    Abstract On March 23, 2022, the U.S. Food and Drug Administration (FDA) approved Pluvicto (lutetium Lu 177 vipivotide tetraxetan), also known as 177Lu-PSMA-617, for the treatment of adult patients with metastatic castration-resistant prostate cancer (mCRPC) who have highly expressed prostate-specific membrane antigen (PSMA) and have at least one metastatic lesion. It is the first FDA-approved targeted radioligand therapy for eligible men with PSMA-positive mCRPC. Lutetium Lu 177 vipivotide tetraxetan is a radioligand that strongly binds to PSMA, making it ideal for treating cancers of the prostate by targeted radiation, resulting in DNA damage and cell death. PSMA is overexpressed in cancer cells while being lowly expressed in normal tissues, which makes it an ideal theranostic target. As precision medicine advances, this is a thrilling turning point for highly individualized treatments. This review aims to summarize the pharmacology and clinical studies of the novel drug lutetium Lu 177 vipivotide tetraxetan for the treatment of mCRPC, emphasizing its mechanism of action, pharmacokinetics and safety.

    Abstract Xin Lu is Professor of Cancer Biology at the University of Oxford, UK, and Director of the Oxford Branch of the Ludwig Institute for Cancer Research, Co-director of the Cancer Research UK Oxford Centre, NIHR Oxford Biomedical Research Centre Multi-Modal Cancer Therapies Theme Lead and Director of the Oxford Centre for Early Cancer Detection. She has a long-standing interest in mechanisms of tumour suppression and cellular plasticity, centred on studies of p53 and the ASPP family of proteins (apoptosis-stimulating protein of p53; ankyrin repeats, SH3 domain and proline-rich sequence-containing proteins). Her laboratory's discovery of ASPPs, regulators or 'molecular switches' of the apoptotic function of p53 led to key insights into the role of cell plasticity in cancer and other diseases, and these could pave the way to new diagnostic and therapeutic interventions. Xin has received many awards and honours in recognition of her significant contributions to cancer biology, including being elected as a Fellow of the Royal Society in 2020. Here, she outlines how her breakthrough discovery of ASPPs came about and its impact on the cancer field, as well as highlighting the importance of mentors including Min Wu, Birgit Lane and Sir David Lane in shaping her early career and helping her to navigate a new research world, having moved to the UK from China in the 80s.

    Abstract Two uranium oxide hydrate frameworks (UOHFs) with either Dy3+ or Lu3+ ions, Dy1.36(H2O)6[(UO2)10UO13(OH)4] (UOHF-Dy) or Lu2(H2O)8[(UO2)10UO14(OH)3] (UOHF-Lu), were synthesized hydrothermally and characterized with a range of structural and spectroscopic techniques. Although SEM-EDS analysis confirmed the same atomic ratio of ~5.5 for U:Dy and U:Lu, they displayed different crystal morphologies, needles for UOHF-Dy in the orthorhombic C2221 space group and plates for UOHF-Lu in the triclinic P-1 space group. Both frameworks are composed of β-U3O8 type layers linked by pentagonal bipyramidal uranium polyhedra, with the Dy3+/Lu3+ ions inside the channels. However, the arrangements of Dy3+/Lu3+ ions are different, with disordered Dy3+ ions well aligned at the centers of the channels and single Lu3+ ions well-separated in a zigzag pattern in the channels. While the characteristic vibrational modes were revealed by Raman spectroscopy, the presence of a pentavalent uranium center in UOHF-Lu was confirmed with diffuse reflectance spectroscopy. The formation of two types of UOHFs with lanthanide ions, high or low symmetry, and the structure trend were discussed regards to synthesis conditions and lanthanide ionic radius. This work highlights the complex chemistry driving the formation of UOHFs with lanthanide ions and has implications to the spent nuclear fuel under geological disposal.
