    Abstract Objectives: The goal of this investigation was to identify the main compounds and the pharmacological mechanism of the traditional Chinese medicine formulation, Gong Ying San (GYS), by infrared spectral absorption characteristics, metabolomics, network pharmacology, and molecular-docking analysis for mastitis. The antibacterial and antioxidant activities were determined in vitro.
    Methods: The chemical constituents of GYS were detected by ultra-high-performance liquid chromatography Q-extractive mass spectrometry (UHPLC-QE-MS). Related compounds were screened from the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP, http://tcmspw.com/tcmsp.php) and the Encyclopedia of Traditional Chinese Medicine (ETCM, http://www.tcmip.cn/ETCM/index.php/Home/) databases; genes associated with mastitis were identified in DisGENT. A protein-protein interaction (PPI) network was generated using STRING. Gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment screening was conducted using the R module. Molecular-docking analyses were performed with the AutoDockTools V1.5.6.
    Results: Fifty-four possible compounds in GYS with forty likely targets were found. The compound-target-network analysis showed that five of the ingredients, quercetin, luteolin, kaempferol, beta-sitosterol, and stigmasterol, had degree values >41.6, and the genes TNF, IL-6, IL-1β, ICAM1, CXCL8, CRP, IFNG, TP53, IL-2, and TGFB1 were core targets in the network. Enrichment analysis revealed that pathways associated with cancer, lipids, atherosclerosis, and PI3K-Akt signaling pathways may be critical in the pharmacology network. Molecular-docking data supported the hypothesis that quercetin and luteolin interacted well with TNF-α and IL-6.
    Conclusions: An integrative investigation based on a bioinformatics-network topology provided new insights into the synergistic, multicomponent mechanisms of GYS's anti-inflammatory, antibacterial, and antioxidant activities. It revealed novel possibilities for developing new combination medications for reducing mastitis and its complications.

    Abstract Objective: Diabetic retinopathy (DR) is a severe diabetic complication that leads to severe visual impairment or blindness. He-Ying-Qing-Re formula (HF), a traditional Chinese medicinal concoction, has been identified as an efficient therapy for DR with retinal vascular dysfunction for decades and has been experimentally reported to ameliorate retinal conditions in diabetic mice. This study endeavors to explore the therapeutic potential of HF with key ingredients in DR and its underlying novel mechanisms.
    Methods: Co-expression gene modules and hub genes were calculated by weighted gene co-expression network analysis (WGCNA) based on transcriptome sequencing data from high-glucose-treated adult retinal pigment epithelial cell line-19 (ARPE-19). The chromatographic fingerprint of HF was established by ultra-performance liquid chromatography coupled with high-resolution mass spectrometry (UPLC-Q-TOF-MS). The molecular affinity of the herbal compound was measured by molecular docking. Reactive oxygen species (ROS) was measured by a DCFDA/H2DCFDA assay. Apoptosis was detected using the TUNEL Assay Kit, while ELISA, Western blot, and real-time quantitative reverse transcription polymerase chain reaction (qRT-PCR) were used for detecting the cytokine, protein, and mRNA expressions, respectively.
    Results: Key compounds in HF were identified as luteolin, paeoniflorin, and nobiletin. For WGCNA, ME-salmon ("protein deacetylation") was negatively correlated with ME-purple ("oxidative impairment") in high-glucose-treated ARPE-19. Luteolin has a high affinity for SIRT1 and P53, as indicated by molecular docking. Luteolin has a hypoglycemic effect on type I diabetic mice. Moreover, HF and luteolin suppress oxidative stress production (ROS and MDA), inflammatory factor expression (IL-6, TNF-α, IL1-β, and MCP-1), and apoptosis, as shown in the
    Conclusion: Using HF and its active compound luteolin as therapeutic agents offers a promising approach to diabetic retinopathy treatment. It primarily suppressed protein acetylation and oxidative stress via the SIRT1/P53 pathway in retinal pigment epithelial cells.

    Abstract Two quality control standards, total flavonoid glycosides of Epimedii Folium and epimedin C of Epimedii Wushanensis Folium, were used to systematically evaluate the quality of Epimedium wushanense T. S. Ying, so as to provide reference for its germplasm screening and resource utilization. Seven representative populations of E. wushanense covering its main distribution areas were uniformly sampled during the flowering period. There were significant quality differences among the populations of E. wushanense. According to the quality standard of total flavonoid glycosides, all populations were superior to the quality standard of the Chinese Pharmacopoeia for Epimedii Folium, with more than 1.5 % total flavonoid glycosides. The variation ranges of epimedin A, epimedin B, epimedin C, icariin and total flavonoid glycosides were 0.40-0.76 %, 0.51-0.83 %, 1.70-9.31 %, 0.40-1.23 % and 3.05-10.61 %, respectively. According to the quality standard of epimedin C, all populations were better than the quality standard of the Chinese Pharmacopoeia for Epimedii Wushanensis Folium, with more than 1.0 % epimedin C. The variation range of epimedin C was 2.22-10.06 %. When comparing the results of the two methods, a trend of slightly lower mean values was found for total flavonoid glycosides, except for the HBXW population. The quality of E. wushanense was superior to both the quality standard of Epimedii Folium and Epimedii Wushanense Folium in the Chinese Pharmacopoeia. Epimedin C was the most abundant component. Among the investigated populations, HBXW and HBGK exhibited the highest quality, and may provide excellent genetic resources for standardized cultivation. In addition, the habitat of these populations can also serve a reference for cultivation conditions.

    Abstract Objective: To establish a gas chromatography-mass spectrometry(GC-MS) method for determination of seven sugars and sugar alcohols in infants Ying Yang Bao nutritional supplements.
    Methods: The samples were extracted with pure water and diluted with 95% ethanol. After being dried by nitrogen, methoxyamine hydrochloride oxime was dissolved in pyridine and derivatized by MSTFA. The capillary column TG-5 Ms(30 m×0. 25 mm, 0. 25 μm) was used for determination by GC-MS.
    Results: The limits of detection(LODs)were 1. 0-3. 0 mg/g and the limits of quantification(LOQs)were 3. 3-10. 0 mg/g. The average recoveries of seven kinds of sugar and sugar alcohols were 86. 7%-96. 7%, and the relative standard deviation was less than 5. 1%(n=6). The contents of seven sugars and sugar alcohols in soybean matrix nutritional supplements were determined in the range of 0. 25-13. 70 g/100 g, which was consistent with the nutrition label of the products.
    Conclusion: The method is convenient, mild and fit for batch sample analysis.

    Abstract Sinopodophyllum hexandrum (Royle) Ying is a perennial medicinal plant that produces the anti-cancer lignan compositions especially podophyllotoxin. Since podophyllotoxin commercial production via chemical synthesis, biotechnological intervention or cultivation has yet to meet the increasing demand, mining the potentially available plant organs to discover more natural podophyllotoxin sources is essential. This study investigated the reversed-phase high performance liquid chromatography (RP-HPLC) fingerprint, active chemicals (lignans, flavonoids, and phenolic profiles), and bioactivities (antimicrobial and cytotoxic activities) of distinct S. hexandrum plant organs. Among different plant organs, the rhizome is a preponderant organ due to high active compounds especially podophyllotoxin at 71.615☓10³ μg/mL, high bioactivity with low minimum inhibitory concentration (MIC)/minimal bactericidal concentration (MBC)/minimal fungicidal concentration (MFC) value of 5/5/5 mg/mL, and low CIR₅₀ value of 0.0718 μg/mL, followed by root. Chemometrics analysis techniques, including correlation analysis (CA), similarity analysis (SA), hierarchical cluster analysis (HCA), principal component analysis (PCA), and discriminant analysis (DA), were further employed to facilitate accurate quality assessment of different plant organs from S. hexandrum. The findings unambiguously validated the clustering of seven samples into four major groups (G1–G4), with G1 (rhizome)>G2 (root)>G4 (fruit, blossom, and leaf)>G3 (stem and petiole) as the consistent quality ordering, despite significant differences in chemical compositions and bioactivities. Considered the practical application, the rhizome is, therefore, a preferred medicinal organ, and root could be considered as a substitute resource. The rhizome or root extraction could be used as a potential raw material source for natural antimicrobial agents and cancer cell inhibitors in the food, medicine, and healthcare industries. This research added to our understanding of the integrated exploitation and utilization extension of S. hexandrum resources. RP-HPLC fingerprinting in combination with chemometrics approaches was also confirmed as a highly reliable method for assessing the quality of S. hexandrum and other industrial crops.

    Abstract Objective: To compare the therapeutic effect between combination of Shu-stream and Ying-spring acupoints and routine acupoints selected by syndrome differentiation for toothache.
    Methods: Eighty-five cases were randomly divided into a combination of Shu-stream and Ying-spring acupoints group (group A, 43 cases) and a routine acupionts group (group B, 42 cases). In group A, according to the corresponding relationship between viscera-meridians and different kinds of teeth, the Shu-stream and Ying-spring acupoints at both sides of the corresponding meridians were sleeted. The group B was treated with acupuncture at Jiache (ST 6), Xiaguan (ST 7), Hegu (LI 4), erjian (LI 2), Neiting (ST 44). Yifeng (TE 17) and Fengchi (GB 20) were added for wind and fire attacking; Lidui (ST 45) and Quchi (LI 11) were added for intense stomach fire; Taixi (KI 3) and Zhaohai (KI 6) were added for deficiency fire flaming up. Electroacupuncture was applied in two groups, once every day, ten treatments constituted one course, the differences of effect and recurrence rate were observed after one course and half a year later.
    Results: The cured rate were respectively 88.4% (38/43)and 69.0% (29/42) in group A and group B, with a significant difference (P < 0.05). The recurrence rates were respectively 23.7% (9/38) and 55.2% (16/29) in group A and group B, with a significant difference (P < 0.05).
    Conclusion: Both short-term and long-term therapeutic effects of combination of Shu-stream and Ying-spring acupoints are significantly better than those of routine acupoints in treating toothache.
