    Abstract Loss of the von Hippel-Lindau (VHL) tumor suppressor is a hallmark feature of renal clear cell carcinoma. VHL inactivation results in the constitutive activation of the hypoxia-inducible factors (HIFs) HIF-1 and HIF-2 and their downstream targets, including the proangiogenic factors VEGF and PDGF. However, antiangiogenic agents and HIF-2 inhibitors have limited efficacy in cancer therapy due to the development of resistance. Here we employed an innovative computational platform, Mining of Synthetic Lethals (MiSL), to identify synthetic lethal interactions with the loss of VHL through analysis of primary tumor genomic and transcriptomic data.     Abstract On May 21, 2019 at 03:02:29 UTC Advanced LIGO and Advanced Virgo observed a short duration gravitational-wave signal, GW190521, with a three-detector network signal-to-noise ratio of 14.7, and an estimated false-alarm rate of 1 in 4900 yr using a search sensitive to generic transients. If GW190521 is from a quasicircular binary inspiral, then the detected signal is consistent with the merger of two black holes with masses of 85_{-14}^{+21}  M_{⊙} and 66_{-18}^{+17}  M_{⊙} (90% credible intervals). We infer that the primary black hole mass lies within the gap produced by (pulsational) pair-instability supernova processes, with only a 0.32% probability of being below 65  M_{⊙}. We calculate the mass of the remnant to be 142_{-16}^{+28}  M_{⊙}, which can be considered an intermediate mass black hole (IMBH). The luminosity distance of the source is 5.3_{-2.6}^{+2.4}  Gpc, corresponding to a redshift of 0.82_{-0.34}^{+0.28}. The inferred rate of mergers similar to GW190521 is 0.13_{-0.11}^{+0.30}  Gpc^{-3} yr^{-1}.

    Abstract There is much debate within the forensic community around the indications that suggest a head injury sustained by a child resulted from abusive head trauma, rather than from accidental causes, especially when a fall from low height is the explanation given by a caregiver. To better understand this problem, finite element models of the paediatric head have been and continue to be developed. These models require material models that fit the behaviour of paediatric head tissues under dynamic loading conditions. Currently, the highest loading rate for which skull data exists is 2.81 ms

    Abstract We report the observation of a compact binary coalescence involving a 22.2 - 24.3 $M_{\odot}$ black hole and a compact object with a mass of 2.50 - 2.67 $M_{\odot}$ (all measurements quoted at the 90$\%$ credible level). The gravitational-wave signal, GW190814, was observed during LIGO's and Virgo's third observing run on August 14, 2019 at 21:10:39 UTC and has a signal-to-noise ratio of 25 in the three-detector network. The source was localized to 18.5 deg$^2$ at a distance of $241^{+41}_{-45}$ Mpc; no electromagnetic counterpart has been confirmed to date. The source has the most unequal mass ratio yet measured with gravitational waves, $0.112^{+0.008}_{-0.009}$, and its secondary component is either the lightest black hole or the heaviest neutron star ever discovered in a double compact-object system. The dimensionless spin of the primary black hole is tightly constrained to $\leq 0.07$. Tests of general relativity reveal no measurable deviations from the theory, and its prediction of higher-multipole emission is confirmed at high confidence. We estimate a merger rate density of 1-23 Gpc$^{-3}$ yr$^{-1}$ for the new class of binary coalescence sources that GW190814 represents. Astrophysical models predict that binaries with mass ratios similar to this event can form through several channels, but are unlikely to have formed in globular clusters. However, the combination of mass ratio, component masses, and the inferred merger rate for this event challenges all current models for the formation and mass distribution of compact-object binaries.

    Abstract Research consistently finds that men use all types of drugs more frequently and extensively than women. However, the misuse of prescription tranquilizers provides an exception. Recent research has found that women are more likely to misuse tranquilizers than men, yet few efforts have been made to systematically understand why this is the case and whether there are gendered factors that might help explain their misuse. Building on general strain theory and other scholarship concerning the links between psychological strain, mental–emotional health, and illicit drug use, we employed a mixed-methods design to investigate the interrelationships between gender, mental health, and tranquilizer misuse. Using data from the 2010 National Survey of Drug Use and Health, we examined tranquilizer misuse among women using various social, demographic, and health-related characteristics. Following this, we drew on nine in-depth interviews with adult women aged 21 to 69 years who reported a history of misusing tranquilizers. The quantitative data reveal that the odds of tranquilizer misuse are nearly two times higher for each unit increase on the poor mental health scale. Whereas being married increases the risk of misuse, having young children is associated with a decreased risk. Our analysis of the interview data reveals three main themes, related to tranquilizer access, reasons for misuse, and shame related to misuse. The interviews clearly uncover tranquilizer misuse as an attempt by women to manage competing demands between their work and home lives, and more specifically as a means of promoting success in both devotions. We conclude by arguing that women’s misuse of tranquilizers is a gendered behavior in response to gender-specific strains, which in turn reproduces gendering as an institution as well as in individual lives. The implications of these findings for general strain theory are also discussed.

    Abstract Mediterranean mussels (Mytilus galloprovincialis) were exposed over 21 days to polyethylene (PE) particles (0.01 mg ml−1; 50–570 μm) isolated from toothpaste. PE was deployed in the Outer Oslofjord (Norway) for 21 days, before exposing the mussels to both virgin (PE-V) and weathered PE (PE-W) particles. The mussels ingested both types of particles, but significantly more weathered particles were ingested than virgin (p = .0317), based on PE dosed by weight (mg ml−1) but not when considering particle number (PE-V: 1.18 ± 0.16 particles ml−1; PE-W 1.86 ± 0.66 particles ml-1;). PE particle ingestion resulted in structural changes to the gills and digestive gland, as well as necrosis in other tissues such as the mantle. No differences were found regarding the degree of tissue alteration between PE-virgin and PE-weathered exposures. This current study illustrates the importance of using weathered particles in microplastic exposure studies to reflect the behaviour of plastic particles after entering the marine environment. The observed tissue alterations demonstrate the potential adverse effects to mussels exposed to microplastic particles.
