    Abstract Alzheimer's disease, a neurodegenerative disease, is a progressive and irreversible disease that has become a global challenge due to its increasing prevalence and absence of available potential therapies. Protein misfolding and aggregation are known to be the root of several protein neurodegenerative diseases, including Alzheimer's disease. Protein aggregation is a phenomenon where misfolded proteins accumulate and clump together intra-or extracellularly. This accumulation of misfolded amyloid proteins leads to the formation of plaquesin the neuronal cells, also known as amyloid β plaques. The synthesis of amyloid β plaques and tau protein aggregation are the hallmarks of Alzheimer's disease. Potential therapeutics must be developed in conjunction with an understanding of the possible root cause involving complex mechanisms. The development of therapeutics that can inhibit protein misfolding and aggregation, involved in the pathogenesis of Alzheimer's disease, could be one of the potential solutions to the disease.

    Abstract The incidence of cancer is rising globally at a remarkable rate. According to the global cancer Observatory, the total number of cases in Pakistan were 178,388 in 2020; and these figures were provided by the Punjab Cancer Registry which represents the population of Lahore and some parts of central Punjab. Considering the overall 220 million population, these figures are severe underestimation of the true cancer burden in Pakistan. Unfortunately, several attempts at establishing a national cancer registry in the country have failed in the past. Resource limitations, including funds, manpower and infrastructure, and the lack of interest on the part of the governments are behind the failure of the establishment of a national cancer database. ---Continue

    Abstract This case report demonstrates the full mouth prosthetic rehabilitation of a patient with generalised compensated tooth wear and oral sub-mucous fibrosis (OSMF). A 50- year-old female with advanced tooth wear and concomitant OSMF presented to the clinic. She was managed in a sequential manner using an occlusal splint, extractions of hopeless teeth, root canal treatment of strategically important teeth, surgical crown lengthening of abutment teeth, and dental implants to replace missing dentition, followed by crown and bridgework to rehabilitate the dentition. All procedures were done on the principles of re-organised occlusal intervention. The outcome was a functional dentition with an aesthetic smile and a satisfied patient. The regaining of mouth opening among OSMF subjects is a challenge. The usual management is prescribing muscle stretching exercises, drugs, or surgery. In the present case, we managed to achieve significant improvement in mouth opening with occlusal splints. The use of such occlusal orthotics has never been reported.

    Abstract Pancreatic gastrointestinal tumour is an unusual primary tumour of the pancreas. A 31-year-old male came to the clinic with jaundice and weight loss. Cross-sectional imaging showed a mass in the pancreatic uncinate process. Image-guided biopsy revealed gastrointestinal stromal tumour, so pancreaticoduodenectomy was performed, followed by adjuvant Imatinib. The patient had oligo-metastasis in the liver five years post-surgery and underwent liver resection. This is an unusual case where a pancreatic GIST presented with metastasis while on adjuvant treatment. Hepatectomy and multimodal therapy increases the survival if the disease is confined to the liver.

Unfortunately, several attempts at establishing a national cancer registry in the country have failed in the past. Resource limitations, including funds, manpower and infrastructure, and the lack of interest on the part of the governments are behind the failure of the establishment of a national cancer database. There is a dire need for establishing a national cancer registry to understand the actual burden of disease, and to implement a national action plan for cancer prevention, and management strategies to control the growing epidemic.
