    Abstract BACKGROUND: The European beech is arguably the most important climax broad-leaved tree species in Central Europe, widely planted for its valuable wood. Here, we report the 542 Mb draft genome sequence of an up to 300-year-old individual (Bhaga) from an undisturbed stand in the Kellerwald-Edersee National Park in central Germany. FINDINGS: Using a hybrid assembly approach, Illumina reads with short- and long-insert libraries, coupled with long Pacific Biosciences reads, we obtained an assembled genome size of 542 Mb, in line with flow cytometric genome size estimation. The largest scaffold was of 1.15 Mb, the N50 length was 145 kb, and the L50 count was 983. The assembly contained 0.12% of Ns. A Benchmarking with Universal Single-Copy Orthologs (BUSCO) analysis retrieved 94% complete BUSCO genes, well in the range of other high-quality draft genomes of trees. A total of 62,012 protein-coding genes were predicted, assisted by transcriptome sequencing. In addition, we are reporting an efficient method for extracting high-molecular-weight DNA from dormant buds, by which contamination by environmental bacteria and fungi was kept at a minimum. CONCLUSIONS: The assembled genome will be a valuable resource and reference for future population genomics studies on the evolution and past climate change adaptation of beech and will be helpful for identifying genes, e.g., involved in drought tolerance, in order to select and breed individuals to adapt forestry to climate change in Europe. A continuously updated genome browser and download page can be accessed from beechgenome.net, which will include future genome versions of the reference individual Bhaga, as new sequencing approaches develop.

    Abstract This study provides insights into factors that influence the water balance of selected European lakes, mainly in Central Europe, and their implications for water quality. An analysis of isotopic, chemical and land use data using statistical and artificial intelligence models showed that climate, particularly air temperature and precipitation, played a key role in intensifying evaporation losses from the lakes. Water balance was also affected by catchment factors, notably groundwater table depth. The study shows that lakes at lower altitudes with shallow depths and catchments dominated by urban or crop cover were more sensitive to water balance changes. These lakes had higher evaporation-to-inflow ratios and increased concentrations of total nitrogen in the water. On the other hand, lakes at higher elevations with deeper depths and prevailing forest cover in the catchment were less sensitive to water balance changes. These lakes, which are often of glacial origin, were characterized by lower evaporation losses and thus better water quality in terms of total nitrogen concentrations. Understanding connections between water balance and water quality is crucial for effective lake management and the preservation of freshwater ecosystems.

    Abstract We determine the time dependence of pressure and shear stress distributions on the surface of a pitching and deforming hydrofoil from measurements of the three dimensional flow field. Period-averaged stress maps are obtained both in the presence and absence of steady flow around the foil. The velocity vector field is determined via volumetric three-component particle tracking velocimetry and subsequently inserted into the Navier-Stokes equation to calculate the total hydrodynamic stress tensor. In addition, we also present a careful error analysis of such measurements, showing that local evaluations of stress distributions are possible. The consistency of the force time-dependence is verified using a control volume analysis. The flapping foil used in the experiments is designed to allow comparison with a small trapezoidal fish fin, in terms of the scaling laws that govern the oscillatory flow regime. As a complementary approach, unsteady Euler-Bernoulli beam theory is employed to derive instantaneous transversal force distributions on the flexible hydrofoil from its deflection and the results are compared to the spatial distributions of hydrodynamic stresses obtained from the fluid velocity field.

    Abstract Introduction: Chlorpromazine is a commonly used drug in several medical conditions associated with a wide range of side effects. Few cases of hemostatic disorder have been reported in the literature.
    Case report: A 39-year-old man had previously been diagnosed with a cavernous malformation of the medulla oblongata. Chlorpromazine was started to treat persistent hiccups. Twenty days later, the patient presented hepatitis and a pruritic rash. Haemostasis tests revealed a prolonged partial thromboplastin time associated with isolated decrease of factor VIII level and anti-factor VIII antibodies. Magnetic resonance imaging revealed recent asymptomatic bleeding. Introduction of eptacog alfa and prednisone allowed clinical and biological improvement as well as a prolonged remission after 12 months of follow-up.

    Abstract Objective: Stratifying the risk of death in SSc-related interstitial lung disease (SSc-ILD) is a challenging issue. The extent of lung fibrosis on high-resolution CT (HRCT) is often assessed by a visual semiquantitative method that lacks reliability. We aimed to assess the potential prognostic value of a deep-learning-based algorithm enabling automated quantification of ILD on HRCT in patients with SSc.
    Methods: We correlated the extent of ILD with the occurrence of death during follow-up, and evaluated the additional value of ILD extent in predicting death based on a prognostic model including well-known risk factors in SSc.
    Results: We included 318 patients with SSc, among whom 196 had ILD; the median follow-up was 94 months (interquartile range 73-111). The mortality rate was 1.6% at 2 years and 26.3% at 10 years. For each 1% increase in the baseline ILD extent (up to 30% of the lung), the risk of death at 10 years was increased by 4% (hazard ratio 1.04, 95% CI 1.01, 1.07, P = 0.004). We constructed a risk prediction model that showed good discrimination for 10-year mortality (c index 0.789). Adding the automated quantification of ILD significantly improved the model for 10-year survival prediction (P = 0.007). Its discrimination was only marginally improved, but it improved prediction of 2-year mortality (difference in time-dependent area under the curve 0.043, 95% CI 0.002, 0.084, P = 0.040).
    Conclusion: The deep-learning-based, computer-aided quantification of ILD extent on HRCT provides an effective tool for risk stratification in SSc. It might help identify patients at short-term risk of death.

    Abstract Research focused on succinate dehydrogenase (SDH) and its substrate, succinate, culminated in the 1950s accompanying the rapid development of research dedicated to bioenergetics and intermediary metabolism. This allowed researchers to uncover the implication of SDH in both the mitochondrial respiratory chain and the Krebs cycle. Nowadays, this theme is experiencing a real revival following the discovery of the role of SDH and succinate in a subset of tumors and cancers in humans. The aim of this review is to enlighten the many questions yet unanswered, ranging from fundamental to clinically oriented aspects, up to the danger of the current use of SDH as a target for a subclass of pesticides.

    Abstract The Curuaí floodplain in the low Amazon river in the Pará state of Brazil and Juruá basin, a major Solimões tributary. Characterizing the hydrological dynamics of Amazon floodplains is essential to better understand and preserve these environments providing important resources to local populations. Radar altimetry is an effective remote sensing tool for monitoring water levels of continental hydrosystems, including floodplains. An unsupervised classification approach on radar echoes to determine hydrological regimes has recently been tested and showed a strong potential on the Congo River basin. This method is adapted to Envisat and Saral satellite radar altimetry data on two study areas in the Amazon Basin. The aim is to improve inland water detection along altimeter tracks to automatically generate water level time series (WLTS) over rivers, lakes, and poorly monitored floodplains and wetlands. Results show a good agreement with land cover maps obtained with optical imagery over selected Amazonian wetlands (70–80% accuracies with Envisat data and 50–60% with Saral data). Automatically generated WLTS are strongly correlated to the manually generated WLTS ( ≈ 0.9; RMSE < 1 m). Compared to the manual method, the automatic method is faster, more efficient and replicable. Densifying the WL network in the floodplains bring crucial information on the connectivity dynamic between lakes and rivers.

    Abstract Background: Reduced gastric motility in Parkinson's disease (PD) has been reported, but hardly any study exists in subjects with isolated rapid-eye-movement (REM) sleep behavior disorder (iRBD), a specific prodrome of α-synucleinopathies.
    Objectives: We compared the gastric motility of 17 iRBD subjects with that of 18 PD subjects (15 drug naive, 3 early treated in defined off) and 15 healthy controls (HC) with real-time magnetic resonance imaging (rtMRI).
    Methods: After overnight fasting, participants consumed a standardized breakfast and underwent a 3-T rtMRI of the stomach. Amplitude and velocity of the peristaltic waves were analyzed under blinded conditions. Gastric motility index (GMI) was calculated. The procedure was repeated in 12 of 17 iRBD subjects ~2.5 years later. Nine of these 12 iRBD subjects were hyposmic.
    Results: In iRBD and PD subjects the amplitude of the peristaltic waves was significantly reduced compared with HCs (iRBD vs. HC: 8.7 ± 3.7 vs. 11.9 ± 4.1 mm, P = 0.0097; PD vs. HC: 6.8 ± 2.2 vs. 11.9 ± 4.1 mm, P = 0.0001). The amplitude in iRBD and PD subjects was decreased to the same extent. The GMI was reduced in only PD subjects (PD vs. HC: P = 0.0027; PD vs. iRBD: P = 0.0203). After ~2.5 years the amplitude in iRBD subjects did not significantly decrease further.
    Conclusion: The amplitude of the peristaltic waves was markedly reduced in iRBD, a prodrome of α-synucleinopathies. This reduction was similar to the extent observed already in manifest early PD. This finding implies that the α-synuclein pathology affects the innervation of the stomach already in the prodromal stage. © 2023 The Authors.     Abstract Since 1990, global forest area has been reducing; tropical forests have suffered from different anthropogenic and natural factors that account for forest loss. Tropical deforestation is the second driver of anthropogenic emissions; increasing demand and investments in tropical forests drive these emissions. These forests attract significant foreign direct investments, but the effects of these investments on carbon emissions from land use, land-use change, and forestry are not well enshrined in the literature for the countries under study. This paper seeks to analyse the impact of foreign direct investments on carbon emissions from land use, land-use change, and forestry amongst 30 tropical forest countries from 1996 to 2019. The sampled countries were disaggregated by tropical blocs: Amazon, Congo basin, Australasia, and Southeast Asia, and by income levels; low-income, lower-middle, upper-middle, and high-income. The findings reveal a U-shape structure of the impact of FDI on carbon emissions from LULUCF within Congo basin and Amazon countries; at higher levels of FDI, emissions from LULUCF will increase while Australasia and Southeast Asian countries show an inverted U-shape impact, thus at higher levels of FDI, there will be a negative and significant impact on carbon emissions from LULUCF. The income levels reveal an inverted U-shape for low-income and high-income countries and a U-shape for upper-middle-income countries; the impact for lower-middle-income countries is not significant. Overall, for the whole sample, the impact depicts a U-shape. This paper proposes high-level development of environmental conditions for FDI for different sectors that align with country and regional green growth plans. Enhancing national and regional governance systems to enforce decisions and fight corruption effectively can significantly promote green FDI for green growth.

    Abstract Background and objectives: To increase the validity of biomarker measures in multiple sclerosis (MS), factors affecting their concentration need to be identified. Here, we test whether the volume of distribution approximated by the patients' estimated blood volume (BV) and body mass index (BMI) affect the serum concentrations of glial fibrillary acidic protein (GFAP). As a control, we also determine the relationship between BV/BMI and GFAP concentrations in CSF. To confirm earlier findings, we test the same hypotheses for neurofilament light chain (NfL).
    Methods: NfL and GFAP concentrations were measured in serum and CSF (sNFL/sGFAP and cNFL/cGFAP) in 157 patients (n = 106 with MS phenotype and n = 51 with other neurologic/somatoform diseases). Using multivariate linear regressions, BV was tested in the MS cohort as a predictor for each of the biomarkers while controlling for age, sex, MS phenotype, Expanded Disability Status Scale score, gadolinium-enhancing lesions, and acute relapse.
