    Abstract The Trichocentrum cepula complex comprises three species, T. caatingaense, T. cepula and T. sprucei, endemic to tropical forests east of the Andes in South America. The delimitation of these species has been diversely interpreted due to the extensive morphological variation in the complex. We applied an integrative approach to achieve a better understanding of these biological units, using geometric morphometrics, cytogenetic analysis (chromosome counts and CMA/DAPI banding) and molecular phylogenetics (ITS and rpl32-trnL). An initial morphometric analysis using the pre-identified specimens into three taxa suggested that T. sprucei is distinct from the other two, which show some overlap. A subsequent analysis of the labellum, including only T. caatingaense and T. cepula organized in six pseudo-populations, suggested the existence of four morphological groups. All analysed specimens presented 2n = 36 chromosomes, CMA⁺/DAPI⁻ terminal bands and CMA⁻/DAPI⁺ pericentromeric bands, which varied in number across species, localities or even individuals from the same locality. The notable variation in DAPI⁺ pericentromeric bands may be related to transposable elements that could also be a factor influencing the wide morphological variation in the flowers. In the phylogenetic analysis, the specimens belonging to T. caatingaense formed a strongly supported clade sister to the rest, whereas the specimens belonging to T. cepula and T. sprucei emerged together, with their relationships tending to be determined by geographic proximity. The evidence we generated suggests that treating the Brazilian populations of this species complex under a single name, T. cepula, provides more taxonomic stability and utility, thus the necessary taxonomic changes are implemented.

    Abstract The cashew, Anacardium occidentale, is a globally important tropical fruit tree, but little is known about its natural infraspecific systematics. Wild Brazilian populations occur in the cerrado biome and coastal restinga vegetation. We investigated whether wild coastal and domesticated populations could be distinguished genetically using inter-simple repeat molecular markers (ISSRs). In total, 94 polymorphic loci from five primers were used to characterise genetic diversity, structure and differentiation in four wild restinga populations and four domesticated ones from eight localities in Piauí state (30 individuals per population). Genetic diversity was greater overall in wild (%P: 57.2%, I: 0.24, Hₑ : 0.15) than domesticated populations (%P: 49.5%, I: 0.19, Hₑ : 0.12). Significant structure was observed among the eight populations (between-population variance 22%, ΦPT = 0.217, P ≥ 0.001), but only weak distinctions between wild and domesticated groups. Cluster and principal coordinate analyses showed marked genetic disparity in populations. No correlation of genetic and geographical inter-population distance was found (Mantel test, r = 0.02032, P = 0.4436). Bayesian analysis found an eight-group optimal model (ΔK = 50.2, K = 8), which mostly corresponded to sampled populations. Wild populations show strong genetic heterogeneity within a small geographical area despite probable gene flow between them. Within-population genetic diversity of wild plants varied considerably and was lower where extractive activities by local people are most intense (Labino population). The study underlines the importance of wild populations as in situ genetic reserves and the urgent need for further studies to support their conservation.

    Abstract Pilocarpus microphyllus Stapf ex Wardlew (Rutaceae), popularly known as jaborandi, is a plant native to the northern and northeastern macroregions of Brazil. Several alkaloids from this species have been isolated. There are few reports of antibacterial and anthelmintic activities for these compounds. In this work, we report the antibacterial and anthelmintic activity of five alkaloids found in P. microphyllus leaves, namely, pilosine, epiisopilosine, isopilosine, epiisopiloturine and macaubine. Of these, only anthelmintic activity of one of the compounds has been previously reported. Nuclear magnetic resonance, HPLC and mass spectrometry were combined and used to identify and confirm the structure of the five compounds. As regards the anthelmintic activity, the alkaloids were studied using in vitro assays to evaluate survival time and damaged teguments for Schistosoma mansoni adult worms.     Abstract Genetic variation was investigated using AFLP markers in 12 populations of Anthurium sinuatum and A. pentaphyllum var. pentaphyllum (Araceae) in north-east Brazil, Amazonia and the Brazilian Atlantic forest. Two unique genetic patterns characterized the populations of A. sinuatum as a group, but no correlation between genetic and geographical interpopulation distance was found; the Amazonian population was not separated from that in Ceará. The isolated Ceará brejo populations of A. sinuatum were genetically distinct, but genetic diversity levels were similar to populations elsewhere, with no evidence of genetic erosion. Anthurium pentaphyllum populations were significantly different from each other; Bayesian genetic structural analysis found no common genetic pattern, but revealed genetic clusters unique to subgroups and individual populations in the Atlantic forest and French Guiana. Anthurium pentaphyllum and A. sinuatum can be distinguished genetically, but individuals of both species formed intermediate genetic clusters that blurred their distinction. We suggest that genetic mixing of A. sinuatum and A. pentaphyllum has occurred in north-east Brazil, possibly connected with cycles of humid forest expansion. The weak genetic structure in A. sinuatum is consistent with the natural fragmentation of continuous forest areas, possibly during the Holocene. This study highlights the scientific importance of the highly threatened brejo forests for tropical American biogeography.

    Abstract The shortage of reliable primary taxonomic data limits the description of biological taxa and the understanding of biodiversity patterns and processes, complicating biogeographical, ecological, and evolutionary studies. This deficit creates a significant taxonomic impediment to biodiversity research and conservation planning. The taxonomic impediment and the biodiversity crisis are widely recognized, highlighting the urgent need for reliable taxonomic data. Over the past decade, numerous countries worldwide have devoted considerable effort to Target 1 of the Global Strategy for Plant Conservation (GSPC), which called for the preparation of a working list of all known plant species by 2010 and an online world Flora by 2020. Brazil is a megadiverse country, home to more of the world's known plant species than any other country. Despite that, Flora Brasiliensis, concluded in 1906, was the last comprehensive treatment of the Brazilian flora. The lack of accurate estimates of the number of species of algae, fungi, and plants occurring in Brazil contributes to the prevailing taxonomic impediment and delays progress towards the GSPC targets. Over the past 12 years, a legion of taxonomists motivated to meet Target 1 of the GSPC, worked together to gather and integrate knowledge on the algal, plant, and fungal diversity of Brazil. Overall, a team of about 980 taxonomists joined efforts in a highly collaborative project that used cybertaxonomy to prepare an updated Flora of Brazil, showing the power of scientific collaboration to reach ambitious goals. This paper presents an overview of the Brazilian Flora 2020 and provides taxonomic and spatial updates on the algae, fungi, and plants found in one of the world's most biodiverse countries. We further identify collection gaps and summarize future goals that extend beyond 2020. Our results show that Brazil is home to 46,975 native species of algae, fungi, and plants, of which 19,669 are endemic to the country. The data compiled to date suggests that the Atlantic Rainforest might be the most diverse Brazilian domain for all plant groups except gymnosperms, which are most diverse in the Amazon. However, scientific knowledge of Brazilian diversity is still unequally distributed, with the Atlantic Rainforest and the Cerrado being the most intensively sampled and studied biomes in the country. In times of “scientific reductionism”, with botanical and mycological sciences suffering pervasive depreciation in recent decades, the first online Flora of Brazil 2020 significantly enhanced the quality and quantity of taxonomic data available for algae, fungi, and plants from Brazil. This project also made all the information freely available online, providing a firm foundation for future research and for the management, conservation, and sustainable use of the Brazilian funga and flora.
