Abstract Polyester plastics are widely used in daily life, but also cause a large amount of waste. Degradation by microbial enzymes is the most promising way for the biobased upcycling of the wastes. However, there is still a shortage of high-performance enzymes, and more efficient polyester hydrolases need to be developed. Here we identified two polyester hydrolases, jmPE13 and jmPE14, from a previously isolated strain

Abstract [Research Purpose] In the era of the digital economy, there is an urgent need to explore solutions to various problems faced by enterprises in their digital transformation, such as the lack of data resources, data silos, and information asymmetry within supply chains. [Method/Contribution] Leveraging evolutionary game theory and adopting a supply chain perspective, this study integrates the government and upstream/downstream enterprises into a unified analysis framework. In this study, a three-party evolutionary game model under government coordination aimed at fostering data openness and sharing among supply chain enterprises is constructed. Simulation analyses are conducted on decision-making strategies concerning data sharing between the government and supply chain enterprises across different scenarios. [Research Conclusion] It is observed that the high level of benefits and low costs associated with data sharing incentivize supply chain enterprises to actively open and share their data. Notably, government incentives significantly encourage data openness among these enterprises by subsidizing the cost of data sharing, "especially evident when the incentive coefficient exceeds 0.6," thereby guiding them toward collaborative data-sharing initiatives. Finally, it is also found that data sharing further promotes the digital transformation of the supply chain, optimizing decision-making processes, resource allocation, and operational efficiency. Through data sharing, better forecasting, inventory management, and risk mitigation strategies can be implemented. Moreover, data sharing fosters collaboration among supply chain partners enhances transparency and trust, and makes the supply chain more synchronized and responsive, which leads to value cocreation within the supply chain, with downstream enterprises being more incentivized than upstream enterprises by this value cocreation.

Abstract Discovery of nitrate/nitrite-dependent anaerobic methane oxidation (DAMO) challenges the conventional biological treatment processes, since it provides a possibility of simultaneously mitigating dissolved methane emissions from anaerobic effluents and reducing additional carbon sources for denitrification. Due to the slow growth of specialized DAMO microbes, this possibility has been just practiced with biofilms in membrane biofilm reactors or granular sludge in membrane bioreactors. In this study, simultaneous elimination of dissolved methane from anaerobic effluents and nitrate/nitrite reduction was achieved in a conventional anoxic reactor with magnetite. Calculations of electron flow balance showed that, with magnetite the eliminated dissolved methane was almost entirely used for nitrate/nitrite reduction, while without magnetite approximately 52 % of eliminated dissolved methane was converted to unknown organics. Metagenomic sequencing showed that, when dissolved methane served as an electron donor, the abundance of genes for reverse methanogenesis and denitrification dramatically increased, indicating that anaerobic oxidation of methane (AOM) coupled to nitrate/nitrite reduction occurred. Magnetite increased the abundance of genes encoding the key enzymes involved in whole reverse methanogenesis and Nir and Nor involved in denitrification, compared to that without magnetite. Analysis of microbial communities showed that, AOM coupled to nitrate/nitrite reduction was proceeded by syntrophic consortia comprised of methane oxidizers, Methanolinea and Methanobacterium, and nitrate/nitrite reducers, Armatimonadetes_gp5 and Thauera. With magnetite syntrophic consortia exchanged electrons more effectively than that without magnetite, further supporting the microbial growth.

Abstract MicroRNAs (miRNAs) are noncoding small regulatory RNAs involved in diverse biological processes. Odontotermes formosanus (Shiraki) is a polyphagous pest that causes economic damage to agroforestry. Serratia marcescens is a bacterium with great potential for controlling this insect. However, knowledge about the miRNA pathway and the role of miRNAs in O. formosanus defense against SM1 is limited. In this study, OfAgo1, OfDicer1 and OfDrosha were differentially expressed in different castes and tissues. SM1 infection affected the expression of all three genes in O. formosanus. Then, we used specific double-stranded RNAs to silence OfAgo1, OfDicer1 and OfDrosha. Knockdown of these genes enhanced the virulence of SM1 to O. formosanus, suggesting that miRNAs were critical in the defense of O. formosanus against SM1. Furthermore, we sequenced miRNAs from SM1-infected and uninfected O. formosanus. 33 differentially expressed (DE) miRNAs were identified, whereby 22 were upregulated and 11 were downregulated. Finally, the miRNA-mRNA networks were constructed, which further suggested the important role of miRNAs in the defense of O. formosanus against SM1. Totally, O. formosanus miRNA core genes defend against SM1 infection by regulating miRNA expression. This study elucidates the interactions between O. formosanus and SM1 and provides new theories for biological control.
