    Abstract Ethnopharmacological relevance: Baoyuan Decoction (BYD) was initially recorded in the classic of "Bo Ai Xin Jian" in the Ming dynasty. It is traditionally used for treating weakness and cowardice, and deficiency of vital energy. In researches related to anti-fatigue effects, the reciprocal regulation of AMPK and circadian clocks likely plays an important role in anti-fatigue mechanism, while it has not yet been revealed. Therefore, we elucidated the anti-fatigue mechanism of BYD through AMPK/CRY2/PER1 pathway.
    Aim of the study: To investigate the effect and mechanism of BYD in reducing fatigue, using pharmacodynamics, network pharmacology and transcriptomics through the AMPK/CRY2/PER1 signaling pathway.
    Materials and methods: Firstly, the chemical constituents of BYD were qualitatively identified by UHPLC-Q-Exactive Orbitrap/MS, establishing a comprehensive strategy with an in-house library, Xcalibur software and Pubchem combined. Secondly, a Na

    Conclusion: Herein, we identified 114 chemical constituents of BYD, performed zebrafish activity validation, while demonstrated that BYD can relieve fatigue by AMPK/CRY2/PER1 signaling pathway through network pharmacology and transcriptome.

    Abstract In this work, we develop and release Yuan 2.0, a series of large language models with parameters ranging from 2.1 billion to 102.6 billion. The Localized Filtering-based Attention (LFA) is introduced to incorporate prior knowledge of local dependencies of natural language into Attention. A data filtering and generating system is presented to build pre-training and fine-tuning dataset in high quality. A distributed training method with non-uniform pipeline parallel, data parallel, and optimizer parallel is proposed, which greatly reduces the bandwidth requirements of intra-node communication, and achieves good performance in large-scale distributed training. Yuan 2.0 models display impressive ability in code generation, math problem-solving, and chatting compared with existing models. The latest version of YUAN 2.0, including model weights and source code, is accessible at Github.

    Abstract Recent work like GPT-3 has demonstrated excellent performance of Zero-Shot and Few-Shot learning on many natural language processing (NLP) tasks by scaling up model size, dataset size and the amount of computation. However, training a model like GPT-3 requires huge amount of computational resources which makes it challengeable to researchers. In this work, we propose a method that incorporates large-scale distributed training performance into model architecture design. With this method, Yuan 1.0, the current largest singleton language model with 245B parameters, achieves excellent performance on thousands GPUs during training, and the state-of-the-art results on NLP tasks. A data processing method is designed to efficiently filter massive amount of raw data. The current largest high-quality Chinese corpus with 5TB high quality texts is built based on this method. In addition, a calibration and label expansion method is proposed to improve the Zero-Shot and Few-Shot performance, and steady improvement is observed on the accuracy of various tasks. Yuan 1.0 presents strong capacity of natural language generation, and the generated articles are difficult to distinguish from the human-written ones.

    Abstract A novel heteropolysaccharide with about twenty sugar residues named DF80-2 was obtained from Angelica pubescens Maxim. f. biserrata Shan et Yuan root, one of the most widely used traditional Chinese medicines for thousands of years in China. The possible structure of DF80-2 was proposed considering the comprehensive results of physicochemical properties, methylation analysis, and 1D/2D NMR spectroscopy, which showed that its main chain was composed of (1→3)-, and (1→4)-linked-α-d-Glcp, (1→4)-linked-β-d-Galp, (1→6)-linked-α-d-Manp, and (1→3)-linked-α-l-Araf, and the branch was present as the α-d-Glcp-(1→3)-β-d-GalpA disaccharides stretched from O-6 position of (1→4)-linked-α-d-Glc moiety in the main chain. Congo red analysis, scanning electron microscopy and atomic force microscopy showed that DF80-2 possessed a triple helical conformation, and its branched monomers were interlaced with one another forming a regular network structure. DF80-2 exhibited antioxidant activity by effectively scavenging DPPH, and hydroxyl radicals, and chelating ferrous ions.

    Abstract Yuan-Zhi Decoction (YZD) is a traditional Chinese medical formulation with demonstrated clinical benefits in Alzheimer's disease (AD). We used liquid chromatography coupled with mass spectrometry to identify 27 unique chemical components of YZD. Analyzing these using network pharmacology and molecular docking models identified 34 potential interacting molecular targets involved in 26 biochemical pathways. When tested in an animal model of AD, the APP/PS1 transgenic mice showed measurable improvements in spatial orientation and memory after the administration of YZD. These improvements coincided with significantly reduced deposition of Aβ plaques and tau protein in the hippocampi in the treated animals. In addition, a decreased BACE1 and beta-amyloid levels, a downregulation of the p-GSK-3β/GSK-3β, and an upregulation of the PI3K and p-AKT/AKT pathway was seen in YZD treated animals. These

    Abstract Objective: To extract and analyze information on the regularity with which wind-dispelling medication was prescribed by LI Dong-Yuan and many doctors of his school of thought and to provide theoretical basis and ideas for modern clinical application to facilitate the use, research, and development of these medications. Methods: Original data on wind-dispelling medication described in LI Dong-Yuan’s works were collected, sorted and organized. Data mining and comprehensive analysis were performed by using a series of data processing softwares, such as SPSS 22.0, Modeler 18.0 and xMiner. By reviewing LI Dong-Yuan's writings on wind-dispelling medication, the number of prescriptions for wind-dispelling medication were counted. Frequency of use of wind-dispelling medication was determined, and association rules analysis, factor analysis, and core drug network analysis were used to reveal associations of the symptoms of syndromes and treatment with wind-dispelling medication and to reveal the regularity with which these medications were prescribed by LI Dong-Yuan. Results: A total of 356 prescriptions and 121 flavors of wind-dispelling medication were observed. Among them, five kinds of medicinal properties, seven kinds of medicinal tastes, and 12 kinds of meridians were identified. Furthermore, herbs were divided into 18 categories according to their efficacy. Statistical analysis showed that there were 23 wind-dispelling medications that were prescribed with a frequency of ≥ 60. The mainly used drugs were Saposhnikoviae Radix (Fang Feng, 防风), Bupleuri Radix (Chai Hu, 柴胡), Cimicifugae Rhizoma (Sheng Ma, 升麻), Notopterygii Rhizoma Et Radix (Qiang Huo, 羌活), Puerariae Lobatae Radix (Ge Gen, 葛根), Chuanxiong Rhizoma (Chuan Xiong, 川芎), and Ephedrae Herba (Ma Huang, 麻黄).     Abstract Ethnopharmacological relevance: Gan Kang Yuan (GKY) is a compound medicine formulated on the basis of traditional Chinese medicine (TCM). It was composed of Herba Cistanchis (Roucongrong), Radix Puerariae (Gegen), Radix Astragali (Huangqi), Fructus Schisandrae (Wuweizi) and Radix Glycyrrhizae (Gancao).
    Aim: The purpose of this study is to research the hepatoprotective effect of GKY against liver injury induced by alcohol, and to elucidate the mechanism of hepatopretective effect.
    Materials and methods: Hepatoprotective activity of GKY was researched both in vivo and vitro. In vitro, effect of GKY on the survival rates of HepG2 cells were assessed. In vivo research, ICR mice were oral administrated with alcohol (Er Guo-tou white spirit, 56%, 6mL/kg, once per day) for 31 days to establish liver injury model. Meanwhile, positive group or experimental groups were treated with bicyclol (300mg/kg) or GKY (200, 600, 1800mg/kg). Serological indexes including aspartate and alanine transaminases (AST, ALT), γ-glutamyl transpeptidase (γ-GTP), total bilirubin (TBil), total cholesterol (TCHO) and serum triglyceride (STG) were estimated. Hepatic indicators including superoxide dismutase (SOD), glutathione S-transferase (GST), glutathione (GSH), glutathione peroxidase (GSH-Px), malondialdehyde (MDA), and liver triglyceride (LTG) were analyzed. Histopathologic changes of liver tissue were observed.
    Results: The survival rates of HepG2 cell were observably promoted by GKY. Alcoholic treatment drastically altered the serum indexes and liver indicators of model animals, while these alteration were significantly ameliorated by GKY (p < 0.05, 0.01 or 0.001) in experimental group. The microvesicular steatosis and necrosis in hepatic histopathology induced by alcoholic treatment also were notably attenuated by GKY administration.
    Conclusions: These findings indicated that GKY possessed hepatoprotective property against liver injury induced by ethanol. GKY significantly promoted activities of relative enzymes and suppressed the contents of MDA and LTG, which might be the mechanism of hepatoprotective effect of GKY.

    Abstract Chronic heart failure (CHF) leads to pulmonary structural remodeling, which may be a key factor for poor clinical outcomes in patients with end-stage heart failure, and few effective therapeutic options are presently available. The aim of the current study was to explore the mechanism of action and pulmonary-protective effects of treatment with Bao Yuan decoction combined with Tao Hong Si Wu decoction (BYTH) on lung structural remodeling in rats with ischemic heart failure. In a model of myocardial infarction (MI) induced by ligation of the left anterior descending (LAD) artery, rats were treated with BYTH. Heart function and morphometry were measured followed by echocardiography, histological staining, and immunohistochemical analysis of lung sections. The levels of transforming growth factor-β1 (TGF-β1), type I collagen, phosphorylated-Smad3 (p-Smad3), tumor necrosis factor-α (TNF-α), toll-like receptor 4 (TLR4), active nuclear factor κB (NF-κB) and alpha smooth muscle actin (α-SMA) were detected using Western blotting. Lung weight increased after an infarct with no evidence of pulmonary edema and returned to normal as a result of BYTH. In addition, BYTH treatment reduced levels of type I collagen, TGF-β1, and α-SMA expression and decreased the phosphorylation of Smad3 in the lungs of rats after MI. BYTH treatment also reduced the elevated levels of lung inflammatory mediators such as TNF-α, TLR4, and NF-κB. Results suggested that BYTH could effectively improve lung structural remodeling after MI because of its anti-inflammatory and anti-fibrotic action, which may be mediated by suppression of the TGF-β1/Smad3 and NF-κB signaling pathways.
