    Abstract The in vitro production of stem-cell-derived islets (SC-islets) has brought forth the potential of transplanting these cells to restore glycemic control in people with diabetes. Nonetheless, alloimmune and autoimmune responses remain considerable challenges for a broad clinical implementation of β-cell replacement therapies. β-cell stress has been implicated in the onset of β-cell immunogenicity and death and is likely to contribute to β-cell failure following transplantation. We show that inducing stress and/or administering cytokines causes SC-islet apoptosis, cellular dysfunction, and an increased expression of β-cell stress- and immune-interaction-related genes. We then demonstrate that manipulating some of these genes results in enhanced protection of SC-islets from apoptosis in vitro.

    Abstract Tension-type-headaches appear as the third most common disease in the general population and have a big impact on quality of life. The aim is to verify the impact of clinical Pilates exercises on pain intensity, impact headaches, neck disability, state of anxiety, depression, stress and quality of sleep in higher-education students. After a sample selection questionnaire was completed, a series of cases was carried out with 9 undergraduate students with tension-type headaches. Five instruments were applied before and after an intervention program: (i) Numerical Rating Scale-NRS, (ii) Headache Impact Test-HIT-6™, (iii) Neck Disability Index, (iv) Depression, Anxiety, Stress Scales and (v) Pittsburgh Sleep Quality Index. Four sessions of 30 min classes of Pilates exercises were held for 2 weeks. The pain intensity decreased in only 2 participants. The impact of tension-type headaches on normal daily life and ability to function was improved in all patients, and only one patient's functional disability caused by pain in the cervical region did not improve. In regard to negative emotional states, 6 individuals reported improvements, and only one individual reported a lower quality of sleep after the program. The exercise program can induce positive effects on disorders associated with tension-type-headaches.

    Abstract Background: Interleukin (IL)-10 knockout (KO) mice, a model for inflammatory bowel disease (IBD), develop chronic enterocolitis due to an aberrant immune response to enteric antigens. Endoscopy, the gold standard for evaluation of human mucosal health, is not widely available for murine models.
    Aims: To assess the natural history of left-sided colitis in IL-10 KO mice via serial endoscopies.
    Methods: BALB/cJ IL-10 KO mice underwent regular endoscopic assessments from 2 up to 8 months of age. Procedures were recorded and blindly evaluated using a 4-component endoscopic score: mucosal wall transparency, intestinal bleeding, focal lesions and perianal lesions (0-3 points each). An endoscopic score ≥ 1 point was considered as the presence of colitis/flare.
    Results: IL-10 KO mice (N = 40, 9 female) were assessed. Mean age at first endoscopy was 62.5 ± 2.5 days; average number of procedures per mouse was 6.0 ± 1.3. A total of 238 endoscopies were conducted every 24.8 ± 8.3 days, corresponding to 124.1 ± 45.2 days of surveillance per mouse. Thirty-three endoscopies in 24 mice (60%) detected colitis, mean endoscopy score 2.5 ± 1.3 (range: 1-6.3). Nineteen mice (47.5%) had one episode of colitis and 5 (12.5%) had 2-3 episodes. All exhibited complete spontaneous healing on subsequent endoscopies.
    Conclusions: In this large-scale endoscopic surveillance study of IL-10 KO mice, 40% of mice did not develop endoscopic left-sided colitis. Furthermore, IL-10 KO mice did not exhibit persistent colitis and universally exhibited complete spontaneous healing without treatment. The natural history of colitis in IL-10 KO mice may not be comparable with that of IBD in humans and requires careful consideration.

    Abstract Este artigo buscou analisar a importância dos programas sociais na mitigação da insegurança alimentar durante a pandemia da COVID-19, considerando os domicílios mais vulneráveis de Ponta Grossa-PR. Para isso, foi utilizado informações de uma pesquisa realizada por Raiher et al (2022), construída a partir de uma amostra probabilística de 302 famílias, as quais representaram a população total atendida com o benefício eventual de auxílio alimentação de Ponta Grossa-PR (total de 5.305 famílias). Com essas informações, utilizou o método Propensity Score Matching visando mensurar o efeito dos programas sociais na insegurança alimentar. Como corolário, teve-se um agravamento da fome no município de Ponta Grossa-PR durante a pandemia, de modo que quase metade da população estava em grave vulnerável alimentar. E os programas sociais se apresentaram essenciais para o rompimento da insegurança alimentar destes domicílios, apresentando um efeito significativo. Abstract : This article sought to analyze the importance of social programs in mitigating food insecurity during the COVID-19 pandemic, considering the most vulnerable households in Ponta Grossa-PR. For this, information from a survey carried out by Raiher et al (2022) was used, constructed from a probabilistic sample of 302 families, which represented the total population served with the occasional benefit of food aid in Ponta Grossa-PR (total of 5,305 families). With this information, he used the Propensity Score Matching method to measure the effect of social programs on food insecurity. As a corollary, there was a worsening of hunger in the municipality of Ponta Grossa-PR during the pandemic, so that almost half of the population was seriously vulnerable to food. And the social programs were essential for breaking the food insecurity of these households, with a significant effect.

    Abstract Viral respiratory infections represent a major threat to the population's health globally. Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) causes COVID-19 disease and in some cases the symptoms can be confused with Influenza disease caused by the Influenza A viruses. A simple, fast, and selective assay capable of identifying the etiological agent and differentiating the diseases is essential to provide the correct clinical management to the patient. Herein, we described the development of a genomagnetic assay for the selective capture of viral RNA from SARS-CoV-2 and Influenza A viruses in saliva samples and employing a simple disposable electrochemical device for gene detection and quantification.
