    Abstract Objective: This study aimed to develop and validate a rapid, simple, accurate and precise analytical method for the quantification of L-AA in vitamin C serums. Moreover, the developed method was further applied to determine L-AA in eight different brands of vitamin C serums. A complementary study was also carried out to evaluate the stability of L-AA in the vitamin C serum samples after 15, 30, 45 and 60 days of storage at ambient temperature (15-35°C).
    Methods: Ultra-high-performance liquid chromatography-tandem mass spectrometry was applied.
    Results: Quantitative analyses were performed with a total chromatographic run time of 1.5 min by matrix-matched calibration, and the analytical curve was linear over the range of 1-1700 µg L
    Conclusion: The proposed method could be powerful in routine analyses to ensure the quantification of L-AA in vitamin C serums since it proved to be a simple, reliable, fast, precise, accurate and sensitive analytical method.

    Abstract The aim of this work was to verify the effects of methanol (MeOH) and hydroalcoholic (HA) extracts and their respective partition phases obtained from white mangrove (Laguncularia racemosa (L.) C.F. Gaertn.) leaves on human thrombin activity. Among the extracts and phases tested, only the ethyl acetate and butanolic partitions significantly inhibited human thrombin activity and the coagulation of plasma in the presence of this enzyme. Chromatographic analyses of the thrombin samples incubated with these phases revealed that different compounds were able to interact with thrombin. The butanolic phase of the MeOH extract had the most potent inhibitory effects, reducing enzymatic activity and thrombin-induced plasma coagulation. Two glycosylated flavonoids in this partition were identified as the most potent inhibitors of human thrombin activity, namely quercetin-3-O-arabinoside (QAra) and quercetin-3-O-rhamnoside (Qn). Chromatographic analyses of thrombin samples incubated with these flavonoids demonstrated the chemical modification of this enzyme, suggesting that the MeOH extract contained other compounds that both induced structural changes in thrombin and diminished its activity. In this article, we show that despite the near absence of the medical use of mangrove compounds, this plant contains natural compounds with potential therapeutic applications.

    Abstract Objetivou-se no trabalho avaliar a influência da enxertia e do porta-enxerto 'Apoatã IAC 2258' no comportamento agronômico de sete cultivares de cafeeiro da espécie Coffea arabica cultivadas em solo isento de fitonematoides. O ensaio foi conduzido em delineamento experimental de blocos casualizados em esquema fatorial (7 x 3) com quatro repetições, sendo sete cultivares de Coffea arabica ('Obatã IAC 1669-20', 'Acauã', 'Oeiras MG 6851', 'Catucaí Amarelo 2SL', 'Topázio MG 1190', 'IBC Palma II' e 'Paraíso MG H 419-1') e três tipos de enxertia (muda enxertada, auto enxertada e pé franco). Foi avaliada a produtividade média de quatro anos, uniformidade de maturação, percentagem de frutos chochos e classificação do grão por tipo de peneira. As cultivares enxertadas estudadas apresentaram comportamento agronômico satisfatório em todas as características avaliadas e semelhante ao pé-franco. Em áreas isentas de fitonematoides, à exceção das cultivares 'IBC Palma II' e 'Paraíso MG H 419-1', as demais cultivares estudadas são adequadas opções de escolha para uso como copas. A técnica da enxertia com o uso do porta-enxerto 'Apoatã IAC 2258' em área isenta de fitonematoides não se justifica para incremento de produtividade. This research had the objective to evaluate the effect of grafting and 'Apoatã IAC 2258' cv. as rootstock on agronomic performance of seven Coffea arabica cvs. planted in area nematodes free. The experimental design was randomized blocks using a 7x3 factorial scheme with four repetitions. The treatments were 'Obatã IAC 1669-20', 'Acauã', 'Oeiras MG 6851', 'Catucaí Amarelo 2SL', 'Topázio MG 1190', 'IBC Palma II' and 'Paraíso MG H 419-1' coffee cultivars grafted on 'Apoatã IAC 2258', self grafted (meaning a cultivar was grafted on a rootstock of the same cultivar) and no grafting. The characteristic evaluated was the average yield of beans between the first and the fourth year, the fruit maturation stage, floating grain percentage and the bean size. The grafting cultivars showed good agronomic performance in the characteristic evaluated. The performance in coffee cultivars grafted on 'Apoatã IAC 2258' is the same in no grafting. In nematodes free areas, with the exception of the cultivars 'IBC Palma II' and 'Paraíso MG H 419-1', all cultivars are good choices to be used as scions. The grafting technique using 'Apoatã IAC 2258' as rootstock on area free of nematodes is not justified for yield increase.

    Abstract In previous studies it was found that thymulin 5cH (thymic hormone) can modulate immune processes in several diseases. Additionally, the Antimonium crudum has used in dogs bearing leishmaniosis, according to the similia principle. We studied the inflammatory and immune modulation by thymulin 5CH and Antimonium crudum 30CH treatment in mice experimentally inoculated with Leishmania (L.) amazonensis. Male adult Balb/c mice were inoculated with Leishmania (2x105 promastigotes) into the footpad to induce inflammatory response and peritoneum and spleen cells were evaluated by flow cytometry after 60 days. Animals were divided in 3 groups (n=10): thymulin 5cH, Antimonium crudum 30cH and vehicle /control. Treatment was made in blind, daily, in water/alcohol 30% diluted 1:2500 in drinking water, during all experimental period. CD11b (activated phagocytes and B1 cells), CD19 (B1 cells and B2), CD4 and CD8 (effective T lymphocytes) markers were used to identify immune cells subsets in peritoneal washing fluid and spleen cell suspension. Mice treated with thymulin 5cH presented increase in peritoneal and spleen B1 stem cells (X2=0.0001) and higher CD8+/CD4+ ratio in spleen, regarding to the control. Also, Antimonium crudum 30CH induced a mild increase in B1 cells in peritoneum and spleen ( both X2, p=0.0001). Further histological analysis of the primary lesion will be done in the next step, to elucidate the impact of these findings in the disease evolution. The results show that both treatments stimulate B1 stem cell proliferation and suggest the cooperation of T spleen lymphocytes in the process.

    Abstract The Cyperaceae show the ability to incorporate silicon by depositing colloidal silica, which is recorded by the occurrence of projections in the form of cones, in inner tangential walls of some epidermal cells or "silica cells". Leaves of C. ligularis and R. aberrans were analyzed through the technique of electron backscatter. Cyperus ligularis accumulates silica, in addition to "silica cells", in some stomata, trichomes and the cell walls that surround the cavities of the aerenchyma. The silica in the latter occurs in various forms; however, the cells located near the vascular bundles have conical projections, similar to those of the epidermis. Rhynchospora aberrans presents "silica cells" whose projections have tapered "satellites". In this species, silica also occurs in stomata and certain epidermal cells adjacent to them. It appears that the silicon deposition occurs in combination with the wall (with no apparent structural changes), and structures of secretion, or projections of the wall. These structural changes in the species, and location, are probably related to functional and environmental factors, especially the soil, in addition to relation with taxonomic groups. As Cyperaceae têm a capacidade de incorporação do ácido silícico mediante depósito de sílica coloidal, que é registrada pela ocorrência de projeções sob a forma de cones, nas paredes tangenciais internas de determinadas células epidérmicas ou "células de sílica". Folhas de C. ligularis e R. aberrans foram analisadas pela técnica de elétrons retroespalhados. C. ligularis além de acumular sílica nas "células de sílica", acumula também em alguns estômatos, nos tricomas e nas paredes das células que contornam as cavidades do aerênquima. Este último apresenta formas diversas, entretanto, as células que se localizam junto aos feixes vasculares mostram projeções cônicas, semelhantes àquelas da epiderme. R. aberrans contém "células de silica" cujas projeções cônicas têm "satélites". A sílica também ocorre em estômatos específicos e em células epidérmicas contíguas aos mesmos. Constata-se que a deposição de sílica ocorre tanto em combinação com a parede (sem modificações estruturais aparentes), como em estruturas de secreção da mesma, ou seja, projeções da parede. Essas variações de ocorrência estrutural e de localização nas espécies devem estar relacionadas com a funcionalidade, fatores ambientais, com destaque ao solo, além da relação com grupos taxonômicos.

    Abstract Leprosis, caused by citrus leprosis virus (CiLV) and transmitted by the tenuipalpid mite Brevipalpus phoenicis, is one of the most important viruses of citrus in the Americas. Sweet oranges (Citrus sinensis L. Osb.) are highly susceptible to CiLV, while mandarins (C. reticulata Blanco) and some of their hybrids have higher tolerance or resistance to this disease. The mechanisms involved in the resistance and its inheritance are still largely unknown. To study the quantitative trait loci (QTL; quantitative trait loci) associated with the resistance to CiLV, progeny analyses were established with 143 hybrid individuals of 'Pêra' sweet orange (C. sinensis L. Osb.) and 'Murcott' tangor (C. reticulata Blanco x C. sinensis L. Osb.) from controlled crossings. Disease assessment of the hybrid individuals was conducted by infesting the plants with viruliferous mites in the field. The experiment consisted of a randomized completely block design with ten replicates. The evaluated phenotypic traits were incidence and severity of the disease on leaves and branches, for a period of 3 years. The MapQTL[trade mark sign] v.4.0 software was used for the identification and location of possible QTL associated with resistance to CiLV on a genetic map obtained from 260 AFLP and 5 RAPD markers. Only consistent QTLs from different phenotypic traits and years of evaluation, with the critical LOD scores to determine the presence or absence of each QTL calculated through the random permutation test, were considered. A QTL was observed and had a significant effect on the phenotypic variation, ranging from 79.4 to 84% depending on which trait (incidence or severity) was assessed. This suggests that few genes are involved in the genetic resistance of citrus to CiLV.

    Abstract Continental and marine conditions during the last millennium off Porto, Portugal (the southern pole of the North Atlantic Oscillation, NAO), are reconstructed from a sediment archive through a high-resolution multiproxy study and instrumental evidence. Results show multidecadal variability and sea surface temperatures (SSTs) that correlate well with previously published land and sea-based Northern Hemisphere temperature records, and appear to be responding to long-term solar insolation variability. Precipitation was negatively correlated with the NAO, whereas strong flooding events occurred at times of marked climate cooling (AD 1100-1150 and 1400-1470) and transitions in solar activity. AD 1850 marks a major shift in the phytoplankton community associated with a decoupling of d18O records of 3 planktonic foraminifera species. These changes are interpreted as a response to a reduction in the summer and/or annual upwelling and more frequent fall-winter upwelling-like events. This shift's coincidence with a decrease in SST and the increase in coherence between our data and the Atlantic Multidecadal Oscillation (AMO) confirms the connection of the upwelling variability to the North Atlantic Ocean's surface and thermohaline circulation on a decadal scale. The disappearance of this agreement between the AMO and our records beyond AD 1850 and its coincidence with the beginning of the recent rise in atmospheric CO2 supports the hypothesis of a strong anthropogenic effect on the last ~150 yr of the climate record. Furthermore, it raises an important question of the use of instrumental records as the sole calibration data set for climate reconstructions, as these may not provide the best analogue for climate beyond AD 1730.

    Abstract Perinatal hypoxia-ischemia represents a significant risk to CNS development, leading to high mortality rates, diverse damages, and persistent neurological deficits. Despite advances in neonatal medicine in recent decades, the incidence of HIE remains substantial. Motor deficits can manifest early, while cognitive impairments may be diagnosed later, emphasizing the need for extended follow-up. This review aims to explore potential candidates for therapeutic interventions for hypoxic-ischemic encephalopathy (HIE), with a focus on cognitive deficits. We searched randomized clinical trials (RCT) that tested drug treatments for HIE and evaluated cognitive outcomes. The results included studies on erythropoietin, melatonin, magnesium sulfate, topiramate, and a combination of vitamin C and ibuprofen. Although there are several indications of the efficacy of these drugs among animal models, considering neuroprotective properties, the RCTs failed to provide complete effectiveness in the context of cognitive impairments derived from HIE. More robust RCTs are still needed to advance our knowledge and to establish standardized treatments for HIE.
