    Abstract Background: Compound Xiao-ai-fei honey ointment (CXHO) is an anticancer preparation with a long history in Uyghur folk medicine in China and has been used for the treatment of gastric cancer (GC) in Xinjiang, China. Nevertheless, the mechanism of its anticancer effect remains to be investigated.
    Methods: Bioactive ingredients of CXHO were retrieved from the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform database. Target genes of ingredients were acquired via the PubChem and Swiss target prediction database. Gene expression profiling of GC was obtained from GSE54129 in the GEO database and analyzed using the limma package in R. The hub genes associated with CXHO in GC were validated using the TIMER2.0 database, GEPIA2 database and Auto Dock tools. The effect of CXHO on migration of GC cells was detected by Transwell chamber assay and Wound healing assay. The effect of CXHO on expression levels of MMP2/MMP9 and NF-κb, PI3K/AKT signaling pathway was detected by Western blot assay.
    Results: Forty-five bioactive ingredients and their 819 related genes were found. A total of 462 differentially expressed genes were identified between GC patients and healthy controls. Seventeen common target genes were identified as hub genes CXHO against GC. Among them, MMP2 and MMP9 were significantly associated with tumor immune infiltrates and had good binding affinity with effective ingredients. Moreover, we validated the mRNA and protein expression levels and prognostic value of MMP2 and MMP9 by different databases. In addition, Kyoto encyclopedia of genes and genomes and gene ontology analyses showed that the 17 common target genes were mainly involved in steroid hormone biosynthesis and cancer-related pathways. Experimental results showed that CXHO inhibited migration of GC cells and down regulated the expression levels of MMP2/MMP9, NF-κb. In addition, CXHO can inhibited PI3K/AKT signaling pathway.
    Conclusion: We identified and experimental validated 2 pivotal target genes of CXHO against GC and preliminarily analyzed the potential mechanisms by which CXHO inhibits the development of GC. All these findings support CXHO as a promising drug for the treatment of GC.

    Abstract Background: The goal was to study lipid profiles (TG, TC, LDL, HDL), effects on serum leptin, and fat tissue adiponectin, and resistin as well as body weight effects of Shan He Jian Fei Granules (SHJFG) in rats on a high fat diet.
    Methods: Rats were randomly divided into five groups: normal control group fed with normal fat diet, rats on high fat diet receiving low dosage, middle dosage, high dosage of Shan He Jian Fei Granules (SHJFG) as well as a high fat diet group receiving placebo. Rats were treated for 8 weeks. Body weight and naso-anal length of each rat were recorded and Lee's index was calculated. Serum TG, TC, LDL, HDL and leptin concentrations were analyzed. The gene expressions of adiponectin and resistin in adipose tissues were tested by RT-PCR.
    Results: Compared to the high-fat diet group, body weights, Lee's indexes, weight of fat tissues and serum TG, TC, LDL and leptin of SHJFG groups significantly decreased (p < 0.05), whereas mRNA expressions of adiponectin and resistin of SHJFG groups significantly increased (p < 0.05).
    Conclusions: SHJFG could significantly lower body weight and serum TG, TC, and LDL of obese rats. The effects of SHJFG in lowering leptin synthesis and raising mRNA expression of adiponectin and resistin in fat tissues may act as part of the mechanisms in lowering body weight of obese rats. Further studies are needed to demonstrate whether SHJFG may also reduce overall cardiovascular morbidity and mortality like other lipid lowering drugs.

    Abstract To characterize the epidemiology of autumn-winter type scrub typhus in a new endemic focus of Fei County, Shandong Province, China, field and laboratory studies were conducted to determine the species composition and seasonal fluctuations of chigger mites and their animal hosts. Orientia tsutsugamushi was isolated, and serotypes of antibodies to O. tsutsugamushi were detected and identified. A total of 1044 rodents and insectivores were field collected: 751 Apodemus agrarius (71.94%), 266 Tscherskia triton (25.48%), 1 Cricetulus barabensis (0.09%), 11 Rattus norvegicus (1.05%), 3 Mus musculus (0.29%), and 12 Crocidura suaveolens (1.15%). Apodemus agrarius, T. triton and R. norvegicus were shown to be naturally infected with O. tsutsugamushi. The serum antibody positive rate of rodents was 14.78%. A total of 11762 chigger mites were collected: 2547 Leptotrombidium scutellare (21.65%), 1596 L. palpale (13.57%), 466 L. taishanicum (3.96%), 2833 L. linhuaikongense (24.09%), and 4320 Walchia pacifica (36.73%). Leptotrombidium scutellare was most numerous from September to November and was the predominant species during the local scrub typhus epidemic season. Natural infection with O. tsutsugamushi was detected in L. scutellare, and transovarial transmission of the pathogen by this chigger was demonstrated. 38 strains of O. tsutsugamushi were isolated from 33 patients (14 strains), 3 species of rodents (9 strains), and 4 species of mites (15 strains). All O. tsutsugamushi strains isolated in this endemic focus were of low virulence. Serotype identification showed that 33 strains were of the Gilliam type, and 3 strains were of the Karp type. Another 2 strains remain to be identified. In the Fei County focus of scrub typhus, the main reservoir host is A. agrarius, the predominant mite vector is L. scutellare, the principal serotype of O. tsutsugamushi is Gilliam, and all strains of O. tsutsugamushi are of low virulence. Most cases of scrub typhus occurred from September to November; therefore, scrub typhus in this focus is autumn-winter type. This focus is located in the premontane flatlands of Fei County.

    Abstract During a search for latent vectors of scrub typhus in a focus of the autumn-winter type of this disease, the authors isolated O. tsutsugamushi from larval and nymphal L. linhuaikongense. Isolated O. tsutsugamushi serotyped by indirect immunofluorescence assay (IFA). Larval L. linhuaikongense were initially found in May on the bodies of rodents captured in the field. The infestation reached a peak in August and disappeared in November; chiggers were thus abundant during summer. Nymphal L. linhuaikongense were reared from larvae collected from rodents. Two strains of O. tsutsugamushi were isolated from 4 pools of larvae and 3 pools of nymphs, respectively. This implies that L. linhuaikongense is a potential vector of O. tsutsugamushi, chiefly to rodents but occasionally to humans, during non-endemic seasons.
