    Abstract We developed a simple coordinate transformation method for quickly locating features of interest (FOIs) of samples in transmission electron microscope (TEM). The method is well suited for conducting sample searches in aberration-corrected scanning/transmission electron microscopes (S/TEM), where the survey can be very time-consuming because of the limited field of view imposed by the highly excited objective lens after fine-tuning the aberration correctors. For implementation, a digital image of the sample and the TEM holder was captured using a simple stereo-optical microscope. Naturally presented geometric patterns on the holder were referenced to construct a projective transformation between the electron and optical coordinate systems. The test results demonstrated that the method was accurate and required no electron microscope or specimen holder modifications. Additionally, it eliminated the need to mount the sample onto specific patterned TEM grids or deposit markers, resulting in universal applications for most TEM samples, holders and electron microscopes for fast FOI identification. Furthermore, we implemented the method into a Gatan script for graphical-user-interface-based step-by-step instructions. Through online communication, the script enabled real-time navigation and tracking of the motion of samples in TEM on enlarged optical images with a panoramic view.

    Abstract The increasing trend in drought events under the background of global warming makes it more important to understand the drought effect on vegetation photosynthesis. While diverse global gross primary production (GPP) datasets were adopted to investigate the drought impact on photosynthesis, few studies focused on the discrepancies of drought response among different GPP datasets, especially for the cumulative drought impact. Therefore, a total of twenty-six global GPP datasets based on process, machine learning (ML), and light-use efficiency (LUE) model schemes were obtained to appraise the cumulative impact of drought stress on photosynthesis from 2001 to 2010. Moreover, a relatively reliable global pattern of drought’s cumulative effect on vegetation photosynthesis was acquired from these global GPP products through probability analysis. The results illustrated that the cumulative impact of drought existed in 52.11% of vegetation cover land with the cumulative time scales dominantly at a short term (1–4 months, 31.81%). Obvious heterogeneity of the drought cumulative effect in space and different vegetation functional types was observed, as the reliability of the drought effect decreased with latitude decreasing and a higher sensitivity to drought in herbaceous vegetation than woody plants. Our findings highlighted the importance of ways in characterizing moisture conditions across vegetation types among various GPP models and the necessity of GPP dataset selection in investigating drought effect on photosynthesis.

    Abstract Ferroptosis is a distinct type of regulated cell death characterized by iron overload and lipid peroxidation. Ferroptosis is regulated by numerous factors and controlled by several mechanisms. This cell death type has a relationship with the immune system, which may be regulated by damage-associated molecular patterns. Ferroptosis participates in the progression of autoimmune diseases, including autoimmune hepatitis, rheumatoid arthritis, systemic lupus erythematosus, inflammatory bowel disease, multiple sclerosis, Parkinson's Disease, psoriasis and insulin-dependent diabetes mellitus. The present review summarizes the role of ferroptosis in autoimmune disorders and discusses ferroptosis as a potential therapeutic target for autoimmune disease.

    Abstract Background: Immune escape remains a major challenge in the treatment of malignant tumors. Here, we studied the mechanisms underlying immune escape in the tumor microenvironment and identified a potential therapeutic target.
    Methods: Pathological specimens from patients with liver cancer, soft tissue sarcoma, and liver metastasis of colon cancer were subjected to immunohistochemistry analysis to detect the expression of programmed death-1 (PD-1) in the tumor microenvironment (TME). Additionally, the expression of regulatory T cells (Tregs) and long non-coding RNAs (lncRNAs), such as highly upregulated in liver cancer (HULC) was evaluated by fluorescence in situ hybridization, and the relationship between HULC, Treg cells, and PD-1 was determined.
