    Abstract As a vital component of blood, platelets play crucial roles in hemostasis and maintaining vascular integrity, and actively participate in inflammation and immune regulation. The unique biological properties of natural platelets have enabled their utilization as drug delivery vehicles. The advancement and integration of various techniques, including biological, chemical, and physicochemical methods, have enabled the preparation of engineered platelets. Platelets can serve as drug delivery platforms combined with immunotherapy and chemokine therapy to enhance their therapeutic impact. This review focuses on the recent advancements in the application of unactivated platelets for drug delivery. The construction strategies of engineered platelets are comprehensively summarized, encompassing internal loading, surface modification, and genetic engineering techniques. Engineered platelets hold vast potential for treating cardiovascular diseases, cancers, and infectious diseases. Furthermore, the challenges and potential considerations in creating engineered platelets with natural activity are discussed.

    Abstract Background: Single umbilical artery (SUA) is strongly associated with foetal structural abnormalities; however, the exact pattern of this association has not been described. We aimed to investigate the occurrence of malformations in singleton pregnancies with SUA in China and to study the association between the absent side of the umbilical artery and foetal malformations.
    Methods: This was a retrospective study of singleton pregnancies for which routine first-trimester anatomical screening was performed at 11
    Results: The incidence of SUA in our study was 2.0% (970/47,894). Of all foetuses with SUA, 387 (39.9%) had structural malformations. The malformation type varied, with cardiovascular complications being the most common. A robust association was observed between SUA and oesophageal stenosis or atresia (OR: 25.33), followed by cardiovascular (OR: 9.98-24.02), scoliosis (OR: 18.62), genitourinary (OR: 2.45-15.66), and brain malformations (OR: 4.73-9.12). The absence of the left umbilical artery (n = 445, 45.9%) was consistent with that of the right umbilical artery (n = 431, 44.4%). Furthermore, a significantly higher rate of an absent right than the left umbilical artery (p<0.01) was observed in SUA with foetal abnormalities than in SUA with no malformations.
    Conclusions: Overall, we observed a higher risk of various specific malformations in foetuses with SUA, and a strong association between SUA and oesophageal stenosis or atresia. The absence of the right umbilical artery was most common in foetuses with SUA and structural malformations. This study provides a reference for ultrasonographers in conducting foetal structural screening for pregnant women with SUA.

    Abstract Opinion statement: Cardio-oncology is an emerging interdisciplinary field dedicated to the early detection and treatment of adverse cardiovascular events associated with anticancer treatment, and current clinical management of anticancer-treatment-related cardiovascular toxicity (CTR-CVT) remains limited by a lack of detailed phenotypic data. However, the promise of diagnosing CTR-CVT using deep phenotyping has emerged with the development of precision medicine, particularly the use of omics-based methodologies to discover sensitive biomarkers of the disease. In the future, combining information produced by a variety of omics methodologies could expand the clinical practice of cardio-oncology. In this review, we demonstrate how omics approaches can improve our comprehension of CTR-CVT deep phenotyping, discuss the positive and negative aspects of available omics approaches for CTR-CVT diagnosis, and outline how to integrate multiple sets of omics data into individualized monitoring and treatment. This will offer a reliable technical route for lowering cardiovascular morbidity and mortality in cancer patients and survivors.

    Abstract Chronic neurodegeneration and acute injuries lead to neuron losses via diverse processes. We compared retinal ganglion cell (RGC) responses between chronic glaucomatous conditions and the acute injury model. Among major RGC subclasses, αRGCs and intrinsically photosensitive RGCs (ipRGCs) preferentially survive glaucomatous conditions, similar to findings in the retina subject to axotomy. Focusing on an αRGC intrinsic factor, Osteopontin (secreted phosphoprotein 1 [Spp1]), we found an ectopic neuronal expression of Osteopontin (Spp1) in other RGCs subject to glaucomatous conditions. This contrasted with the Spp1 downregulation subject to axotomy. αRGC-specific Spp1 elimination led to significant αRGC loss, diminishing their resiliency. Spp1 overexpression led to robust neuroprotection of susceptible RGC subclasses under glaucomatous conditions. In contrast, Spp1 overexpression did not significantly protect RGCs subject to axotomy. Additionally, SPP1 marked adult human RGC subsets with large somata and SPP1 expression in the aqueous humor correlated with glaucoma severity. Our study reveals Spp1's role in mediating neuronal resiliency in glaucoma.

    Abstract Major depressive disorder (MDD) is a highly heterogeneous mental disorder, and its complex etiology and unclear mechanism are great obstacles to the diagnosis and treatment of the disease. Studies have shown that abnormal functions of the visual cortex have been reported in MDD patients, and the actions of several antidepressants coincide with improvements in the structure and synaptic functions of the visual cortex. In this review, we critically evaluate current evidence showing the involvement of the malfunctioning visual cortex in the pathophysiology and therapeutic process of depression. In addition, we discuss the molecular mechanisms of visual cortex dysfunction that may underlie the pathogenesis of MDD. Although the precise roles of visual cortex abnormalities in MDD remain uncertain, this undervalued brain region may become a novel area for the treatment of depressed patients.

    Abstract Objective: The efficacy of single-incision plus one-port laparoscopic surgery (SILS + 1) versus conventional laparoscopic surgery (CLS) for colorectal cancer treatment remains unclear. This study compares the short-term and long-term outcomes of SILS + 1 and CLS using a high-quality systematic review and meta-analysis.
    Method: Literature search followed the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines, drawing from PubMed, Embase, Web of Science, and the Cochrane Library until December 10, 2023. Statistical analysis was conducted using RevMan and Stata.
    Result: The review and meta-analysis included seven studies with 1740 colorectal cancer patients. Compared to CLS, SILS + 1 showed significant improvements in operation time (WMD =  - 18.33, P < 0.00001), blood loss (WMD =  - 21.31, P < 0.00001), incision length (WMD =  - 2.07, P < 0.00001), time to first defecation (WMD =  - 14.91, P = 0.009), time to oral intake (WMD =  - 11.46, P = 0.04), and time to ambulation (WMD =  - 11.52, P = 0.01). There were no significant differences in lymph node harvest, resection margins, complications, anastomotic leakage, hospital stay, disease-free survival, overall survival, and postoperative recurrence.
    Conclusions: Compared to CLS, SILS + 1 demonstrates superiority in shortening the surgical incision and promoting postoperative recovery. SILS + 1 can provide a safe and feasible alternative to CLS.

    Abstract Nowadays, unmanned aerial vehicle (UAV) communication systems are commonly considered as one of the key enabling technologies for 6G. The hybrid free space optical (FSO)/radio frequency (RF) system has the advantages of both FSO and RF links to improve communication system performance, and the relay-assisted system adopts multi-hop transmission and cooperative diversity methods to extend communication coverage. Thus, a joint consideration of UAV-assistedUAV assisted relay in hybrid FSO/RF transmission is meaningful. In this paper, we aim to analyze the performance of UAV-assisted multi-hop parallel hybrid FSO/RF communication systems with and without pointing errors (PE) in terms of Bit Error Rate (BER) and outage probability. In our considered system, the FSO sub-link adopts the Exponential Weibull turbulence model and the RF sub-link suffers the Nakagami fading model. With these, new mathematical formulas of both BER and outage probability are derived under the UAV-assisted hybrid FSO/RF with different modulation methods. Through numerical evaluationnumerical simulations, the performances of UAV-assisted hybrid FSO/RF systems are analyzed under different weather conditions, modulation methods, optical receiver aperture, RF fading parameters, pointing errors, and relay structures. The results demonstrate that (1) compared to hybrid FSO/RF direct links, UAV-assisted hybrid FSO/RF systems can further improve system performance; (2) the performance of UAV-assisted hybrid FSO/RF systems varies with different relay structures; (3) large receiver aperture and RF fading parameters can further improve the communication performance of hybrid FSO/RF direct links and UAV-assisted hybrid FSO/RF systems.

    Abstract To improve the performance of fee-space optical communication systems, this paper analyzes the performance of a relay-aided hybrid fee-space optical (FSO)/radio frequency (RF) cooperation system based on a selective combination and decoding forward transmission scheme. In this system, the FSO sub-link experienced Málaga turbulence with pointing errors and the RF sub-link suffered Nakagami-m fading. Firstly, the probability density function (PDF) and cumulative distribution function (CDF) of the end-to-end output signal-to-noise ratio (SNR) of the relay-aided hybrid FSO/RF system are derived. Then, using the extended generalized bivariate Meijer's G-function (EGBMGF) and the approximate analytical formula of the generalized Gauss-Laguerre integral, mathematical expressions of the end-to-end average bit error rate (ABER) and outage probability of the relay-aided hybrid FSO/RF system with different subcarrier intensity modulation and different detection schemes are derived. Through a simulation analysis of the system, the results show that compared with the other three modulation technologies, the hybrid FSO/RF direct link and relay-aided hybrid FSO/RF system with coherent binary phase shift keying (CBPSK) modulation have the best bit error performance. Compared with direct detection, the hybrid direct link and relay-aided hybrid system with coherent detection can significantly improve the communication performance.
