    Abstract A disponibilização de teses e dissertações no Repositório Institucional da Universidade Federal de Juiz de Fora (RI-UFJF) ocorre aproximadamente 90 dias após a defesa do trabalho e aprovação pela banca examinadora. A lacuna existente entre o aceite e o acesso ocorre devido ao processo de homologação tramitado dentro da instituição para registro e validação do título obtido. Em alusão aos preprints utilizados para acelerar o processo de comunicação dos resultados de pesquisas em periódicos, o registro prévio dos principais dados da pesquisa acadêmica foi a solução encontrada para trazer a celeridade para divulgação das produções. O pré-registro de teses e dissertações é realizado a partir das informações contidas na matéria jornalística realizada pela Equipe de Divulgação Científica da Diretoria de Imagem Institucional da Universidade Federal de Juiz de Fora (UFJF) e divulgada no site da instituição. A coleção “Defesas Recentes – Teses e Dissertações” contém os pré-registros – título do trabalho, orientador, autor, banca – e utiliza a matéria como se fosse o resumo do trabalho. O texto completo do arquivo fica pendente e é inserido por ocasião do recebimento do processo homologado no Repositório Institucional. Com a divulgação do pré-registro imediatamente após a defesa, as principais informações, bem como os resultados das pesquisas que foram objeto de tese e dissertações já podem ser recuperados em pesquisas realizadas no RI-UFJF e em serviços de busca.

    Abstract We report the first record of Turdus hauxwelli Lawrence, 1869, Hauxwell’s Thrush, from the state of Goiás, central Brazil, based on photographic and vocal records obtained at Taquaral de Goiás. This new record substantially expands the species’ distribution beyond the Amazonian domain and marks its new easternmost geographical limit. The new record extends this species’ distribution by 650 km from the nearest recent record. We also discuss the importance of this record in understanding the biogeographical affinities of the seasonal forests of central-western Brazil.

    Abstract Conventional anticancer therapies present low specificity, leading to several secondary effects. To improve these drawbacks, aptamers able to fold into G-quadruplex (G4) are being used to promote drug accumulation in cancer cells. AS1411 is a G4 aptamer able to recognize nucleolin, a protein overexpressed on cancer cells’ surfaces. This aptamer was tested in clinical trials, but it showed low response rates and suboptimal pharmacokinetics. Nevertheless, AS1411 is being used as a targeting agent. Moreover, AS1411 derivatives were developed, with improved toxicity and high affinity to nucleolin. Thus, we propose to use AT11, an AS1411 derivative, to functionalize liposomes and improve the selectivity of C 8 (a potential anticancer drug) into oral cancer. Therefore, we produced liposomes (blank or C 8- associated) by the ethanol injection method to, then, we functionalized them with AT11-TEG-Cholesteryl. The resulting liposomes were characterized by DLS. C 8 association was determined by UV/vis spectroscopy, and the AT11 functionalization was determined by SDS-PAGE. The effect of blank and C 8- associated liposomes on oral cancer and healthy cells’ viability was determined by MTT, and its internalization of was visualized by confocal microscopy. Liposomes with hydrodynamic diameters of 148–168 nm were obtained, and C 8 was efficiently associated (~100%). When the cells were treated with blank liposomes, cell viability was almost unaffected. After treating with C 8 -associated liposomes, both cell lines showed a dose–response effect. Additionally, we observed that AT11-liposomes can internalize and reach the cytoplasm of cells. Overall, these findings suggest that the tested liposomes are promising drug carriers for oral cancer therapy.

    Abstract One of the major problems facing humanity in the coming decades is the production of food on a large scale. The production of large quantities of food must be conducted in a sustainable and responsible manner for nature and humans. In this sense, the appropriate application of agricultural pesticides plays a fundamental role since pesticide application in a qualified manner reduces human and environmental risks as well as the costs of food production. Evaluation of the quality of application using sprayers is an important issue, and several quality descriptors related to the average diameter and distribution of droplets are used. This paper describes the construction of a data-driven soft sensor using the parametric principal component regression (PCR) method based on principal component analysis (PCA), which works in two configurations: with the input being the operating conditions of the agricultural boom sprayers and its outputs being the prediction of the quality descriptors of spraying, and vice versa. The soft sensor provides, in one configuration, estimates of the quality of pesticide application at a certain time and, in the other, estimates of the appropriate sprayer-operating conditions, which can be used for control and optimization of the processes in pesticide application. Full cone nozzles are used to illustrate a practical application as well as to validate the usefulness of the soft sensor designed with the PCR method. The selection of historical data, exploration, and filtering of data, and the structure and validation of the soft sensor are presented. For comparison purposes, the results with the well-known nonparametric k-Nearest Neighbor (k-NN) regression method are presented. The results of this research reveal the usefulness of soft sensors in the application of agricultural pesticides and as a knowledge base to assist in agricultural decision-making.

    Abstract Aim of study: Germination assays are vital in the management of material preserved in germplasm banks. The rules published by the International Seed Testing Association (ISTA) are generally those followed in such assays. In wild species, seed dormancy and inter-population variability increase the difficulty in estimating seed viability. The aim of the present work was to determine the germination requirements of the seeds from different wild populations of pasture grasses species. Area of study: Northwestern Spain Material and methods: Seeds from eight wild populations of different species of Festuca, all from northwestern Spain, were studied. Germination assays were performed under constant and alternating temperature conditions. Treatments for removing seed dormancy (cold stratification and gibberellic acid application) were also applied. A full parametric time-to event model was used for data analysis. Main results: In general, the optimum environmental temperature for germination was around 15°C for the populations of Festuca group ovina, F. gr. rubra and F. gigantea; temperatures of 20-30ºC had a negative effect. All the examined populations, except that of tall fescue (Lolium arundinaceum), showed non-deep physiological dormancy at suboptimal germination temperatures, but this was breakable by the application of gibberellic acid and by cold stratification. Research highlights: There are clear inter- and intra-specific differences in germination requirements that might be associated with place of origin. The ISTA germination assay recommendations for wild members of fescues may not be the most appropriate.

    Abstract Abstract In this work, we demonstrate the development of a rapid and label-free electrochemical biosensor to detect Listeria monocytogenes using a novel stimulus–response thiomer nanobrush material. Nanobrushes were developed via one-step simultaneous co-deposition of nanoplatinum (Pt) and alginate thiomers (ALG-thiomer). ALG-thiomer/Pt nanobrush platform significantly increased the average electroactive surface area of electrodes by 7 folds and maintained the actuation properties (pH-stimulated osmotic swelling) of the alginate. Dielectric behavior during brush actuation was characterized with positively, neutral, and negatively charged redox probes above and below the isoelectric point of alginate, indicating ALG-thiomer surface charge plays an important role in signal acquisition. The ALG-thiomer platform was biofunctionalized with an aptamer selective for the internalin A protein on Listeria for biosensing applications. Aptamer loading was optimized and various cell capture strategies were investigated (brush extended versus collapsed). Maximum cell capture occurs when the ALG-thiomer/aptamer is in the extended conformation (pH > 3.5), followed by impedance measurement in the collapsed conformation (pH < 3.5). Low concentrations of bacteria (5 CFU mL−1) were sensed from a complex food matrix (chicken broth) and selectivity testing against other Gram-positive bacteria (Staphylococcus aureus) indicate the aptamer affinity is maintained, even at these pH values. The new hybrid soft material is among the most efficient and fastest (17 min) for L. monocytogenes biosensing to date, and does not require sample pretreatment, constituting a promising new material platform for sensing small molecules or cells.

    Abstract During cell division, drastic cellular changes characteristic of mitosis result in the inactivation of the transcriptional machinery, and global downregulation of transcription. Sequence-specific transcription factors (TFs) have thus been considered mere bystanders, devoid of any regulatory function during mitosis. This view changed significantly in recent years, upon the conclusion that many TFs associate with condensed chromosomes during cell division, even occupying a fraction of their genomic target sites in mitotic chromatin. This finding was at the origin of the concept of mitotic bookmarking by TFs, proposed as a mechanism to propagate gene regulatory information across cell divisions, by facilitating the reactivation of specific bookmarked genes. While the underlying mechanisms and biological significance of this model remain elusive, recent developments in this fast-moving field have cast new light into TF activity during mitosis, beyond a bookmarking role. Here, we start by reviewing the most recent findings on the complex nature of TF–chromatin interactions during mitosis, and on mechanisms that may regulate them. Next, and in light of recent reports describing how transcription is reinitiated in temporally distinct waves during mitosis-to-G1 transition, we explore how TFs may contribute to defining this hierarchical gene expression process. Finally, we discuss how TF activity during mitotic exit may impact the acquisition of cell identity upon cell division, and propose a model that integrates dynamic changes in TF–chromatin interactions during this cell-cycle period, with the execution of cell-fate decisions.
