    Abstract Myostatin (MSTN) is a member of the transforming growth factor-β (TGF-β) family, and functions as an inhibitor of muscle growth. Disrupting the inhibitory effect of MSTN on growth can provide an effective way to increase the muscle yield of livestock and poultry. The cysteine knot motif of TGF-β can stabilize the structure of MSTN protein and plays an important regulatory role in the biological function of MSTN. Accordingly, in this study, we used the CRISRP/Cas9 to edit the exon 3 of MSTN in the kidney cells of Liang Guang Small Spotted pig (LPKCs), in order to disrupt the cysteine knot motif of MSTN and remove the inhibitory effect of MSTN on its target genes.MSTN-edited LPKCs were obtained through fluorescence-activated cell sorting (FACS) and used as donor cells for somatic cell nuclear transfer (SCNT) to generate cloned embryos, which were then transferred to surrogate sows to finally obtain eight MSTN-edited Liang Guang Small Spotted piglets. Among them, two survived to 10 days old. Genotyping revealed that these two piglets were gene edited heterozygotes with base deletion and substitution occurred within the coding sequence of C106 and C108 at the cystine knot motif of MSTN. These changes resulted in frameshift mutations, and conversion of C106 and C108 to other amino acids. More developments of muscles were observed at the shoulders and hips of the heterozygotes of MSTN-edited Liang Guang Small Spotted pigs. H&E analysis showed that the cross-sectional area (CSA) of myofiber inMSTN-edited pigs was significantly decreased, and the number of myofiber were significantly increased. Western blot analysis showed that the disruption of C106 and C108 did not affect the expression of MSTN protein, but significantly up-regulated the expression of its target genes such as Myf5, MyoD, Myogenin and other myogenic regulatory factors. In summary, the gene-edited pig model obtained in this study did not cause complete loss of MSTN expression, and could retain other biological functions of MSTN, thereby promoting muscle growth while minimizing the potential adverse effects on complete loss of MSTN in the Liang Guang Small Spotted pigs.

    Abstract Background: Qing Guang An Granule (QGAG), a Chinese patent medicine, has been used clinically to treat glaucoma for more than 20 years.
    Objective: To explore the possible mechanism of treatment of QGAG in glaucoma by using network pharmacology and molecular docking in this study.
    Methods: Active compounds and targets of each herb in QGAG were retrieved via the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP). Glaucoma-related targets were acquired from OMIM and DisGeNET database. Key targets of QGAG against glaucoma were acquired by overlapping the above targets via the Venn diagram. Using the DAVID, Gene Ontology (GO) enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis of the key targets were performed. The docking process was performed using the AutoDock 4.2.6 and AutoDock Vina 1.1.2.
    Results: The 55 active compounds and 173 targets were obtained and constructed a compound-target network. The 20 key targets of QGAG in treating glaucoma were acquired, and these targets are involved in the apoptotic process, cellular response to hypoxia, negative regulation of cell growth, and ovarian follicle development. The main pathways are p53, HIF-1, PI3K-Akt, and neurotrophin signaling pathway.
    Conclusion: QGAG may exert a protective effect by acting on the optic nerve at a molecular and systemic level. This study can provide a certain basis for future researches on exploring the QGAG in treating glaucoma and provide new ideas for developing new drugs.

    Abstract AIM: To observe effects of four effective components of Qing Guang An on inoblast and typeⅠcollagen of scarring tissue of filtration canal after glaucoma surgery, and discuss their effect and function mechanism on preventing its formation. METHODS: Four effective components of Qing Guang An were used in rabbit eyes after glaucoma surgery(Group D.E.F.G), with the blank group(A), model group(B), MMC group comparison(C). Effects of four effective components of Qing Guang An on inoblast and typeⅠcollagen of scarring tissue of filtration canal after glaucoma surgery were observed. RESULTS: After surgery, MMC group(C), effective component 2(E)groups of intraocular pressure rose slowly. At the fourth week, intraocular pressure remained minimum. The compare of these two groups of intraocular pressure and those of group A, B, D, F, G had significance on statistics. Comparing with the number of inoblast of each group, except group D and E, which result was P>0.05 and had no obvious significance on statistics, others were significant. Comparing with the expression of typeⅠcollagen, except group C and E, B and G, F and G, result of others had obvious significance on statistics showing as P<0.05. CONCLUSION: The scaring of filter canal after glaucoma surgery is main cause of the filtration surgery's failure assuming abnormal backup intraocular pressure. The effective componment 2 of Qing Guang An and MMC can obviously inhibit the multiplication of inoblast and the expression of typeⅠcollagen, reduce the hyperplasia of scar tissue, which has obvious effect for preventing scarring of filtration canal.
