    Abstract Anhand des Werks des österreichischen Dichters Thomas Bernhard (1931 bis 1989) wird der Frage nachgegangen, wie die Psychoanalyse als Methode zur Interpretation und zum Erkenntnisgewinn der literarischen Kunst angewandt und sinnvoll genutzt werden? Die Beziehung zwischen Psychoanalyse und Literatur wird reflektiert, erforscht und diskutiert, und auf dieser Grundlage werden Leben und Werk Thomas Bernhards untersucht. Dabei werden zwei Schwerpunkte verfolgt: zum einen die kritische, theoretische Auseinandersetzung mit bisherigen Leistungen der psychoanalytischen Literaturwissenschaft, zum anderen eine direkte, praxisbezogene Anwendung und Überprüfung der Ergebnisse dieser Auseinandersetzung am Gegenstand der Literatur selbst. Der erste Band enthält die komplexe Entwicklung der Beziehung zwischen Psychoanalyse und Kunst, setzt diese in einen synoptischen Zusammenhang und diskutiert psychoanalytische Bedingungen künstlerischer Kreativität und Produktivität. Im zweiten Band werden Rezeptionsprozesse untersucht und im Zuge einer Interpretation von Thomas Bernhards Werk der Einfluss von Biografischem in Beziehung mit Fragen zur Ästhetik und der literarischen Form gerückt. - Inhalt: (I) Geschichte und Produktion.     Abstract Background and purpose: To evaluate the feasibility of a multimodal approach involving dynamic contrast-enhanced (DCE) perfusion imaging and diffusion kurtosis imaging (DKI) in the preoperative imaging of brain tumors in a multicenter setting, and to evaluate the effect on diagnostic confidence and accuracy for tumor grade and type prediction.
    Materials and methods: One hundred and thirty-three patients with brain tumors were imaged in six hospitals with a standardized multimodal protocol. Standard imaging and six parameter maps derived from DCE and DKI sequences were reviewed off-site by two independent readers. Image quality and diagnostic confidence were evaluated in qualitative analyses. Quantitative analyses were performed to assess diagnostic accuracy and the performance of DKI and DCE parameters for tumor grade differentiation and molecular tumor type determination.
    Results: Standardized acquisition of DCE and DKI maps was feasible with excellent image quality. Diagnostic confidence was significantly improved from 85 % to 96 % (p = 0.0005) by additional review of the DCE and DKI maps. The combination of mean kurtosis and CBV was particularly advantageous for differentiating low-grade and high-grade glioma, oligodendroglial vs. astrocytic, and IDH1/2 wild type vs. mutated tumors.
    Conclusion: A multimodal imaging approach with DCE and DKI improves diagnostic confidence and yields higher diagnostic accuracy for predicting tumor grade and type in adult-type glioma.

    Abstract Tn7-like transposons have co-opted CRISPR-Cas systems to facilitate the movement of their own DNA. These CRISPR-associated transposons (CASTs) are promising tools for programmable gene knockin. A key feature of CASTs is their ability to recruit Tn7-like transposons to nuclease-deficient CRISPR effectors. However, how Tn7-like transposons are recruited by diverse CRISPR effectors remains poorly understood. Here, we present the cryo-EM structure of a recruitment complex comprising the Cascade complex, TniQ, TnsC, and the target DNA in the type I-B CAST from Peltigera membranacea cyanobiont 210A. Target DNA recognition by Cascade induces conformational changes in Cas6 and primes TniQ recruitment through its C-terminal domain. The N-terminal domain of TniQ is bound to the seam region of the TnsC spiral heptamer. Our findings provide insights into the diverse mechanisms for the recruitment of Tn7-like transposons to CRISPR effectors and will aid in the development of CASTs as gene knockin tools.

    Abstract The conversion of phosphatidylinositol 4,5-bisphosphate to phosphatidylinositol 3,4,5-triphosphate by phosphoinositide 3-kinase γ (PI3Kγ) is critical for neutrophil chemotaxis and cancer metastasis. PI3Kγ is activated by Gβγ heterodimers released from G protein-coupled receptors responding to extracellular signals. Here we determined cryo-electron microscopy structures of Sus scrofa PI3Kγ-human Gβγ complexes in the presence of substrates/analogs, revealing two Gβγ binding sites: one on the p110γ helical domain and another on the p101 C-terminal domain. Comparison with PI3Kγ alone reveals conformational changes in the kinase domain upon Gβγ binding that are similar to Ras·GTP-induced changes. Assays of variants perturbing the Gβγ binding sites and interdomain contacts altered by Gβγ binding suggest that Gβγ recruits the enzyme to membranes and allosterically regulates activity via both sites. Studies of zebrafish neutrophil migration align with these findings, paving the way for in-depth investigation of Gβγ-mediated activation mechanisms in this enzyme family and drug development for PI3Kγ.
