    Abstract Various studies have shown the anti-inflammatory and anti-oxidant effects of Descorainia sophia. The purpose of this study was to evaluate the hepatoprotective ability of Descorainia sophia ethanolic extract in liver damage induced by carbon tetrachloride in male Wistar rats. In this experimental study, 60 male Wistar rats were randomly divided into 10 groups of 6 consisting of normal control, intoxified control (intraperitoneal injection of 0.5 ml/kg of carbon tetrachloride), normal experimental (Descorainia sophia ethanolic extract at doses of 10, 50, 100 and 200 mg/kg via intragastric gavage) and intoxified experimental (intraperitoneal injection of 0.5 ml/kg of carbon tetrachloride and Descorainia sophia ethanolic extract at doses of 10, 50, 100 and 200 mg/kg via intragastric gavage). After 28 days, the levels of antioxidant enzymes viz. superoxide dismutase, catalase, glutathione peroxidase and level of malondialdehyde in liver homogenate were evaluated. Data were analyzed using one-way ANOVA and the Tukey post hoc test with statistical significance defined as p<0.05. Our results showed that administration of carbon tetrachloride significantly decreased the levels of antioxidant enzymes and increased the level of malondialdehyde in the intoxified control group in comparison to normal control group (p<0.001). Also, the administration of Descorainia sophia ethanolic extract significantly increased the levels of antioxidant enzymes and decreased the level of malondialdehyde in the liver of intoxified experimental groups in comparison with the intoxified control group (p<0.05) in a dose dependent manner. The results of the study indicated that Descorainia sophia removes free radicals and reduces oxidative stress caused by carbon tetrachloride in hepatic tissue of rats probably due to its flavonoid compounds.

    Abstract A biosensor based on glutamate oxidase (GluOx) was developed to measure glutamate concentration. The main function of this type of biosensor is related to the structure and catalytic activity of GluOx. Since radiofrequency, as the widest spectrum of electromagnetic fields, can affect the catalytic activity and structure of GluOx, in this study, the effect of these fields on the analytical parameters of the fabricated biosensor was investigated. To build the biosensor a sol-gel solution of chitosan and native GluOx were prepared and then immobilized on the surface of the platinum electrode. Similarly, to investigate the effect of radiofrequency fields on the analytical parameters of the biosensor, instead of the native GluOx, irradiated GluOx was used to build the biosensor. To evaluate the biosensor responses, cyclic voltammetry experiments were performed and voltammograms were considered as biosensor responses. To determine the analytical parameters including detection limit, linear range, and saturation region of the responses, calibration curves were drawn for each of the biosensors. Also the long-term stability and selectivity of the fabricated biosensor were evaluated. Thereafter, the optimum pH and temperature for each of these two biosensors were examined. The results showed that radiofrequency waves harmed the detection and response of biosensors in the saturation region, while they had little effect on the linear region. Such results could be due to the effect of radiofrequency waves on the structure and function of glutamate oxidase. In general, the results indicate that when a glutamate oxidase-based biosensor is used to measure glutamate in radiofrequency fields, corrective coefficients for this type of biosensor should be considered to accurately measure glutamate concentration.

    Abstract Varicocele is a pathological dilation of the venous network of the spermatic cord and considering that magnesium oxide nanoparticles play a key role in various physiological functions, the aim of this study was to evaluate the performance of nanoparticles on sperm characteristics affected by experimental varicocele. A total of 54 male Wistar rats were randomly divided into 9 equal groups including healthy control group (untouched animals), sham-operated group (underwent sham surgery), three healthy experimental groups (animals in these groups received magnesium oxide nanoparticles at doses of 1.25, 2.5 and 5 mg/kg respectively by gavage for 6 weeks), varicocele control group (varicocele was induced by renal vein ligation) and three experimental varicocele groups (in addition to varicocele induction, magnesium oxide nanoparticles were given at doses of 1.25, 2.5 and 5 mg/kg respectively by gavage for 6 weeks). At the end of the 6th week, the abdomen was opened and semen samples were collected from the tail of the epididymis to determine the indices of concentration, survival and motility of sperm and the data were statistically analyzed (p<0.05). The results showed that magnesium oxide nanoparticles were able to significantly increase the concentration, viability, progressive, moderate and non-progressive movements of sperm compared to the varicocele group (p<0.001) and also caused a significant decrease (p<0.001) in the number of non-moving sperms in the varicocele experimental groups. Therefore, magnesium oxide nanoparticles may possibly reduce the destructive effects of varicocele due to their antioxidant activity and be effective in its treatment by improving sperm properties during varicocele.

    Abstract Introduction: Many factors affect memory and learning. One of the most important ones is magnesium, which is essential for the proper functioning of our memory. Magnesium is the fourth most important cation and the second most important intracellular cation after potassium in the body. Magnesium plays an important role in neurotransmission. This study aimed to examine magnesium oxide nanoparticles' effect on the parameters of oxidative stress to treat memory deficit in the rat model of Alzheimer's disease with the help of Amyloid-β. Material & Methods: In this experimental study, 54 adult male rats were divided randomly into nine groups, such as 1. Healthy control group, 2. Alzheimer's control group (rats that underwent stereotactic surgery and received 2nmol/µl of Amyloid-β by intracerebroventricular injection [ICVI]), 3. Sham group (rats that underwent stereotactic surgery and received saline as an Amyloid-β’s solvent), 4, 5, and 6. The healthy experimental groups (healthy rats receiving 2.5, 5, and 10 mg/kg body-weight doses of magnesium oxide nanoparticles by intraperitoneal injection), 7, 8, and 9. Alzheimer's experimental group (Alzheimer's rats receiving 2.5, 5, and 10 mg/kg body-weight doses of magnesium oxide nanoparticles by intraperitoneal injection). The duration of oral treatment of nanoparticles was 30 days. At the end of the treatment period (30 days), oxidative stress parameters, including superoxide dismutase (SOD), glutathione peroxidase (GPX), catalase (CAT), and malondialdehyde (MDA) were measured in neural (brain tissue), and all data were analyzed by one-Factor ANOVA and Tukey post hoc test in SPSS software (version 21) considering a significance level of P<0.05.     Abstract Background and Objectives: Many herbal medicines have been recommended for the treatment of diabetes. The genus Artemisia has always been of great botanical and pharmaceutical interest, and is used in traditional medicine for the treatment of various diseases. The present study was performed aiming at investigating the effect of alcoholic extract of the aerial parts of Artemisia deserti in normal and streptozotocin-induced diabetic rats. Methods: To induce diabetes, the rats received a single intraperitoneal injection of streptozotocin (70mg/kg). Ethanolic extract of Artemisia deserti in amounts of (50, 100, 200, and 400mg/kg body weight) were administrated orally for 28 days in normal and diabetic rats. At the end of treatment, the animals were anesthetized and blood samples were collected from their heart. Then, serum levels of glucose, insulin, total cholesterol, triglyceride, urea, uric acid, and creatinine were measured. Data were analyzed by one-way ANOVA and Tukey’s test. The significance level was defined as p<0.05. Results: Oral administration of ethanolic extract of aerial parts of Artemisia deserti for 28 days significantly decreased serum levels of glucose, triglyceride, total cholesterol, urea, uric acid, and creatinine in the diabetic rats. Also, oral administration of the plant extract significantly increased serum insulin level in diabetic rats. Oral administration of the extract caused no significant change in the levels of the above mentioned parameters in normal rats. Conclusion: The results of the present study showed that Artemisia deserti has antidiabetic effect in rats and can be considered for therapeutic purposes.

    Abstract Background and purpose: Oxidative stress and neuroinflammation are the main causes of cerebral ischemia. Saponin with steroid or triterpenoid compounds with a sugar part has strong antioxidant and anti-inflammatory properties. The aim of this study was to evaluate the effect of saponin on cerebral edema, Bax gene expression level, and ischemic neurological defects. Materials and methods: In this experimental study, 24 male Wistar rats were divided into three groups: sham, stroke, and stroke+saponin. Rats in the stroke+saponin group received intravenous injection of saponin (3mg/kg) 30 minutes before cerebral ischemia. Each group was divided into two subgroups: brain edema (n=5) and gene expression level investigation (n=3). Cerebral ischemia was induced by middle cerebral artery occlusion model. After 24 hrs, we investigated neurological deficits resulting from ischemia (Kruskal-Wallis test), the degree of brain edema, and Bax gene expression levels in the cortex, piriform cortex-amygdala, striatum, and hippocampus (one-way ANOVA). Results: Findings indicated a significant decrease in the score of neurological defects resulting from ischemia, the amount of cerebral edema, and the level of Bax gene expression in stroke+saponin group (P<0.05). Conclusion: This study showed that saponin pretreatment in rats with ischemia-reperfusion injury can be effective in reducing the damage caused by cerebral ischemia.

    Abstract Zinc transporters (ZnT) and ZIP proteins maintain Zinc homeostasis in the live organisms. On the other hand, the impacts of zinc oxide nanoparticles (ZnO NPs) on the expression of the Znt genes in biological systems were not clear yet. So that in this experimental study we have tried to find the effects of ZnO NPs on Znt1-4 genes expression in the hippocampus of male rats under acute stress. Adult male rats were divided into groups of control and treated with 5 or 10 mg/kg of ZnO NPs alone and under acute restraint stress for 90 min. The changes in the expression of the selected genes were monitored using real-time qRT-PCR. The ZnT4 protein expression also was measured by Western blotting. Real-time qRT-PCR expression analysis revealed that the Znt1 gene expression was up-regulated in the stress group, while the expression of the Znt1 and Znt4 genes was significantly up-regulated in the group receiving 10 mg/kg of ZnO NPs. Furthermore, in the ZnO NPs 10 mg/kg group under stress, the Znt2 gene expression was down-regulated, while the Znt4 gene expression was up-regulated. Moreover, the levels of ZnT4 protein were significantly increased after 10 mg/kg of ZnO NPs injection in the stress and normal groups. According to these results ZnO NPs administration can cause changes in the expression of a number of zinc transporter genes under stress conditions and increases the ZnT4 protein level. Therefore, this is a valuable approach for forecast investigation in biomedicine and pharmacogenetics studies.

    Abstract Background: Multiple sclerosis (MS) is one of the most common diseases of the nervous system, characterized by inflammation of the central nervous system and oxidative stress. Carvacrol is a monoterpenoid phenol with antioxidant effects against free radical. The aim of this study was to evaluate the effect of carvacrol on the expression of Hmox-1, iNOS, Nrf2 and NF-ҚB genes in the spinal cord of Experimental Autoimmune Encephalomyelitis (EAE) rats as an animal model of MS. Materials and Methods: EAE was induced in female Lewis rats and they were then divided into three groups: control, EAE model, and EAE treated with carvacrol. Carvacrol was daily injected intraperitoneally for 17 days after immunization. RNA was extracted from rat spinal cord tissue and changes in the expression of the Hmox-1, iNOS, Nrf2 and NF-ҚB genes were examined by Real-Time PCR. Results: The results showed that carvacrol treatment stopped severe weight loss in the EAE model group and their mean weight increased. Furthermore, carvacrol was found to reduce the expression of iNOS and NF-ҚB genes and increase the levels of Hmox-1 mRNA and Nrf2 mRNA. Conclusion: The effect of carvacrol on weight changes in treated rats, along with the increased HO-1 gene expression in their spinal cord, associated with increased Nrf2 mRNA and decreased expression of iNOS and NF-ҚB, led to a reduction in inflammation and oxidative stress, and an increase in the neuroprotection. Furthermore, the invasive environment created for some cells, such as oligodendrocytes, was modulated.

    Abstract Introduction: Angiography is a safe cardiovascular technique for the diagnosis and treatment of cardiovascular disorders. The potential effects of angiography on the levels of cytokines are yet to be clarified completely. Interleukin-8 (IL-8) and tumor necrosis factor-alpha (TNF-α) are important pro-inflammatory cytokines that participate in the pathogenesis of artery stenosis. The aim of this study was to investigate the effects of angiography on the serum levels of IL-8 and TNF-α.Methods: Sixty participants in three groups, without, with one, and with more than one artery stenosis, were explored in this project. Serum levels of IL-8 and TNF-α were measured in the participants before and after angiography using the enzyme linked immunosorbent assay (ELISA) technique.Results: Serum levels of IL-8, but not TNF-α, significantly decreased following angiography from 61.45 (9.80-266.72) to 7.00 (3.82-17.85) pg/mL (P = 0.008) in participants without artery stenosis, from 79.40 (7.20-197.40) to 6.80 (3.85-29.50) pg/mL (P = 0.002) in participants with one artery stenosis, and from 91.25 (12.80-145.97) to 10.75 (6.97-49.82) pg/mL (P = 0.004) in participants with more than one artery stenosis. The X-ray dose had a moderate positive correlation with serum levels of TNF-α in patients with more than one artery stenosis (r = 0.584, P = 0.009). Serum levels of IL-8 and TNF-α were not different between male and female participants in none of the groups. Conclusion: Angiography may be a protective factor against inflammation by suppressing IL-8.

    Abstract Introduction: Zinc plays an important role in the function of vital organs, especially the central nervous system. Zinc homeostasis disorder causes and progresses nervous system diseases such as Alzheimer, depression, learning disabilities and stress. Zinc homeostasis in the body is mediated by ZnT and ZIP proteins. The aim of this study was to investigate the effect of zinc oxide nanoparticles on the expression of Znt1, Znt2, Znt3, and Znt4 genes in hippocampus cells as one of the tissues with high zinc density. Material and methods: First, the cell passage of the hippocampus cell line was performed, then the MTT assay test was performed for zinc oxide nanoparticles. In the next step, RNA extraction and CDNA synthesis were performed, and nanodrop spectrophotometer was used to ensure the purity of the RNA samples. Specific and appropriate primers of the desired genes were designed and synthesized. Then, changes in the expression of Znt1, Znt2, Znt3, and Znt4 genes were investigated using Real-Tim e RT-PCR. Results: Concentrations of 10 and 20 μg/mL of zinc oxide nanoparticles, significantly increased the expression of Znt1, Znt2, Znt3, and Znt4 genes in the hippocampus cell line of rat, while creating the lowest cytotoxicity. Conclusion: Zinc oxide nanoparticles can be investigated pharmacologically by increasing the expression of Znt1, Znt2, Znt3, and Znt4 genes in the treatment of zinc homeostasis disorders such as Alzheimer, depression, learning disabilities and stress.
