    Abstract In the present work, different hydrolases were adsorbed onto polypropylene beads to investigate their activity both in short-esters and polyesters synthesis. The software MODDE ® Pro 13 (Sartorius) was used to develop a full-factorial design of experiments (DoE) to analyse the thermostability and selectivity of the immobilized enzyme towards alcohols and acids with different chain lengths in short-esters synthesis reactions. The temperature optima of Candida antarctica lipase B (CaLB), Humicola insolens cutinase (HiC), and Thermobifida cellulosilytica cutinase 1 (Thc_Cut1) were 85 °C, 70 °C, and 50 °C. CaLB and HiC preferred long-chain alcohols and acids as substrate in contrast to Thc_Cut1, which was more active on short-chain monomers. Polymerization of different esters as building blocks was carried out to confirm the applicability of the obtained model on larger macromolecules. The selectivity of both CaLB and HiC was investigated and best results were obtained for dimethyl sebacate (DMSe), leading to polyesters with a M w of 18 kDa and 6 kDa. For the polymerization of dimethyl adipate (DMA) with BDO and ODO, higher molecular masses were obtained when using CaLB onto polypropylene beads (CaLB_PP) as compared with CaLB immobilized on macroporous acrylic resin beads (i.e., Novozym 435). Namely, for BDO the M n were 7500 and 4300 Da and for ODO 8100 and 5000 Da for CaLB_PP and for the commercial enzymes, respectively. Thc_Cut1 led to polymers with lower molecular masses, with M n < 1 kDa. This enzyme showed a temperature optimum of 50 °C with 63% of DMA and BDO when compared to 54% and 27%, at 70 °C and at 85 °C, respectively.

    Abstract This study analyses 371 yearly series of 1-day precipitation maxima extracted from a data set of high-quality homogenized rain gauge records covering an area of about 25,000 km2 in the western part of northern-central Italy that includes eastern Liguria and northern Tuscany and is very prone to severe floods and shallow landslides. Return levels of 1-day extreme precipitation (corresponding to 10-, 50-, and 100-year return periods as predicted by a generalized extreme value distribution (GEV)) are estimated both for station sites and for the nodes of a high-resolution grid (30 arcsec). GEV parameters are estimated by regional frequency analysis using the station-year approach. Grid-point index floods are estimated from yearly cumulative precipitation normals exploiting the very high correlation existing at local scale between station index flood and the corresponding yearly precipitation normals. Results clearly show the high vulnerability of this region to extreme precipitation events and highlight the different role played by total annual precipitation normals and by both scale and shape parameters in regulating the longer period return levels. The results presented in this paper allow better estimating the climatology of extreme events in the study area, contributing to a more effective hazard assessment at different spatial and temporal scales.

    Abstract Background: Multiple sclerosis (MS) is a chronic inflammatory and demyelinating disease of the CNS. An intrathecal IgM synthesis is associated with a more rapid progression of MS and the intrathecal immune response to measles -, rubella -and varicella zoster virus (MRZR) which, if present, increases the likelihood of a diagnosis of MS in adults.
    Objective: To evaluate the frequency of an intrathecal IgM synthesis and MRZR in children with MS. MethodsChildren with MS and a data set including clinical and treatment history, MRI at onset, in addition to a CSF analysis, and determination of antibody index (AI) of measles, rubella, and zoster antibodies, were eligible. The presence of an intrathecal IgM synthesis and/or a positive MRZ reaction were compared to biomarkers of a more progressive disease course.
    Results: In 75 children with MS, OCBs were present in 93.3 %). 49,2 % experienced their first relapse within 6 months. 50.7 % had a total lesion load of more than 10 lesions in the first brain MRI. Spinal lesions were identified in 64 %. 23.5 % had a positive MRZR and 40.3 % an intrathecal IgM synthesis. No significant associations were detected between the presence of an intrathecal IgM synthesis and MRZR and parameters including the relapse rate in the first two years.
    Conclusion: An intrathecal IgM synthesis and a positive MRZR are found in a subset of MS children but are not associated with markers associated with a poor prognosis.

    Abstract In the past 20 years, scientific research focused on the identification of valid alternatives to materials of fossil origin, in particular, related to biobased polymers. Recently, the efforts led to the synthesis of thiophene-based polymers (TBPs), a new class of polyesters based on 2,5-thiophenedicarboxylic acid (TPCA) that can be industrially produced using biomass-derived molecules. In this study, TBPs were synthesized using diols with different chain length (from C4 to C6) leading to poly(butylene 2,5-thiophenedicarboxylate) (PBTF), poly(pentamethylene 2,5-thiophenedicarboxylate) (PPeTF), and poly(hexamethylene 2,5-thiophenedicarboxylate) (PHTF), respectively, that were processed to thin films. To investigate enzymatic hydrolysis of these polymer films, cutinase 1 (Thc_cut1) and cutinase 2 (Thc_cut2) from Thermobifida cellulosilytica were recombinantly expressed in the host E. coli and purified. After 72 h of incubation at 65°C with 5 µM Thc_cut1, weight loss and HPLC analysis indicated 9, 100, and 80% degradation of PBTF, PPeTF, and PHTG with a concomitant release of 0.12, 2.70, and 0.67 mM of TPCA. The SEM analysis showed that tiny holes were formed on the surface of the films and after 72 h PPeTF was completely degraded. The LC-TOF/MS analysis indicated that Thc_cut2 in particular released various oligomers from the polymer during the reaction. In addition, the FTIR analysis showed the formation of novel acid and hydroxyl groups on the polymer surfaces. The results showed that the two used thermostable cutinases are promising biocatalysts for the environmentally friendly degradation of TPCA-based polyesters, in view of a possible sustainable recycling of plastic waste through resynthesis processes.

    Abstract Gradiometric gravimetry is a survey technique widely used in geological structure investigation. This work demonstrates the feasibility of a new class of low frequency accelerometers for geodynamics studies and space applications. We present the design features of a new low noise single-axis differential accelerometer; the sensor is suitable to be used in a Gravity Gradiometer (GG) system for land geophysical survey and gravity gradient measurements. A resolution of 1 Eötvös (1 Eö=10?9s?2) at one sample per second is achievable in a compact, lightweight (less than 2 kg) portable instrument, operating at room temperature. The basic components of the sensor are two identical rigidly connected accelerometers separated by a 15-cm baseline vector and the useful signal is extracted as the subtraction of the two outputs, by means of an interferometric microwave readout system. The structure will be engraved in a monocrystal of sapphire by means of Computer-Numerically-Controlled (CNC) ultrasonic machining: the material was chosen because of its unique mix of outstanding mechanical and dielectric properties.

    Abstract The prevalence of hepatitis B virus (HBV) in Brazil increases from South to North but moderate to elevated prevalence has been detected in the Southwest of Paraná State. The prevalence of serological markers of HBV was evaluated in 3188 pregnant women from different counties in Paraná State and relevant epidemiological features were described. The prevalence of HBV markers in pregnant women for the state as a whole was 18.5% (95% CI = 17.2-19.9), ranging from 7.2% in Curitiba to 38.5% in Francisco Beltrão. The endemicity of HBV marker prevalence in pregnant women was intermediate in Cascavel, Foz do Iguaçu, and Francisco Beltrão, and low in Curitiba, Londrina, Maringá, and Paranaguá. Multiple logistic regression showed that HBV marker prevalence increased with age, was higher among black women, among women of Italian and German descent, and among women who had family members in neighboring Rio Grande do Sul State. Univariate analysis showed that HBV marker prevalence was also higher among women with no education or only primary education, with a lower family income and whose families originated from the South Region of Brazil. Pregnant women not having positive HBV markers (anti-HBc, HBsAg or anti-HBs detected by ELISA) corresponded to 73.7% of the population studied, implying that HBV vaccination needs to be reinforced in Paraná State. The highest prevalence was found in three counties that received the largest number of families from Santa Catarina and Rio Grande do Sul, where most immigrants were of German or Italian ascendance. This finding probably indicates that immigrants that came to this area brought HBV infection to Southwestern Paraná State.

    Abstract No Estado do Paraná, a primeira notificação de leishmaniose tegumentar americana ocorreu em 1917 e a partir de 1980 observou-se um aumento do número de casos, mantendo-se endêmica e acometendo pessoas de todas as faixas etárias e em ambos os sexos. Este estudo teve como objetivo realizar um levantamento epidemiológico sobre a ocorrência de LTA em pacientes atendidos no Laboratório de Ensino e Pesquisa em Análises Clínicas da Universidade Estadual de Maringá (LEPAC/UEM). Foi realizado um estudo retrospectivo e descritivo em base de dados secundário de 1986 a 2005, com 1656 pacientes segundo as variáveis: sexo, idade, ocupação, procedência, local de moradia, forma clínica e diagnóstico. A maioria dos pacientes era do sexo masculino (72,6%) e adquiriu a infecção no Estado do Paraná (97,8%), residia em área urbana (64,3%) dos quais 51,3% adquiriu a infecção durante atividades de lazer. O diagnóstico da maioria dos pacientes foi estabelecido nos dois primeiros meses de evolução das lesões (54,0%) e apresentando a forma cutânea da doença (88,9%). Uma parcela (34,4%) significativa dos pacientes que residiam em área rural adquiriu a infecção no domicílio ou peridomicílio. O estudo mostra a predominância da forma cutânea da leishmaniose tegumentar americana e sugere a atividade de lazer e o ambiente do domicilio como fatores preditivos importantes para a infecção.     Abstract Realizou-se inquérito sorológico e epidemiológico para cisticercose em indivíduos de cinco municípios da região Norte do Estado do Paraná, Brasil. De 2.180 indivíduos investigados através da reação de imunofluorescência indireta, 69 (3,2%) apresentaram títulos significativos de anticorpos anti-Cysticercus cellulosae. Os percentuais de indivíduos com títulos significativos encontrados em Sarandi (6,6%) e Marialva (4,7%) não diferem estatisticamente (Z=1.319, P=0,0936), mas diferem dos percentuais encontrados em Mandaguaçu, Paiçandu e Maringá (P<0,01). Destes indivíduos, 47,9% estavam na faixa etária de 21 a 49 anos e 79,4% eram do sexo feminino. Foi comum o relato de queixas como "dores de cabeça" (70,6%), "tonturas" (57,4%) e "convulsões" (7,4%), além de história de teníase (22,1%) e hábitos de ingestão de carne crua bovina (41,2%) ou suína (27,9%) e carne com "canjiquinha" (25,0%). An epidemiological and serological study was carried out on a sample of 2,180 individuals, in five counties in the north of Paraná State - Brazil, using the indirect immunofluorescence test to detect anti-Cysticercus cellulosae antibodies. These individuals, 69 (3.2%) showed significant titers of antibodies. No single significant difference between the proportion of reactivity in Sarandi (6.6%) and in Marialva (4.7%) was observed (Z=1,319, P=0,0936), but it was significantly higher than that observed in Mandaguaçu, Paiçandu and Maringá (P<0.01). Of these individuals, 47.9% were within 21-49 years old and 79.4% were of female sex. "Headache" (70.6%), "faintness" (57.4%), and "convulsions" (7.4%) were among the most frequent by reported, moreover, cases of Taenia infections (22.1%) and the custom of eating uncooked beef (41.2%) or pork (27.9%) and meat containing cysticerci (25.0%) were also related.

    Abstract We search for gravitational-wave transients associated with gamma-ray bursts detected by the Fermi and Swift satellites during the first part of the third observing run of Advanced LIGO and Advanced Virgo (1 April 2019 15:00 UTC - 1 October 2019 15:00 UTC). 105 gamma-ray bursts were analyzed using a search for generic gravitational-wave transients; 32 gamma-ray bursts were analyzed with a search that specifically targets neutron star binary mergers as short gamma-ray burst progenitors. We describe a method to calculate the probability that triggers from the binary merger targeted search are astrophysical and apply that method to the most significant gamma-ray bursts in that search. We find no significant evidence for gravitational-wave signals associated with the gamma-ray bursts that we followed up, nor for a population of unidentified subthreshold signals. We consider several source types and signal morphologies, and report for these lower bounds on the distance to each gamma-ray burst.

    Abstract Realizou-se inquérito sorológico e epidemiológico para cisticercose em indivíduos de cinco municípios da região Norte do Estado do Paraná, Brasil. De 2.180 indivíduos investigados através da reação de imunofluorescência indireta, 69 (3,2%) apresentaram títulos significativos de anticorpos anti-Cysticercus cellulosae. Os percentuais de indivíduos com títulos significativos encontrados em Sarandi (6,6%) e Marialva (4,7%) não diferem estatisticamente (Z=1.319, P=0,0936), mas diferem dos percentuais encontrados em Mandaguaçu, Paiçandu e Maringá (P<0,01). Destes indivíduos, 47,9% estavam na faixa etária de 21 a 49 anos e 79,4% eram do sexo feminino. Foi comum o relato de queixas como "dores de cabeça" (70,6%), "tonturas" (57,4%) e "convulsões" (7,4%), além de história de teníase (22,1%) e hábitos de ingestão de carne crua bovina (41,2%) ou suína (27,9%) e carne com "canjiquinha" (25,0%).
