    Abstract To address the challenges faced by forensic examiners in degraded DNA analysis, we have developed two different panels for various forensic applications, encompassing an AIM-InDel panel for ancestry inference and a Multi-InDel panel for individual identification, respectively. Herein, the efficiencies of these two panels were tested in the Chinese Hui group. By calculating forensic parameters and simulating family relationships, we verified that the Multi-InDel panel could be an effective tool for individual identification, paternity testing, and could assist in kinship identification in the Hui group.     Abstract Background: Global nonvalvular AF rises, impacting health severely. In Qinghai, China's diverse setting, studying AF among varied ethnic groups is crucial OBJECTIVES: The purpose of this study was to compares cardiac features in AF among Tibetan, Han, and Hui patients to develop tailored prevention and treatment strategies for this region, the goal was to enhance the understanding of AF and provide an empirical basis for developing prevention and treatment strategies specific to this region METHODS: This study included a total of 3445 Tibetan, Han, and Hui patients diagnosed with nonvalvular atrial fibrillation and treated at the Qinghai Cardiovascular and Cerebrovascular Specialist Hospital, China, between January 2019 and January 2021. We analyzed the differences in cardiac structure, comorbidities, and other influencing factors among the different ethnic groups RESULTS: We found significant differences in gender, age, smoking history, lone atrial fibrillation, left heart failure, dilated cardiomyopathy, and diabetes between Tibetan, Han, and Hui patients (P < 0.05). Tibetan, Han, and Hui patients also differed with regard to left ventricular end-diastolic volume, left ventricular ejection fraction, fractional shortening, NT-proBNP, glycated hemoglobin, red blood cell distribution width, platelet count, platelet hematocrit, platelet distribution width, homocysteine (Hcy), C-reactive protein, and superoxide dismutase (SOD) (P < 0.05) CONCLUSION: Our study revealed variations in comorbidities, cardiac structure, and blood indexes among Tibetan, Han, and Hui AF patients, highlighting distinct patterns in complications and biomarker levels across ethnic groups.

    Abstract Background: Alzheimer's disease (AD) has gradually increased as society has aged and is now a serious social problem. In the clinical treatment of AD, patients show improvement in cognitive function after treatment with the traditional Chinese medicine compound, Yizhi Sheng Hui (YZSH) decoction. This study systematically investigates the effect and pharmacology of the YZSH decoction on AD.
    Methods: In this study, 24 SAMP8 AD model mice were randomly divided into three groups: an untreated control group, a group treated with the YZSH decoction, and a positive control group treated with donepezil hydrochloride. Eight SAMR1 mice were placed in the normal control group. A Morris water maze test and a step-down test were conducted at 8 and 13 weeks after continuous intragastric administration of the two drugs. After 13 weeks of administration, the hippocampal expression of Aβ1-42 and tau protein were measured.
    Results: There was no change in the latent period duration and the number of platform crossings in each group after 8 weeks of administration, but after 13 weeks of administration, the latent period of the treatment group and the positive control group were significantly shorter than the untreated control group. Initially, the SAMP8 mice showed a lower spatial exploration ability than the SAMR1 mice. However, after 13 weeks of administration, the treatment group and the control group exhibited a better exploration ability. Compared with the SAMR1 mice, the on-stage evasion time and step-down errors significantly increased in the untreated group. Compared with the untreated group, mice in the treatment group and the positive control group showed a shorter latent period after 8 weeks of administration, and the on-stage evasion time for both groups was significantly reduced after 13 weeks of administration. The treatment group showed fewer instances of electric shock. Hippocampal expression of Aβ1-42 was high in the untreated group, was much lower in the positive control group, and no Aβ1-42 expression was observed in the treatment group.
    Conclusions: The YZSH decoction improved the learning and memory of mice with AD, related to the inhibition of Aβ1-42 expression.

    Abstract With the development of cytology, the establishment of cell models in vitro has become a powerful means to study the mechanism and treatment of diseases. Here we successfully generated the IPSC-derived modeling system of a 25-year-old healthy male. His peripheral blood mononuclear cells (PBMC) were reprogrammed using human OKSM (SOX2, OCT3/4, KLF4, and C-MYC) transcription factors using a non-integrated additional vector system. Immunocytochemistry demonstrated that IPSCS expressed all the markers of pluripotency and demonstrated their ability to differentiate spontaneously from three hypoderms in vitro. Karyotype is normal.

    Abstract Transferring the genome of distant species to crops is an efficient way to create new germplasms. However, the molecular mechanisms involved are unclear. In this study, a new rice restorer line R21 with heat tolerance was created by introgressing the genomic DNA of sorghum into the recipient restorer line Jin Hui 1. Assembly of rice R21 and Jin Hui 1 genomes was performed using PacBio sequencing technology. Comparative genome analysis and coverage statistics showed that the repetitive sequence atr0026 was a candidate introgression fragment of sorghum DNA. Sequence similarity analysis revealed that atr0026 was distributed at different copy numbers on the telomeric position of chromosomes 9 or 10 in R21, Jin Hui 1, and several rice varieties, indicating that the repetitive sequence from sorghum was highly conserved in rice. The repeat annotation in Gramineae indicated that ribosomal DNA loci that existed in atr0026 may be cause a rearrangement of chromosomes 9 and 10 of the R21 genome, resulting in a copy number variation at the 5' end of it. Our study lays the foundation for further elucidation of the molecular mechanisms underlying the heat tolerance of sorghum DNA introgression variant line R21, which is of great significance for guiding crop genetic breeding.

    Abstract Important for promoting the integrated protection and systematic management of mountains, rivers, forests, farmlands, lakes, grasslands, and sandy areas, ecological networks form the backbone of the regional ecological security pattern. An improved morphological spatial pattern analysis coupled with a minimum cumulative resistance model (MSPA–MCR) based on multi-source data was used to study, on a provincial scale, the ecological security pattern of Ningxia, an ecologically fragile region in the upper reaches of the Yellow River in China. The results show the following: (1) A reasonable classification of ecological sources and ecological corridors is key to constructing hierarchical ecological networks. Classifying ecological sources by replacing patch areas with energy factors and identifying the importance of ecological corridors by modifying the gravity model with the energy factors proposed in this paper could improve the rationality of the hierarchical structure division of ecological networks. (2) Grassland as the substrate vegetation type is an important ecological source type in arid and semi-arid ecologically fragile areas, and forests and lake wetlands are the main ecological source types in mountainous areas and oasis areas, respectively. The study area was located in the arid–semi-arid transitional area, with a variety of ecological types, such as mountain, oasis, and desert. Therefore, the complex ecological source types of forest–grassland–wetland appear in some areas. (3) There are 45 ecological patch groups that can be classified as ecological sources in Ningxia, including 10 primary source groups. The number of primary source groups is small, and their spatial distribution is unbalanced. There are two categories of ecological corridors, the river corridor and the mountain corridor, and the network connectivity is poor. (4) The ecological network structure of Ningxia is presented as an ecological security structure consisting of one belt, three screens, three corridors, and five clusters, forming a hierarchical nested ecological network security structure system.

    Abstract Since the implementation of the grassland ecological protection policy of prohibition grazing on natural grasslands throughout the territory in 2003, the growth of grasslands in Ningxia has improved. This study investigated the spatial differentiation mechanism of normalized vegetation index (NDVI) in Ningxia grasslands from 1988 to 2018, analyzed the relative contributions of climate change (CC) and human activities (HA) to NDVI changes, and predicted the future trend of grassland changes. The results show that except in winter, the annual, seasonal and monthly average values of NDVI after grazing prohibition were higher than those before grazing prohibition. After grazing prohibition, the growth rate decreased by 17.91%, but the degradation rate increased by 3.92%. After grazing prohibition, the proportion of medium coverage increased by 16.15%, mainly in the path of “lower coverage grassland→medium coverage grassland”. The transformation trend was mainly positive, and the ecological construction project has achieved remarkable results. The main factors affecting NDVI differentiation in Ningxia grassland were snow depth, potential evapotranspiration, radiation, and precipitation. After grazing prohibition, the explanatory power of each factor and the interaction between the factors decreased significantly, but the explanatory power of wind speed was greatly improved. After the grazing prohibition, 53.22% of the total area was affected by human activities and climate change. The relative contribution of human activities decreased in NDVI-increased areas but increased in NDVI-decreased areas.
