    Abstract A visual measurement method based on a key point detection network is proposed for the difficulty of fitting the cross-sectional profile of ultra-narrow gap welds and the low efficiency and accuracy of manual measurement of geometric parameters. First, the HRnet (High-Resolution Net) key point detection algorithm was used to train the feature point detection model, and 18 profile feature points in a “measurement unit” were extracted. Secondly, the feature point coordinates are transformed from the image coordinate system to the weld coordinate system, and the weld profiles are fitted by the least squares method. Finally, the measurement system is calibrated with a coplanar linear calibration algorithm to perform pixel distance to actual distance conversion for quantitative detection of geometric dimensions. The experimental results show that the accuracy of the proposed method for key point localization is 95.6%, the mean value of the coefficient of determination R-square for curve fitting is greater than 94%, the absolute error of measurement is between 0.06 and 0.15 mm, and the relative error is between 1.27% and 3.12%. The measurement results are more reliable, and the efficiency is significantly improved compared to manual measurement.

    Abstract At the field of nanometer positioning and machining, high-precision tracking is a key technology of the micro-positioning platform which is driven by a voice coil motor. To improve the tracking accuracy and response speed, the sliding-mode active disturbance rejection control is proposed. The mathematical model of the micro-positioning platform control system is established, in which the perturbation and spring-damping force are set as the unknown terms, and an extended state observer is used to estimate and compensate for the unknown terms. To improve the robustness of the system, the equivalent sliding-mode term is constructed to replace the PD control term in the conventional active disturbance rejection. Further, the stability of the system is proved by the Lyapunov stability theory, and compared with the conventional sliding-mode controller, the effectiveness of the proposed control strategy is verified by simulation.

    Abstract Background:Podocytes are essential components of the glomerular filtration barrier and essential for the proper filtration function of the glomerulus. Podocyte injury under various stress conditions is the primary pathogenesis and key determinant of focal segmental glomerulosclerosis (FSGS) with prominent clinical manifestations of proteinuria or nephrotic syndrome. Summary:Under physiological conditions, a highly coordinated mitochondrial quality control system, including antioxidant defenses, mitochondrial dynamics (fusion, fission and mitophagy), and mitochondrial biogenesis, guarantees the sophisticated structure and various functions of podocytes. However, under FSGS pathological conditions, mitochondria encounter oxidative stress, dynamics disturbances, and defective mitochondrial biogenesis. Moreover, mutations in mtDNA and mitochondria-related genes are also strongly associated with FSGS. Based on these evidences, bioactive agents that function to relieve mitochondrial oxidative stress and promote mitochondrial biogenesis have been proven effective in pre-clinical FSGS models. Targeting the mitochondrial network is expected to provide new therapeutic strategies for the treatment of FSGS and delay its progression to end-stage renal disease (ESRD). Key messages:Mitochondrial dysfunction plays a key role in podocyte injury and FSGS progression. This review summarized recent advances in the study of mitochondrial homeostatic imbalance and dysfunction in FSGS and discussed the potential of mitochondria-targeted therapeutics in improving FSGS and retarding its progression to ESRD.

    Abstract Yunnan Baiyao is a famous Chinese patent medicine in Yunnan Province. However, its mechanism for promoting blood circulation and removing blood stasis is not fully explained. Our study used metabonomics technology to reveal the regulatory effect of Yunnan Baiyao on small molecular metabolites in promoting blood circulation and removing blood stasis, and exploring the related urine biomarkers. The coagulation function, blood rheology, and pathological results demonstrated that after Yunnan Baiyao treatment, the pathological indexes in rats with epinephrine hydrochloride-induced blood stasis syndrome improved and returned to normal levels. This is the basis for the effectiveness of Yunnan Baiyao. UPLC-G2Si-HDMS was used in combination with multivariate statistical analysis to conduct metabonomic analysis of urine samples. Finally, using mass spectrometry technology, 28 urine biomarkers were identified, clarifying the relevant metabolic pathways that play a vital role in the Yunnan Baiyao treatment. These were used as the target for Yunnan Baiyao to promote blood circulation and remove blood stasis. This study showed that metabolomics strategies provide opportunities and conditions for a deep and systematic understanding of the mechanism of action of prescriptions.

    Abstract BackgroundIn addition to the typical signs of skin damage, long-term arsenic exposure is often accompanied by signs and symptoms of neurobehavioral abnormalities.ObjectiveTo investigate potential intervention effect of sciadopitysin on senescence of neurons induced by sodium arsenite in rats and possible underlying mediating effect of cell cycle-related transcription factor E2F1.MethodsSH-SY5Y cells were treated with 4 μmol·L−1 sodium arsenite for 24 h and intervened with 50 μg·mL−1 Ginkgo biloba extract (EGb761) or four major biflavonoids in Ginkgo biloba leaves (isoginkgetin, bilobetin, sciadopitysin, and ginkgetin) for 24 h respectively. Then, cell viability was measured by CCK-8 assay. Thirty-two 180-200 g SPF rats were randomly divided into a control group, an arsenic treatment group (10 mg·L−1), a Ginkgo biloba extract intervention group (10 mg·kg−1), and a sciadopitysin intervention group (10 mg·kg−1), 8 rats in each group, half male and half female. The rats were treated with sodium arsenite by free drinking water for 3 consecutive months, and the intervention treatment was conducted after 2 months of poisoning with drug intake by gavage for 1 month. HE staining was used to detect structural changes in the hippocampus, while Nissl's staining was used to detect changes in hippocampal morphology and neuron numbers. Moreover, senescence-associated β galactosidase (SA-β-gal) staining and Western blotting were used to detect senescence of hippocampal neurons and the expression level of E2F1, respectively.ResultsCompared to the arsenic treatment group, EGb761 and the four biflavonoids in Ginkgo biloba leaves effectively antagonized the inhibitory effect of sodium arsenite on cell viability (all Ps<0.05), and sciadopitysin showed better restoration of cellular viability than Ginkgo biloba extract (P<0.05).     Abstract In fault-diagnosis classification, a pressing issue is the lack of target-fault samples. Obtaining fault data requires a great amount of time, energy and financial resources. These factors affect the accuracy of diagnosis. To address this problem, a novel fault-diagnosis-classification optimization method, namely TLSCA-SVM, which combines the sine cosine algorithm and support vector machine (SCA-SVM) with transfer learning, is proposed here. Considering the availability of fault data, this thesis uses the data generated by analog circuits from different faults for analysis. Firstly, the data signal is collected from different faults of the analog circuit, and then the characteristic data are extracted from the data signals by the wavelet packets. Secondly, to employ the principal component analysis (PCA) reduces the feature-value dimension. Lastly, as an auxiliary condition, the error-penalty item is added to the objective function of the SCA-SVM classifier to construct an innovative fault-diagnosis model namely TLSCA-SVM. Among them, the Sallen–Key bandpass filter circuit and the CSTV filter circuit are used to provide the data for horizontal- and vertical-contrast classification results. Comparing the SCA with the five optimization algorithms, it is concluded that the performance of SCA optimization parameters has certain advantages in the classification accuracy and speed. Additionally, to prove the superiority of the SCA-SVM classification algorithm, the five classification algorithms are compared with the SCA-SVM algorithm. Simulation results showed that the SCA-SVM classification has higher precision and a faster response time compared to the others. After adding the error penalty term to SCA-SVM, TLSCA-SVM requires fewer fault samples to process fault diagnosis. Ultimately, the method which is proposed could not only perform fault diagnosis effectively and quickly, but also could run effectively to achieve the effect of transfer learning in the case of less failure data.

    Abstract HSP70s play crucial roles in plant growth and development, as well as in stress response. Knowledge of the distribution and heat response of HSP70s is important to understand heat adaptation and facilitate thermotolerance improvement in wheat. In this study, we comprehensively analyzed the distribution of HSP70s in hexaploid wheat ( TaHSP70s ) and its relatives, and we found an obvious expansion of TaHSP70s in the D genome of hexaploid wheat. Meanwhile, a large portion of tandem duplication events occurred in hexaploid wheat. Among the 84 identified TaHSP70s , more than 64% were present as homeologs. The expression profiles of TaHSP70s in triads tended to be expressed more in non-stressful and heat stress conditions. Intriguingly, many TaHSP70s were especially heat responsive. Tandem duplicated TaHSP70s also participated in heat response and growth development. Further HSE analysis revealed divergent distribution of HSEs in the promoter regions of TaHSP70 homeologs, which suggested a distinct heat regulatory mechanism. Our results indicated that the heat response of TaHSP70s may experience a different regulation, and this regulation, together with the expression of tandem duplicated TaHSP70s , may help hexaploid wheat to adapt to heat conditions.

    Abstract Long-term exposure to high levels of arsenic has been documented to induce skin and liver damage, affecting hundreds of millions of people. While arsenic-induced skin and liver damage and trace element alterations have been studied, their correlations and risks have not been explained. Based on the above premise, this study included a total of 172 subjects from a coal-burning arsenic poisoning area. The levels of 18 trace elements in hair and six liver function indices in serum were detected, and the associations between and risks of trace elements related to skin and liver damage were analyzed. Finally, the receiver operating characteristic (ROC) curve and areas under the curve (AUC) were used to analyze the diagnostic values of certain trace elements for arsenic-induced skin and liver damage. The results found that a decrease in Se was a risk factor for arsenic-induced skin and liver damage (OR = 8.33 and 1.92, respectively). Furthermore, increases in Al and V were risk factors for arsenic-induced skin damage (OR = 1.05) and liver damage (OR = 13.16), respectively. In addition, the results found that Se and Al possessed certain diagnostic values for arsenic-induced skin damage (AUC = 0.93, 0.80), that Se possessed a diagnostic value for liver damage (AUC = 0.93), and that the combination of Se and Al increased the diagnostic value for skin damage (AUC = 0.96). This study provides an important research basis for further understanding the reasons for arsenic-induced skin and liver damage, for screening and identifying candidate diagnostic biomarkers, and for improving prevention and control strategies for arsenism.
