    Abstract Methylation-controlled J protein (MCJ) is a newly identified member of the DnaJ family of cochaperones. Hypermethylation-mediated transcriptional silencing of the MCJ gene has been associated with increased chemotherapeutic resistance in ovarian cancer. However, the biology and function of MCJ remain unknown. Here we show that MCJ is a type II transmembrane cochaperone localized in the Golgi network and present only in vertebrates. MCJ is expressed in drug-sensitive breast cancer cells but not in multidrug-resistant cells. The inhibition of MCJ expression increases resistance to specific drugs by inducing expression of the ABCB1 drug transporter that prevents intracellular drug accumulation. The induction of ABCB1 gene expression is mediated by increased levels of c-Jun due to an impaired degradation of this transcription factor in the absence of MCJ. Thus, MCJ is required in these cells to prevent c-Jun-mediated expression of ABCB1 and maintain drug response.

    Abstract Paleoenvironmental proxy data for ocean properties, eolian sediment input, and continental rainfall based on high-resolution analyses of sediment cores from the southwestern Black Sea and the northernmost Gulf of Aqaba were used to infer hydroclimatic changes in northern Anatolia and the northern Red Sea region during the last ~7500 years. Pronounced and coherent multicentennial variations in these records reveal patterns that strongly resemble modern temperature and rainfall anomalies related to the Arctic Oscillation/North Atlantic Oscillation (AO/NAO). These patterns suggest a prominent role of AO/NAO-like atmospheric variability during the Holocene beyond interannual to interdecadal timescales, most likely originating from solar output changes.

    Abstract Hip arthroscopy continues to increase in popularity and has an ever-expanding range of indications; however, the steep learning curve introduces significant risk of iatrogenic chondrolabral injury when accessing the joint and establishing arthroscopic portals. This article presents a technique for establishing the modified midanterior portal and is particularly useful when the available space is tight. We present "the air-lift" as a safe and simple adjunct to standard portal creation when performing hip arthroscopy in the supine position.

    Abstract Evidence from North Atlantic deep sea cores reveals that abrupt shifts punctuated what is conventionally thought to have been a relatively stable Holocene climate. During each of these episodes, cool, ice-bearing waters from north of Iceland were advected as far south as the latitude of Britain. At about the same times, the atmospheric circulation above Greenland changed abruptly. Pacings of the Holocene events and of abrupt climate shifts during the last glaciation are statistically the same; together, they make up a series of climate shifts with a cyclicity close to 1470 +/- 500 years. The Holocene events, therefore, appear to be the most recent manifestation of a pervasive millennial-scale climate cycle operating independently of the glacial-interglacial climate state. Amplification of the cycle during the last glaciation may have been linked to the North Atlantic's thermohaline circulation.

    Abstract Oxygen isotope measurements in Greenland ice demonstrate that a series of rapid warm-cold oscillations -called Dansgaard-Oeschger events- punctuated the last glaciation (Dansgard et al., 1993, doi:10.1038/364218a0). Here we present records of sea surface temperature from North Atlantic sediments spanning the past 90 kyr which contain a series of rapid temperature oscillations closely matching those in the ice-core record, confirming predictions that the ocean must bear the imprint of the Dansgaard-Oeschger events (Broecker et al., 1988, doi:10.1016/0033-5894(88)90082-8; 1990, doi:10.1029/PA005i004p00469). Moreover, we show that between 20 and 80 kyr ago, the shifts in ocean-atmosphere temperature are bundled into cooling cycles, lasting on average 10 to 15 kyr, with asymmetrical saw-tooth shapes. Each cycle culminated in an enormous discharge of icebergs into the North Atlantic (a 'Hein-rich event' (Bond et al., 1992, doi:10.1038/360245a0; Broecker et al., 1992, doi:10.1007/BF00193540), followed by an abrupt shift to a warmer climate. These cycles document a previously unrecognized link between ice sheet behaviour and ocean-atmosphere temperature changes. An important question that remains to be resolved is whether the cycles are driven by external factors, such as orbital forcing, or by inter-nal ice-sheet dynamics.

    Abstract Although still a rare bacterial infection of the endocardium of the heart, the incidence of infective endocarditis continues to increase with the increased use of intracardiac devices, indwelling lines, and surgical procedures being done on patients. The diagnosis of infective endocarditis remains challenging and requires a high level of suspicion to initiate the appropriate investigation and treatment. Serious complications can still occur despite optimal care, so it is helpful that these patients be managed by a team that includes infectious disease, cardiology, and cardiac surgeons.
