    Abstract Confined field trials (CFT) of genetically engineered (GE) crops are used to generate data to inform environmental risk assessments (ERA). ERAs are required by regulatory authorities before novel GE crops can be released for cultivation. The transportability of CFT data to inform risk assessment in countries other than those where the CFT was conducted has been discussed previously in an analysis showing that the primary difference between CFT locations potentially impacting trial outcomes is the physical environment, particularly the agroclimate. This means that data from trials carried out in similar agroclimates could be considered relevant and sufficient to satisfy regulatory requirements for CFT data, irrespective of the country where the CFTs are conducted. This paper describes the development of an open-source tool to assist in determining the transportability of CFT data. This tool provides agroclimate together with overall crop production information to assist regulators and applicants in making informed choices on whether data from previous CFTs can inform an environmental risk assessment in a new country, as well as help developers determine optimal locations for planning future CFTs. The GEnZ Explorer is a freely available, thoroughly documented, and open-source tool that allows users to identify the agroclimate zones that are relevant for the production of 21 major crops and crop categories or to determine the agroclimatic zone at a specific location. This tool will help provide additional scientific justification for CFT data transportability, along with spatial visualization, to help ensure regulatory transparency.

    Abstract Rationale: Mycobacterium avium
    Objectives: To develop and optimize a CRISPR-MAC assay for MAC infection detection and to evaluate its diagnostic and prognostic performance in two MAC disease cohorts.
    Methods: MAC-cfDNA serum levels were measured in individuals diagnosed with MAC disease or who had bronchiectasis or COPD diagnoses without a history of NTM-PD or NTM-positive sputum cultures. Diagnostic performance was analyzed with pre-treatment serum from two cohorts. Serum MAC-cfDNA changes during MAC-PD treatment were evaluated in a subset of MAC-PD patients who received macrolide-based multi-drug regimens.
    Measurements and main results: CRISPR-MAC assay detected MAC-cfDNA in MAC-PD with 97.6% (91.6 - 99.7%) sensitivity and 97.6% (91.5 - 99.7%) specificity overall. Serum MAC-cfDNA levels markedly decreased after MAC-directed treatment initiation in MAC-PD patients that demonstrated MAC culture conversion.
    Conclusions: This study provides preliminary evidence for the utility of a serum-based CRISPR-MAC assay to rapidly detect MAC infection and monitor their response to treatment.

    Abstract Wildfire regimes are being altered in ecosystems worldwide. The density of reptiles responds to fires and changes to habitat structure. Some of the most vulnerable ecosystems to human-increased fire frequency are old-growth Araucaria araucana forests of the southern Andes. We investigated the effects of wildfires on the density and richness of a lizard community in these ecosystems, considering fire frequency and elapsed time since last fire. During the 2018/2019 southern summer season, we conducted 71 distance sampling transects to detect lizards in Araucaria forests of Chile in four fire "treatments": (1) unburned control, (2) long-term recovery, (3) short-term recovery, and (4) burned twice. We detected 713 lizards from 7 species. We found that the density and richness of lizards are impacted by wildfire frequency and time of recovery, mediated by the modification of habitat structure. The lizard community varied from a dominant arboreal species (L. pictus) in unburned and long-recovered stands, to a combination of ground-dwelling species (L. lemniscatus and L. araucaniensis) in areas affected by two fires. Araucaria forests provided key habitat features to forest reptiles after fires, but the persistence of these old-growth forests and associated biodiversity may be threatened given the increase in fire frequency.
