Germany from 2017 to 2020 were analysed. The aim was to identify perioperative risk factors which significantly impact treatment outcomes but are currently not included in the RAPID score. 245 patients with pleural empyema surgically treated at the Department of General, Visceral and Thoracic Surgery at the University Medical Centre, Hamburg, Germany (admitted from January 2017 to April 2020) were retrospectively analysed. All patients which received either minimally invasive or open thoracic surgery were included. Epidemiological as well as perioperative data was analysed to identify risk factors which impact long-term overall outcomes. 90-day mortality rate was the primary endpoint. The mean age was 59.4 years with a bimodal distribution. There was a male predominance across the cohort (71.4% compared to 28.6%), with no significant differences across ages below or above 60 years. 53 (21.6%) patients died within the first 90 days. Diabetes type 1 and 2, renal replacement therapy, immunosuppression, postoperative bleeding, intraoperative transfusion as well as microbiologically confirmed bacterial invasion of the pleura all led to higher mortality rates. Higher RAPID scores accurately predicted higher 90-day mortality rates. Modifying the RAPID score by adding the comorbidities diabetes and renal replacement therapy significantly increased the predictive value of the score. We demonstrated various perioperative and patient related risk-factors not included in the RAPID score which negatively impact postoperative outcome in patients receiving surgical treatment for pleural empyema. These should be taken into consideration when deciding on the best course of treatment. If confirmed in a prospective study including non-surgical patients with a significantly larger cohort, it may be worth considering expanding the RAPID score to include these.

    Abstract An all-soft multi-material combination consisting of a hydrogel based on poly(ethylene glycol) (PEG) coated with spatially defined spots of gelatin methacryloyl (GM) containing selectively addressable viral nanorods is presented, and its basic application as a qualitative biosensor with reporter enzymes displayed on the tobacco mosaic virus (TMV) bioscaffolds within the GM is demonstrated. Biologically inert PEG supports are equipped with GM spots serving as biological matrix for enzymes clustered on TMV particles preventing diffusion out of the gel. For this multi-material combination, i) the PEG-based hydrogel surface is modified to achieve a clear boundary between coated and non-coated regions by introducing either isothiouronium or thiol groups. ii) Cross-linking of the GM spots is studied to achieve anchoring to the hydrogel surface. iii) The enzymes horseradish peroxidase or penicillinase (Pen) are conjugated to TMV and integrated into the GM matrix. In contrast to free enzymes, enzyme-decorated TMVs persist in GM spots and show sustained enzyme activity as evidenced by specific color reaction after 7 days of washing, and for Pen after 22 months after dry storage. Therefore, the integration of enzyme-coupled TMV into hydrogel matrices is a promising and versatile approach to obtaining reusable and analyte-specific sensor components.

    Abstract The mRNA technology has emerged as a rapid modality to develop vaccines during pandemic situations with the potential to protect against endemic diseases. The success of mRNA in producing an antigen is dependent on the ability to deliver mRNA to the cells using a vehicle, which typically consists of a lipid nanoparticle (LNP). Self-amplifying mRNA (SAM) is a synthetic mRNA platform that, besides encoding for the antigen of interest, includes the replication machinery for mRNA amplification in the cells. Thus, SAM can generate many antigen encoding mRNA copies and prolong expression of the antigen with lower doses than those required for conventional mRNA. This work describes the morphology of LNPs containing encapsulated SAM (SAM LNPs), with SAM being three to four times larger than conventional mRNA. We show evidence that SAM changes its conformational structure when encapsulated in LNPs, becoming more compact than the free SAM form. A characteristic "bleb" structure is observed in SAM LNPs, which consists of a lipid-rich core and an aqueous RNA-rich core, both surrounded by a DSPC-rich lipid shell. We used SANS and SAXS data to confirm that the prevalent morphology of the LNP consists of two-core compartments where components are heterogeneously distributed between the two cores and the shell. A capped cylinder core-shell model with two interior compartments was built to capture the overall morphology of the LNP. These findings provide evidence that bleb two-compartment structures can be a representative morphology in SAM LNPs and highlight the need for additional studies that elucidate the role of spherical and bleb morphologies, their mechanisms of formation, and the parameters that lead to a particular morphology for a rational design of LNPs for mRNA delivery.
